Inflammatory bowel diseases(IBD)are chronic inflammatory disorders of the gastrointestinal tract associated with multifactorial conditions such as ulcerative colitis and Crohn’s *** the underlying mechanisms of IBD remain unclear,growing evidence has shown that dysregulated immune system reactions in genetically susceptible individuals contribute to mucosal ***,conventional treatments have been effective in inducing remission of IBD but not in preventing the relapse of *** this way,mesenchymal stromal cells(MSC)therapy has been recognized as a promising treatment for IBD due to their immunomodulatory properties,ability to differentiate into several tissues,and homing to inflammatory *** so,literature is conflicted regarding the location and persistence of MSC in the body after *** this reason,recent studies have focused on the paracrine effect of the biofactors secreted by MSC,especially in relation to the immunomodulatory potential of soluble factors(cytokines,chemokines,and growth factors)and extracellular vehicles that are involved in cell communication and in the transfer of cellular material,such as proteins,lipids,and nucleic ***,treatment with interferon-γ,tumor necrosis factor-α,and interleukin-1βcauses MSC to express immunomodulatory molecules that mediate the suppression via cell-contact dependent *** together,we present an overview of the role of bioactive factors and cell membrane proteins derived from MSC as a cell-free therapy that can improve IBD treatment.

Mesenchymal stromal cells (MSCs) are multipotent and self-renewing stem cellsthat have great potential as cell therapy for autoimmune and inflammatorydisorders, as well as for other clinical conditions, due to their immunoregulatoryand regenerative properties. MSCs modulate the inflammatory milieu by releasingsoluble factors and acting through cell-to-cell mechanisms. MSCs switch theclassical inflammatory status of monocytes and macrophages towards a nonclassicaland anti-inflammatory phenotype. This is characterized by an increasedsecretion of anti-inflammatory cytokines, a decreased release of pro-inflammatorycytokines, and changes in the expression of cell membrane molecules and inmetabolic pathways. The MSC modulation of monocyte and macrophage phenotypesseems to be critical for therapy effectiveness in several disease models, sincewhen these cells are depleted, no immunoregulatory effects are observed. Here,we review the effects of living MSCs (metabolically active cells) and metabolicallyinactive MSCs (dead cells that lost metabolic activity by induced inactivation) andtheir derivatives (extracellular vesicles, soluble factors, extracts, and microparticles)on the profile of macrophages and monocytes and the implications forimmunoregulatory and reparative processes. This review includes mechanisms ofaction exhibited in these different therapeutic appro-aches, which induce the antiinflammatoryproperties of monocytes and macrophages. Finally, we overviewseveral possibilities of therapeutic applications of these cells and their derivatives,with results regarding monocytes and macrophages in animal model studies andsome clinical trials.

Movement behaviors and their association with depressive symptoms in Brazilian adolescents:A cross-sectional study

Background:Physical activity,sleep,and sedentary behaviors compose 24-h movement behaviors and have been independently associated with depressive ***,it is not clear whether it is the movement behavior itself or other contextual factors that are related to depressive *** objective of the present study was to examine the associations between self-reported and accelerometer-measured movement behaviors and depressive symptoms in ***:Cross-sectional data from 610 adolescents(14-18 years old)were *** answered questions from the Center for Epidemiological Studies Depression scale and reported time spent watching videos,playing videogames,using social media,time spent in various physical activities,and daytime ***-worn accelerometers were used to measure sleep duration,sleep efficiency,sedentary time,and physical ***-effects logistic regressions were ***:Almost half of the adolescents(48%)were classified as being at high risk for depression(score≥20).No significant associations were found between depressive symptoms and accelerometer-measured movement behaviors,self-reported non-sport physical activity,watching videos,and playing ***,higher levels of self-reported total physical activity(odd ratio(OR)=0.92,95%confidence interval(95%CI):0.86-0.98)and volume of sports(OR=0.88,95%CI:0.79-0.97),in minutes,were associated with a lower risk of depression,while using social media for either 2.0-3.9 h/day(OR=1.77,95%CI:1.58-2.70)or>3.9 h/day(OR=1.67,95%CI:1.10-2.54),as well as higher levels of daytime sleepiness(OR=1.17,95%CI:1.12-1.22),were associated with a higher risk of ***:What adolescents do when they are active or sedentary may be more important than the time spent in the movement behaviors because it relates to depressive *** daytime sleepiness,promoting sports,and limiting social media use may benefit adolescents.

