Induction of chronic cholestasis without liver cirrhosis-Creation of an animal model

AIM To analyze time intervals of inflammation and regeneration in a cholestatic rat liver *** In 36 Lewis rats, divided into six groups of 6 animals(postoperative observation periods: 1, 2, 3, 4, 6, 8 wk), the main bile duct was ligated with two ligatures and observed for the periods mentioned above. For laboratory evaluation, cholestasis parameters(bilirubin, γ-GT), liver cell parameters(ASAT, ALAT) and liver synthesis parameters(quick, albumin) were determined. For histological analysis, HE, Ev G, ASDCL and HMGB-1 stainings were performed. Furthermore, we used the m RNA of IL-33, GADD45 a and p-21 for analyzing cellular stress and regeneration in cholestatic *** In chemical laboratory and histological evaluation, a distinction between acute and chronic cholestatic liver injury with identification of inflammation and regeneration could be demonstrated by an increase in cholestasis(bilirubin: 1-wk group, 156.83 ± 34.12 μmol/L, P = 0.004) and liver cell parameters(ASAT: 2-wk group, 2.1 ± 2.19 μmol/L.s, P = 0.03; ALAT: 2-wk group, 1.03 ± 0.38 μmol/L.s, P = 0.03) after bile duct ligation(BDL). Histological evaluation showed an increase of bile ducts per portal field(3-wk group, 48 ± 6.13, P = 0.004) during the first four weeks after bile duct ligation. In addition to inflammation, which is an expression of acute cholestasis, there was an increase of necrotic areas in the histological sections(2-wk group, 1.38% ± 2.28% per slide, P = 0.002). Furthermore, the inflammation could be verified by ASDCL(4-wk group, 22 ± 5.93 positive cells per portal field, P = 0.041) and HMGB-1 [2-wk group, 13 ± 8.18 positive cells per field of view(Fo V), P = 0.065] staining. Therefore, in summary of the laboratory evaluation and histological studies, acute cholestasis could be found during the first four weeks after bile duct ligation. Dielectrophoresis is a powerful and well-established technique that allows label-free,non-invasive manipulation of cells and particles by leveraging their electrical *** practical implementation of the associated electronics and user interface in a biology laboratory,however,requires an engineering background,thus hindering the broader adoption of the *** order to address these challenges and to bridge the gap between biologists and the engineering skills required for the implementation of DEP platforms,we report here a custom-built,compact,universal electronic platform termed ADEPT(adaptable dielectrophoresis embedded platform tool)for use with a simple microfluidic chip containing six *** versatility of the open-source platform is ensured by a custom-developed graphical user interface that permits simple reconfiguration of the control signals to address a wide-range of specific applications:(i)precision positioning of the single bacterium/cell/particle in the micrometer range;(ii)viability-based separation by achieving a 94%efficiency in separating live and dead yeast;(iii)phenotype-based separation by achieving a 96%efficiency in separating yeast and Bacillus subtilis;(iv)cell–cell interactions by steering a phagocytosis process where a granulocyte engulfs *** RGB-S ***,the set of experiments and the platform form a complete basis for a wide range of possible applications addressing various biological questions exploiting the plug-and-play design and the intuitive GUI of ADEPT.

We report on an experiment performed at the FLASH2 free-electron laser(FEL)aimed at producing warm dense matter via soft x-ray isochoric *** the experiment,we focus on study of the ions emitted during the soft x-ray ablation process using time-of-flight electron multipliers and a shifted Maxwell–Boltzmann velocity distribution *** find that most emitted ions are thermal,but that some impurities chemisorbed on the target surface,such as protons,are accelerated by the electrostatic field created in the plasma by escaped *** morphology of the complex crater structure indicates the presence of several ion groups with varying *** find that the ion sound velocity is controlled by the ion temperature and show how the ion yield depends on the FEL radiation attenuation length in different materials.

Three different technologies for the low-temperature separation(LTS)of gas condensate from the Achimov deposits in the Russian Urengoyskoe gas and condensate field were assessed using exergy *** options examined included turbo-expansion and *** exergy values were calculated for material streams and exergy losses and efficiencies were estimated for dedicated equipment used in the *** lowest exergy loss of 4221.2 kW was obtained using turboexpansion and electricity *** carbon release associated with each scenario was calculated while considering different production rates,technological parameters and natural decreases in wellhead *** integral carbon footprint after 40 years of LTS operation was estimated for all cases.A classical ejector-based LTS scheme was shown to produce 1200 t of CO_(2)emissions over 40 years of *** same scheme combined with a turboexpander and electricity generator produced 59%less CO_(2)in the same *** expansion-cogeneration LTS scheme was found to be the most effective and ecologically friendly among the various options based on this analysis.

