The prediction accuracy of a simulation method is limited by its theoretical background. This fact can lead to disadvantages regarding the simulation quality when investigating systems of high complexity, e.g. containing components showing a fairly different behavior. To overcome this limitation, co-simulation approaches are used more and more, combining the advantages of different simulation disciplines. That is why we propose a new strategy for the dynamic simulation of cutting processes. The method couples Lagrangian particle methods, such as the smoothed particle hydrodynamics （SPH） method, and multibody system （MBS） tools using co-simulations. We demonstrate the capability of the new approach by providing simulation results of an orthogonal cutting process and comparing them with experimental data. @ 2013 The Chinese Society of Theoretical and Applied Mechanics. Residential short-term electric load forecasting is essential in modern decentralized power *** forecasting methods mostly rely on neural networks and require access to private and sensitive electric load data for model *** neural network training aggregates all data on a centralized server to train one global ***,the aggregation of user data introduces security and data privacy *** contrast,this study investigates the modern neural network training methods of federated learning and model personalization as potential solutions to security and data privacy *** an extensive simulation approach,the investigated methods are compared to the conventional centralized method and a pre-trained baseline predictor to compare their respective *** study identifies that the underlying data structure of electric load data has a significant influence on the loss of a *** therefore conclude that a comparison of loss distributions will in fact be considered a comparison of data structures,rather than a comparison of the model *** an alternative method of comparison of loss values,this study develops the"differential comparison".The method allows for the isolated comparison of model loss differences by only comparing the losses of two models generated by the same data sample to build a distribution of *** differential comparison method was then used to identify model personalization as the best performing model training method for load forecasting among all analyzed methods,with a superior performance in 59.1%of all cases.

Riboflavin （Rf） receptors bind and translocate Rf and its phosphorylated forms （e.g. flavin mononucleotide, FMN） into cells where they mediate various cellular metabolic pathways. Previously, we showed that FMN-coated ultrasmall superparamagnetic iron oxide （FLUSPIO） nanoparticles are suitable for labeling metabolically active cancer and endothelial cells in vitro. In this study, we focused on the in vivo application of FLUSPIO using prostate cancer xenografts. Size, charge, and chemical composition of FLUSPIO were evaluated. We explored the in vitro specificity of FLUSPIO for its cellular receptors using magnetic resonance imaging （MRI） and Prussian blue staining. Competitive binding experiments were performed in vivo by injecting free FMN in excess. Bio-distribution of FLUSPIO was determined by estimating iron content in organs and tumors using a colorimetric assay. AFM analysis and zeta potential measurements revealed a particulate morphology approximately 20-40 nm in size and a negative zeta potential （-24.23±0.15 mV） in water. X-ray photoelectron spectroscopy and time-of-flight secondary ion mass spectrometry data confirmed FMN present on the USPIO nanoparticle surface. FLUSPIO uptake in prostate cancer cells and human umbilical vein endothelial cells was significantly higher than that of control USPIO, while addition of excess of free FMN reduced accumulation. Similarly, in vivo MRI and histology showed specific FLUSPIO uptake by prostate cancer cells, tumor endothelial cells, and tumor-associated macrophages. Besides prominent tumor accumulation, FLUSPIO accumulated in the liver, spleen, lung, and skin. Hence, our data strengthen our hypothesis that targeting riboflavin receptors is an efficient approach to accumulate nanomedicines in tumors opening perspectives for the development of diagnostic and therapeutic systems.