Antiglycation and antioxidant activities of the crude extract and saponin fraction of Tribulus terrestris before and after microcapsule release

Objective:The present study investigated antiglycation and antioxidant activities of crude dry extract and saponin fraction of Tribulus *** also developed a method of microencapsulation and evaluated antiglycation and antioxidant activities of crude dry extract and saponin fraction before and after microcapsule ***:Antiglycation activity was determined by relative electrophoretic mobility(REM),free amino groups and inhibition of advanced glycation end-product(AGE)*** activity was determined by 2,2-diphenyl-1-picrylhydrazyl(DPPH),ferric ion-reducing antioxidant power(FRAP),nitric oxide(NO)and thiobarbituric acid reactive species(TBARS)*** were prepared using maltodextrin as wall material and freeze-drying as encapsulation *** characterization of microcapsules was evaluated by scanning electron microscopy,and encapsulation efficiency and microcapsule release were determined by total saponins *** and antioxidant assays were performed using crude dry extract and saponin fraction of *** before and after ***:Saponin fraction showed an increase of 32.8%total ***-performance liquid chromatography-mass spectrometry analysis showed the presence of saponins in the obtained *** evaluation by REM demonstrated that samples before and after release presented antiglycation activity similar to bovine serum albumin treated with ***,samples inhibited AGE formation,highlighting treatment with saponin fraction after release(89.89%).Antioxidant tests demonstrated antioxidant activity of the *** dry extract before encapsulation presented the highest activities in DPPH(92.00%)and TBARS(32.49%)*** fraction before encapsulation in FRAP test(499μmol Trolox equivalent per gram of dry sample)and NO test(15.13μmol nitrite formed per gram of extract)presented the highest ***:This study presen

Reconstruction of the Neutron Flux in a Slab Reactor

In this electronic article we use the one-dimensional multigroup neutron diffusion equation to reconstruct the neutron flux in a slab reactor from the nuclear parameters of the reactor, boundary and symmetry condition, initial flux and?keff. The diffusion equation was solved analytically for one single homogeneous fuel region and for two regions considering fuel and reflector. To validate the method proposed, the results obtained in this article were compared using reference methods found in the literature.

The historic centres revitalization addresses the challenges related to the preservation of fundamental heritage values. At a time, when everyone looks with concern to our cities＇ future, it is important to reflect on the received heritage, seeking the most appropriate answers to the planning of the historic centres. These fabrics are reference places in the urban space, due to their role of memorial testimony and of generators of cultural and economic dynamics. However, often times, inherited urban fabrics are affected by the limitations of the heritage policies which, for being too general and based on theoretical and abstract frameworks, have difficulty incorporating the characteristics of each area and neglect the formulation of specific criteria and intervention methods. The purpose of this paper is to provide a comparative reading of the levels of urban renewal allowed by the planning tools. This study chooses two historic centers in Portugal： Oporto and Guimarfies historic centres （World Heritage Sites since 1996 and 2001, respectively, and were the last to get this classification in Portugal）. This reflection is a contribution to peer trends and raise the discussion on the role that the different heritage policies have to the revitalization of the historic centres.

Dynamic culture improves MSC adhesion on freeze-dried bone as a scaffold for bone engineering

AIM: To investigate the interaction between mesenchymal stem cells (MSCs) and bone grafts using two different cultivation methods: static and dynamic. METHODS: MSCs were isolated from rat bone marrow. MSC culture was analyzed according to the morphology, cell differentiation potential, and surface molecular markers. Before cell culture, freeze-dried bone (FDB) was maintained in culture for 3 d in order to verify culture medium pH. MSCs were co-cultured with FDB using two different cultivation methods: static co-culture (two-dimensional) and dynamic co-culture (three-dimensional). After 24 h of cultivation by dynamic or static methods, histological analysis of Cell adhesion on FDB was performed. Cell viability was assessed by the Trypan Blue exclusion method on days 0, 3 and 6 after dynamic or static culture. Adherent cells were detached from FDB surface, stained with Trypan Blue, and quantified to determine whether the cells remained on the graft surface in prolonged non-dynamic culture. Statistical analyses were performed with SPSS and a P 0.05). The objective of this work was to evaluate the effect of different water deficiency and rehydration levels on the concentrations of osmoregulators in two plant species（Hymenaea courbaril and H. Stigonocarpa） in the Amazon. We adopted a 2×5×5 factorial system,referring to 2 species（H. courbaril and H. stigonocarpa）and 5 stages of hydration and rehydration. The five hydration and rehydration stages were established in:（1）Control treatment E0;（2） Plants with 13 days of stress after incubation—E13;（3） Plants with 26 days of stress E26;（4）The plants that were established after 26 days after incubation and rehydrated for two days（RD2）;（5） rehydrated for two days（RD4）. The plants that were established after 26 days after incubation and rehydrated for four days. The experiment totaled fifty young plants with five *** measurements were performed at the beginning of the experiment（E0） at 13（E13） and 26（E26） days after the water stress, in which the plants were rehydrated,repeating the analyses after two（RD2） and four（RD4）days. Both species increased the sucrose concentration by18%, with a decrease of 52% in starch content. The RD4 time presented the highest mean starch concentration(0.19 mmol g-1 of the residue for H. courbaril and0.27 mmol g;of residue for H. stigonocarpa). Increased proline concentrations were recorded for controls until RD2 for both species. For glycine betaine, the highest increases in treatments E26 and RD2 were observed for the H. courbaril species. Our rehydration period was not sufficient for total recovery of pre-stress concentrations of all studied solutes.
