Insulin improves cardiac myocytes contractile function recovery in simulated ischemia-reperfusion:Key role of Akt

The present study examined cardiac myocytecontractile and Ca2+ transient responses to insulin duringsimulated ischemia/reperfusion (I/R) and furtherinvestigated the role of protein kinase B (Akt) in the insulin-induced inotropic effect. Ventricular myocytes wereenzymatically isolated from adult Sprague-Dawley rats and perfused with Tyrode solution while electrically field-stimulated. Simulated I/R was induced by perfusing the cells with chemical anoxic solution including sodiumcyanide-sodium lactate for 15 min followed by reperfusionwith normal oxygenated Tyrode solution with or withoutinsulin. It is found that insulin only at concentration as high as 10 IU/L could increase cell shortening (16±5%, P < 0.05) in normal myocytes, whereas it concentration-dependently (0.01—10 IU/L) increased the contraction,the velocity ofshortening/releng- thening and Ca2+ transient in I/Rmyocytes. In addition, insulin treatment (1 IU/L) increasedAkt phosphorylation of I/R cardiomyocytes by 2.4-foldcompared with that of the control (P < 0.01). Mostimportantly, pretreatment with LY 294002, a specificinhibitor of phosphatidylinositol 3′-kinase (PI3-kinase), significantly inhibited both Akt phosphorylation and thepositive inotropic response to insulin in the I/Rcardiomyocytes. These results suggest that insulin exertsdirect positive inotropic effect by increasing Ca2+ transient of cardiomyocytes, which is enhanced in the pathologicalcondition of I/R. Akt activation plays an important role inthe insulin-induced improvement of myocyte contractile function following I/R.

To study the relationship between OLS and adrenergic neurotransmis-sion is the purpose of this *** inhibitory effect of phentolamine(5×10-8M)on OLS was *** results showed that on the basiccontraction of myenteric plexus longitudinal muscle（MPLM）,phentolaminehad no direct effects;phentolamine could not antagonize the light dose(2×10-7M)of NE-inhibitory on MPLM basic contraction;light dose phento-1amine could antagonize the inhibitory effect of OLS induced by high fre-quence（10Hz）stimulation to the MPLM basic contraction;phentolaminecould potentiate the effect of naloxone in antagonizing the OLS. For re-searching the interaction of adrenergic and enkephalinergic neurotransmis-sion,the present results may be helpful,and also provide a new idea aboutresearching the reeeptokinetics of two types of receptor.

＜正＞P2X3 and P2X2/3 receptors are expressed in peripheral tissues and dorsal root ganglia（DRG） and participate in peripheral ***,the mechanisms underlying P2X receptor-mediated nociception at different ovarial hormone levels has not been *** this study,twenty-four female rats were randomly divided into sham-operated（sham）, ovariectomized（OVX）,estrogen-treated,and estrogen-progesterone-treated groups with *** each group,the visceromotor reflex（VMR） to colorectal distention was tested and the DRG were harvested for a real-time PCR analysis of P2X3 and P2X2 receptor *** OVX rats with colitis,we found that the VMR to colorectal distention and P2X3 receptor mRNA in DRG were both significantly *** replacement reversed the ***,neither the VMR nor the P2X3 mRNA level in DRG from OVX colitis rats was reversed by the complex of estrogen and progesterone. Patch-clamp recording showed that in colitis rats,estradiol rapidly potentiated the sustained and transient currents evoked by ATP to（336±49）%and（122±12）%of controls,respectively,in a subpopulation of DRG neurons,which were blocked by ICI 182,780,an antagonist of the estrogen *** progesterone rapidly inhibited the transient currents induced by ATP to（67±10）%of control and had no effect on the sustained currents evoked by the same agonist. These results indicate that P2X3 receptors are likely to be an important contributor to the altered colonic functions in colitis rats,where the underlying mechanisms are closely related to endogenous estrogen modulation.

In a tail suspension rat model, we investigated changes in myofilament proteinduring cardiac adaptation in simulated microgravity. Contractile force and velocity ofcardiac muscle were decreased in the tail suspension rats as compared with thecontrol. Ca-dependent actomyosin ATPase activity was also de-creased;however,sensitivity of cardiac muscle to Ca activation was unchanged. There was no changein expression of myosin heavy chain, tropomyosin, troponin T, or troponin I isoformsin hearts of tail suspension rats. ＜正＞Objective Advances in stem cell research have raised the hope and possibility for the therapy of neurodegenerative *** Alzheimer’s disease（AD） transgenic animal models,stem cell transplantation has been demonstrated to reverse behavioral *** recent study has demonstrated that embryonic stem（ES） cell-derived neural precursor cells improve memory dysfunction in rats with injections ofβ-amyloid peptide（1-40）（Aβ1-40） in the dorsal ***,the underlying mechanisms have remained unknown as most studies suggested that the observed behavioral improvements appeared to be related to structural reorganization such as specific neuronal differentiation and synapse *** present study was performed to test a murine ES cell-based approach in rats subjected to Aβ1 -40 injection into hippocampus dentate *** Efficacy of cell therapy regarding graft survival,neuronal yield and diversity,and synapse formation of the grafted cells were tested after transplanting ES cell-derived neural precursors into the hippocampus of adult rats with immunohistochemistry,quantum labeling and electric microscopy. Results Here,we show that grafted cells can survive,and differentiate with high yield into immunohistochemically mature glial cells and neurons of diverse *** importantly,transplanted cells demonstrate characteristics of proper synapse formation between host and grafted neural *** Our observations show that ES cell-based transplantation approach may be promising in treatment of AD.