Fluorescent reporter proteins that allow repeated switching between a fluorescent and a non-fluorescent state in response to specific wavelengths of light are novel tools for monitoring of protein trafficking and super-resolu- tion fluorescence microscopy in living organisms. Here, we describe variants of the reversibly photoswitchable fluores- cent proteins rsFastLime, bsDronpa, and Padron that have been codon-optimized for the use in transgenic Arabidopsis plants. The synthetic proteins, designated rsFastLIME-s, bsDRONPA-s, and PADRON C-s, showed photophysical properties and switching behavior comparable to those reported for the original proteins. By combining the ＇positively switchable＇ PADRON C-s with the ＇negatively switchable＇ rsFastLIME-s or bsDRONPA-s, two different fluorescent reporter proteins could be imaged at the same wavelength upon transient expression in Nicotiana benthamiana cells. Thus, co-localiza- tion analysis can be performed using only a single detection channel. Furthermore, the proteins were used to tag the RNA-binding protein AtGRP7 （Arabidopsis thaliana glycine-rich RNA-binding protein 7） in transgenic Arabidopsis plants. Because the new reversibly photoswitchable fluorescent proteins show an increase in signal strength during each pho- toactivation cycle, we were able to generate a large number of scans of the same region and reconstruct 3-D images of AtGRP7 expression in the root tip. Upon photoactivation of the AtGRP7：rsFastLIME-s fusion protein in a defined region of a transgenic Arabidopsis root, spreading of the fluorescence signal into adjacent regions was observed, indicating that movement from cell to cell can be monitored. Our results demonstrate that rsFastLIME-s, bsDRONPA-s, and PADRON C-s are versatile fluorescent markers in plants, Furthermore, the proteins also show strong fluorescence in mammalian cells including COS-7 and HeLa cells.

Dielectrophoresis is a powerful and well-established technique that allows label-free,non-invasive manipulation of cells and particles by leveraging their electrical *** practical implementation of the associated electronics and user interface in a biology laboratory,however,requires an engineering background,thus hindering the broader adoption of the *** order to address these challenges and to bridge the gap between biologists and the engineering skills required for the implementation of DEP platforms,we report here a custom-built,compact,universal electronic platform termed ADEPT(adaptable dielectrophoresis embedded platform tool)for use with a simple microfluidic chip containing six *** versatility of the open-source platform is ensured by a custom-developed graphical user interface that permits simple reconfiguration of the control signals to address a wide-range of specific applications:(i)precision positioning of the single bacterium/cell/particle in the micrometer range;(ii)viability-based separation by achieving a 94%efficiency in separating live and dead yeast;(iii)phenotype-based separation by achieving a 96%efficiency in separating yeast and Bacillus subtilis;(iv)cell–cell interactions by steering a phagocytosis process where a granulocyte engulfs *** RGB-S ***,the set of experiments and the platform form a complete basis for a wide range of possible applications addressing various biological questions exploiting the plug-and-play design and the intuitive GUI of ADEPT.

Hepatic steatosis as the most prevalent liver disorder can either be related to alcoholic liver disease (ALD) or non-alcoholic fatty liver disease (NAFLD). In both conditions, hepatocytes excessively accumulate fatcontaining vacuoles within their cytoplasm, which is the key histological feature. In contrast to ALD, NAFLD is commonly associated with metabolic syndrome, obesity and insulin resistance. To determine increased liver fat content, liver biopsy is currently considered the gold standard. Besides the invasive technique, various other non-invasive techniques have been developed, such as ultrasound, computed tomography (CT), magnetic resonance spectroscopy (MRS) and magnetic resonance imaging (MRI) based methods. Among these techniques, ultrasound and CT provide only qualitative information about hepatic steatosis, whereas MRS-or MRI-based methods are able to determine even small amounts of fat accurately. These non-invasive magnetic resonance techniques have already proven their great potential, especially in longitudinal and cross-sectional studies regarding various metabolic conditions and medical treatment regimens. In this review, the most common, non-invasive MRS/MRI techniques for assessment of intrahepatic lipid content are described with their inherent advantages and limitations.

The open science movement has gained significant momentum within the last few *** comes along with the need to store and share research artefacts,such as publications and research *** this purpose,research repositories need to be established.A variety of solutions exist for implementing such repositories,covering diverse features,ranging from custom depositing workflows to social media-like *** this article,we introduce the FAIREST principles,a framework inspired by the well-known FAIR principles,but designed to provide a set of metrics for assessing and selecting solutions for creating digital repositories for research *** goal is to support decision makers in choosing such a solution when planning for a repository,especially at an institutional *** metrics included are therefore based on two pillars:(1)an analysis of established features and functionalities,drawn from existing dedicated,general purpose and commonly used solutions,and(2)a literature review on general requirements for digital repositories for research artefacts and related *** further describe an assessment of 11 widespread solutions,with the goal to provide an overview of the current landscape of research data repository solutions,identifying gaps and research challenges to be addressed.