Tens of thousands of demoiselle cranes’crossing the Himalayas to the Indian subcontinent have been reported for decades,but their exact spring migration route remained a mystery until our previous study found they made a detour in spring along the western edge of the Himalayas and crossed the Mongolian Plateau to their breeding areas based on satellite telemetry of 3 *** corroborate the loop migration pattern and explore whether demoiselle crane’s loop migration route is shaped by time-and energy-minimization strategies in spring and autumn and how the temporal and spatial variation of environmental conditions contribute to crane’s selection of migration routes,we tracked 11 satellite-tagged demoiselle cranes from their breeding area in China and Russia,simulated 2 pseudo migration routes,and then compared the environmental conditions,time,and energy cost between true and pseudo routes in the same *** show that demoiselles’spring migration obeyed time-minimization hypothesis,avoiding the colder Qinghai-Tibet Plateau,beneﬁted by abundant food and higher thermal and orographic uplift along the route;autumn migration follows energy-minimization hypothesis with the shorter *** research will contribute to uncover the mechanical reasons why demoiselle crane avoids crossing the giant barrier of the Himalayas in spring,and shapes a loop migration route.

The study of structure, thermodynamic state, equation of state(EOS) and transport properties of warm dense matter(WDM) has become one of the key aspects of laboratory astrophysics. This field has demonstrated its importance not only concerning the internal structure of planets, but also other astrophysical bodies such as brown dwarfs, crusts of old stars or white dwarf stars. There has been a rapid increase in interest and activity in this field over the last two decades owing to many technological advances including not only the commissioning of high energy optical laser systems, zpinches and X-ray free electron lasers, but also short-pulse laser facilities capable of generation of novel particle and X-ray sources. Many new diagnostic methods have been developed recently to study WDM in its full complexity. Even ultrafast nonequilibrium dynamics has been accessed for the first time thanks to subpicosecond laser pulses achieved at new facilities. Recent years saw a number of major discoveries with direct implications to astrophysics such as the formation of diamond at pressures relevant to interiors of frozen giant planets like Neptune, metallic hydrogen under conditions such as those found inside Jupiter’s dynamo or formation of lonsdaleite crystals under extreme pressures during asteroid impacts on celestial bodies. This paper provides a broad review of the most recent experimental work carried out in this field with a special focus on the methods used. All typical schemes used to produce WDM are discussed in detail. Most of the diagnostic techniques recently established to probe WDM are also described. This paper also provides an overview of the most prominent examples of these methods used in experiments. Even though the main emphasis of the publication is experimental work focused on laboratory astrophysics primarily at laser facilities, a brief outline of other methods such as dynamic compression with z-pinches and static compression using diamond anvil cells(

Background:Cholestasis should be relieved by biliary drainage prior to major liver *** condition is often associated with bacterial colonization of the otherwise sterile biliary *** reduces the regenerative capacity of the remaining ***,targeted antibiotic therapy is a key feature in perioperative treatment in patients with perihilar cholangiocarcinoma(pCCC).Methods:Between December 1999 and December 2017,251 pCCC patients were treated in our *** total,115 patients underwent a microbiological *** addition to the characterization of the specific microorganisms and antibiotic resistance,we analyzed subgroups according to preoperative ***:Enterococci(87/254,34%)and Enterobacteria(65/254,26%)were the most frequently detected *** 43%(50/115)of patients,Enterococcus faecalis was found in the bile duct *** faecium(29/115)and Escherichia coli(29/115)were detected in 25%of *** patients with percutaneous transhepatic biliary drainage(3/8,38%)or stents(24/79,30%),Enterococcus faecium was diagnosed most frequently(P<0.05).Enterococcus faecium and Klebsiella oxytoca were significantly more frequently noted in the time period after 2012(P<0.05).With regard to fungal colonization,the focus was on various Candida strains,but these strains generally lacked ***:pCCC patients exhibit specific bacterial colonization features depending on the type of preoperative biliary ***,targeted antibiosis should be applied in this patient cohort to minimize the risk of biliary complications after major liver *** our cohort,the combination of meropenem and vancomycin represents an effective perioperative medical approach.