＜正＞Objective It has been suggested that the anterior cingulate cortex（ACC） and prelimbic cortex（PL） of medial prefrontal cortex（mPFC） have critical roles in the acquisition and retrieval（expression） processes of trace eyeblink conditioning（TEC）,*** was also reported that re-training after PL lesions following initial learning could recover the impaired behavioral *** on these results,we hypothesized that ACC substituted for the impaired retrieval role of PL in *** present study is to investigate（1） the hypothesis of TEC retrieval substitution and（2） the natural roles of ACC and PL in *** Lidocaine hydrochloride（LH） or saline was microinfused into ACC,or PL,or both ACC and PL after daily pre-infusion session training following 8 days initial acquisition ***（1） Infusion of lidocaine into area 24 does not significantly affect TEC expression.（2） Area 24 infusions affect TEC acquisition as evidenced by a difference in pre-infusion and post-infusion performance between sham and lesion groups that develops over several infusion days.（3） Area 32 infusions have immediate effect on TEC expression.（4） The expression deficits with area 32 infusions fades over 5 infusion sessions,and（5） subsequent dual infusions into both regions produces immediate deficits that do not *** The results support our hypothesis of substitution and also indicate that reversible inactivation of ACC prevented the acquisition of TEC but did not affect its retrieval,whereas reversible inactivation of PL caused significant deficits in retrieval of TEC,but did not affect its acquisition.

＜正＞Neural stem cells（NSCs） could be applied for neuronal regeneration and structural brain repair only when they migrate within the CNS and reach the precise regions where neuronal replacement is *** the brain was injuried, NSCs in the hippocampus and subventricular zone（SVZ） of the adult changed their migratory route and moved toward the injury *** present study,we observed the migration and differentiation of neural precursor cells（NPCs） derived from murine embryonic stem（ES） cells when transplanted into the A β-injuried hippocampus of *** between migration distance and differentiation ratio of the engrafted NPCs was also *** ES cell line MESPU35,which expressing enhanced green fluorescent protein（EGFP）,was differentiated into nestin-positive NPCs with modified serumfree methods before *** migration distance was increased about 5 times 12 weeks *** differentiation ratio to GFAP-positive cells of migratory NPCs kept rising from about 30.41±1.45%in 4 weeks to 49.25±1.23%in 16 weeks,and differentiation ratio to NF200-positive cells was increased from 16.68±0.95%in 4 weeks to 27.94±1.21%in 16 ***,the percentage of migratory GFAP-positive cells distributed at the ipsilateral of Aβplaques were increased from 60.2%to 81.3%,and the percentage of migratory NF200-positive cells localized at the ipsilateral was increased from 61.3%to 84.1%.It showed that there was a significant positive linear correlation between migration distance and the neural differentiation ratio（correlation coefficient,r=0.991）,a significant positive linear correlation between migration distance and differentiation ratio to astrocyte（correlation coefficient,r = 0.953）.Our findings suggested that engrafted NPCs migrated targetedly to the A β-injured site and differentiated into neurons and astrocyte.

目的　构建表达绿色荧光蛋白的小鼠胚胎干细胞 (MES)克隆 ,观察基因转染对MES细胞增殖和无血清诱导为nestin阳性神经前体细胞 (NPCs)的影响。　方法　原代分离小鼠胚胎成纤维细胞并经丝裂霉素 C处理作为饲养层细胞。质粒的转化、抽提、纯化。电穿孔法基因转染 ,MES细胞克隆的NBT/BClP染色 ,NPCs的nestin免疫组织化学染色。　结果　EGFP基因转染后可得到表达绿色荧光蛋白的MES细胞克隆 ,采用N2或ITSF无血清条件培养基可将其定向诱导为nestin阳性的并能发绿色荧光的NPCs。基因转染后的MES细胞增殖、神经前体细胞诱导率和未转染组相比均无明显下降 ,但NPCs绿色荧光的表达较诱导前明显下降。　结论　基因转染对ES细胞的增殖和神经分化无明显影响。
