Targeted approaches for HER2 breast cancer therapy: News from nanomedicine?

About 30% of human breast cancers are human epidermal growth factor receptor 2 （HER2）＋. This particular biological portrait is characterized by the overexpression of HER2 receptor with the subsequent deregulation of downstream pathways, which control cellular survival and proliferation. The most effective treatment for HER2＋ cancer is represented by therapy with HER2-targeted agents. Anti-HER2 therapy dramatically improves clini-cal outcomes, although it shows some limitations in achieving a proper treatment. These drawbacks of HER2-targeted therapy may be overcome with the develop-ment of HER2-targeted drug delivery nanodevices. These nanoparticles possess an internal three-dimensional com-partimentalization, which allows to combine the specifc target recognition with their capability to act as a drug reservoir for the selective delivery of chemotherapics to tumor sites. Moreover, nanoparticles useful in photo-thermal ablation or in photodynamic therapy have been functionalized in order to match specifcity in tumor cell recognition and suitable chemical properties. Here, we summarize the state of the art concerning the HER2＋ breast cancer and anti-HER2 therapy, in particular deep-ening the contribution of the nanomedicine. Description of preclinical studies performed with HER2-targeted nanoparticles for HER2＋ breast cancer therapy will be preceded by an overview on HER2-targeting molecules and nano-conjugation strategies. Further investigation will be necessary to introduce these nano-drugs in clinical prac-tice; however promising results encourage an upcoming translation of this research for the next future.

Dear Editor,Microcalcifications(MCs)are common findings in mammography and can be indicative of different degrees of malignancy,thus requiring multiple stereotaxis vacuumassisted biopsies under mammography to identify and characterize breast cancer[1,2].In many cases,however,the presence of MCs is not associated with the presence of a ***,a better understanding of the physical-chemical properties of MCs is needed to assess the relationship between their presence and breast cancer.

Abstract： In HIV-1 management, eradication of the virus from sanctuaries represents a major and challenging goal. The genital tract, gut associated lymphoid tissue, lymph nodes, central nervous system, macrophages and latently infected CD4＋ T lymphocytes are typical sites where H1V-1 compartmentalizes. To circumvent this problem, a consistent number of studies have focused on improving ARVs （antiretroviral drugs） delivery into sanctuary sites and different nanoteehnological approaches have been developed. Cellular HIV-1 sanctuaries （i.e. macrophages） can be reached by nanoformulation of ARVs or by activation of latently infected cells. Anatomical sanctuaries （i.e. brain or male genital tract） can be addressed by increasing the permeation of ARVs across tissue barriers, such as the blood-brain barrier or the blood-testis barrier, while ARVs concentration in lymph nodes can be enhanced by drug encapsulation in CD4-targeted nanoparticles.

Environmental parameters assessment of a new diffuser for air cooling/heating system:Measurements and numerical validation

Air distribution of HVAC systems is the most popular type used in the building sector,having a relevant impact on indoor air quality and occupant *** types of research developed optimal solutions for the HVAC system’s design,focusing on specific components of the distribution system,on the airflow and geometry of ducts,on the size of ducts,on the shape and position of air ***,few works in literature proposed a globally experimental and simulation analysis of an air distribution system with a variable mass flaw *** this line,the presented research investigates the potentialities of a new ceiling diffuser,installed in an exhibition *** system provides a variable mass flow rate thanks to its configuration,providing adequate thermal comfort.A warm wall is chosen as the heating *** tests are carried out,six for cooling and two for heating with different volumetric air rates and supply air temperature of the *** combination of two methods,the measurement campaigns and the computational fluid dynamics(CFD)technique represent a suitable approach to examine the thermal indoor *** general,results show a strong capability of this diffuser to provide a uniform temperature and velocity field inside the ***,experimental and numerical data are significantly comparable with an average deviation of 1%for the velocity and lower 1%for the temperature,guaranteeing an optimal distribution of the understudy environmental parameters on the vertical and horizontal planes.

Air quality nearby road traffic tunnel portals: BTEX monitoring

A monitoring campaign of BTEX （benzene, toluene, ethylbenzene, o- m- and p-xylene） was carried out nearby two tunnel portals in the urban area of Naples with the aim to verify air quality in this kind of urban sites. Sampling was carried out using the active adsorption technique. Sampling time was 1 h. Ambient temperature and traffic flow measurements were carried out during each sampling operation. The results indicate that average benzene concentrations at both sites exceed the limit value of 10 μg/Nm^3 established by the European Community （EC）（Dir. 2000/69）. Concentration levels of other BTEX are relatively high as well. A correlation between BTEX concentration and two wheeler vehicle flow was observed.

