Expression of Recombinant Human FADD,Preparation of Its Polyclonal Antiserum and the Application in Immunoassays

The wild-type human Fas-associated death domain （FADD） protein was expressed as a His-tag fusion protein in Escherichia coll. Recombinant FADD proteins were purified under the denatured condition. After denatured protein purification, it was refolded and obtained at a yield of about 23 mg/L. Purified FADD exhibited as a homogenous band corresponding to the molecular weight of 31 kDa. Immunization of rabbits against the refolded FADD protein was allowed the production of high titre polyclonal antiserum. This new polyelonal antibody could recognize recombinant FADD protein in Western blot. Immunoreactivity was also observed in immunofiuorescence assay. The low cost polyclonal antiserum was applicable to extensive detection of FADD in various immunoassays. Cellular ＆ Molecular Immunology. 2008;5（6）：471-474.

Post-harvest storage of lime fruits (Citrus aurantifolia) following high humidity and low temperature in a modified brick wall cooler

The use of a modified brick wall Evaporative Cooling Chamber(ECC)for extending the shelf life of limes(Citrus aurantifolia)was compared with ambient *** shelf life inside of the ECC was extended by five to twenty days relative to ambient *** inside temperatures were about 4-6℃less than the ambient temperatures,and humidity was about 10%-20%*** was conducted to check the weight loss and freshness of lime fruits,and data were observed *** loss in limes kept inside the chamber for ten days was 1.8%and in ambient conditions it was around 9.8%.The color of the fruit changed to yellowish inside the ECC and the control which was kept outside became *** storage did not adversely affect fruit firmness,or the percentage of soluble solids or titratable acidity of the *** modified ECC does not require mechanical or electrical energy input and can be constructed with locally available material by unskilled labour.

Metallothionein 2A(MT2A)is a small stress response protein that can be induced by exposureto toxic *** is highly expressed in breast cancer *** this study,the cDNA encoding the humanMT2A protein was expressed as glutathione S-transferase(GST)fusion protein in Escherichia *** MT2A proteins were loaded onto 12% sodium dodecylsulfate-polyacrylamide gel and separatedby electrophoresis,the recombinant protein was visualized by Coomassie blue staining and the 33 kDarecombinant GST-MT2A fusion protein band was cut out from the *** gel slice was minced and used togenerate polyclonal *** of rabbit against MT2A protein allowed the production of hightiter polyclonal *** new polyclonal antibody recognized recombinant MT2A protein in Westernblot *** low-cost antibody will be useful for detection in various immuno-assays.