A refined Frequency Domain Decomposition tool for structural modal monitoring in earthquake engineering

Output-only structural identification is developed by a refined Frequency Domain Decomposition（rFDD） approach, towards assessing current modal properties of heavy-damped buildings（in terms of identification challenge）, under strong ground motions. Structural responses from earthquake excitations are taken as input signals for the identification algorithm. A new dedicated computational procedure, based on coupled Chebyshev Type Ⅱ bandpass filters, is outlined for the effective estimation of natural frequencies, mode shapes and modal damping ratios. The identification technique is also coupled with a Gabor Wavelet Transform, resulting in an effective and self-contained time-frequency analysis framework. Simulated response signals generated by shear-type frames（with variable structural features） are used as a necessary validation condition. In this context use is made of a complete set of seismic records taken from the FEMA P695 database, i.e. all 44 ＂Far-Field＂（22 NS, 22 WE） earthquake signals. The modal estimates are statistically compared to their target values, proving the accuracy of the developed algorithm in providing prompt and accurate estimates of all current strong ground motion modal parameters. At this stage, such analysis tool may be employed for convenient application in the realm of Earthquake Engineering, towards potential Structural Health Monitoring and damage detection purposes.

Variable-order fracture mechanics and its application to dynamic fracture

This study presents the formulation,the numerical solution,and the validation of a theoretical framework based on the concept of variable-order mechanics and capable of modeling dynamic fracture in brittle and quasi-brittle *** specifically,the reformulation of the elastodynamic problem via variable and fractional-order operators enables a unique and extremely powerful approach to model nucleation and propagation of cracks in solids under dynamic *** resulting dynamic fracture formulation is fully evolutionary,hence enabling the analysis of complex crack patterns without requiring any a priori assumption on the damage location and the growth path,and without using any algorithm to numerically track the evolving crack *** evolutionary nature of the variable-order formalism also prevents the need for additional partial differential equations to predict the evolution of the damage field,hence suggesting a conspicuous reduction in complexity and computational ***,the variable-order formulation is naturally capable of capturing extremely detailed features characteristic of dynamic crack propagation such as crack surface roughening as well as single and multiple *** accuracy and robustness of the proposed variableorder formulation are validated by comparing the results of direct numerical simulations with experimental data of typical benchmark problems available in the literature.

Shoreline evolution and modern beach sand composition along a coastal stretch of the Tyrrhenian Sea, southern Italy

This contribution focuses on a multidisciplinary research showing the geomorphological evolution and the beach sand composition of the Tyrrhenian shoreline between Capo Suvero promontory and Gizzeria Lido village(Calabria,southern Italy). The aim of the geomorphological analysis was to reconstruct the evolutionary shoreline stages and the present-day sedimentary dynamics along approximately 6 km of coastline. The results show a general trend of beach nourishment during the period 1870–2019. In this period, the maximum shoreline accretion value was estimated equal to + 900 m with an average rate of + 6.5 m/yr. Moreover, although the general evolutionary trend is characterized by a remarkable accretion, the geomorphological analysis highlighted continuous modifications of the beaches including erosion processes. The continuous beach modifications occurred mainly between 1953 and1983 and were caused mainly by human activity in the coastal areas and inside the hydrographic basins. The beach sand composition allowed an assessment of the mainland petrological sedimentary province and its dispersal pattern of the present coastal dynamics. Petrographic analysis of beach sands identified a lithic metamorphi-clastic petrofacies, characterized by abundant fine-grained schists and phyllites sourced from the crystalline terrains of the Coastal Range front and carried by the Savuto River. The sand is also composed of a mineral assemblage comparable to that of the Amato River provenance. In terms of framework detrital constituents of QFL(quartz:feldspars:aphanitic lithic fragments) and of essential extraclasts, such as granitoid:sedimentary:metamorphic phaneritic rock fragments(Rg:Rs:Rm), sand maturity changes moderately from backshore to shoreface, suggesting that transport processes had a little effect on sand maturity.
