Genome-wide identification and characterization of the PbrATG family in Pyrus bretschneideri and functional analysis of PbrATG1a in response to Botryosphaeria dothidea

The pear is an economic fruit that is widely planted around the world and is loved by people for its rich nutritional value. Autophagy is a self-protection mechanism in eukaryotes, and its occurrence often accompanied by the degradation of damaged substances in cells and the recycling of nutrients. Autophagy is one of the mechanisms through which plants respond to environmental stress and plays an important role in plant development and stress resistance. functional studies of autophagy-related genes (ATGs) have been performed on a variety of plant species, but little information is available on the ATG family in pear (Pyrus bretschneideri Rehd). Therefore, we analyzed the evolutionary dynamics and performed a genome-wide characterization of the PbrATG gene family. A total of 28 PbrATG members were *** analysis showed that PbrATGs were more closely related to ATGs of European pear and apple. Evolutionary analysis revealed that whole-genome duplication (WGD) and dispersed duplication events were the main driving forces of PbrATG family *** analysis of different pear tissues showed that all the genes were expressed in different pear tissues, and different PbrATGs are expressed at different times and in different locations. Moreover, all PbrATGs also responded to different abiotic stresses, especially salt and drought stress, which elicited the highest expression levels. Pear seedlings were subsequently infected with Botryosphaeria dothidea (***). The results showed that different PbrATGs had different expression patterns at different infection stages. According to the gene expression data, PbrATG1a was selected as a key autophagy gene for further analysis. Silencing of PbrATG1a reduced the resistance of pear to B. dothidea, which resulted in increased lesions, reactive oxygen species (ROS) contents, antioxidant enzyme activity, and gene expression levels in the silenced pear seedlings after B. dothidea inoculation. 学术规范的法律制度体系构建,从法理逻辑上讲具有消除各种学术不端行为产生的不当得利、预防学术不端行为对社会造成的损害,以及构建良好的学术秩序等功能。然而,现有学术规范制度体系在防止学术不端行为的效果方面明显存在重大不足,包括缺乏良好的外部制度环境、学术机构的学术自律严重不足,以及学术规范的制度体系缺失等诸多方面,从而成为当前学术不端行为屡禁不止的重要原因。通过对这些原因进行分析,并借鉴域外同类法律制度体系的经验,可以发现构建良好的学术规范法律制度体系应当包括建立公平的学术成果评价审核制度体系、建立严格的学术机构学术规范自律制度体系,以及建立严密的学术规范法律责任体系等主要内容,构建这样的体系才能有效遏制学术不端行为可能对技术进步和社会发展产生的破坏性作用,促进良好学术氛围的形成。

Study on the clogging mechanism of punching screen in sand control by the punching structure parameters

As an independent sand control unit or a common protective shell of a high-quality screen,the punching screen is the outermost sand retaining unit of the sand control pipe which is used in geothermal well or oil and gas ***,most screens only consider the influence of the internal sand retaining medium parameters in the sand control performance design while ignoring the influence of the plugging of the punching screen on the overall sand retaining performance of the *** explore the clogging mechanism of the punching screen,this paper established the clogging mechanism calculation model of a single punching screen sand control unit by using the computational fluid mechanics-discrete element method(CFD-DEM)combined *** to the combined motion of particles and fluids,the influence of the internal flow state on particle motion and accumulation was *** results showed that(1)the clogging process of the punching sand control unit is divided into three stages:initial clogging,aggravation of clogging and stability of *** the initial stage of blockage,coarse particles form a loose bridge structure,and blockage often occurs preferentially at the streamline gathering place below chamfering inside the sand control *** the stage of blockage intensification,the particle mass develops into a relatively complete sand bridge,which develops from both ends of the opening to the center of the *** the stable plugging stage,the sand deposits show a“fan shape”and form a“V-shaped”gully inside the punching slot element.(2)Under a certain reservoir particle-size distribution,The slit length and opening height have a large influence on the permeability and blockage rate,while the slit width size has little influence on the permeability and blockage *** microscopic clogging mechanism and its law of the punching screen prevention unit are proposed in this study,which has some field guidance significance for the design of punching screen

Improved forecasting via physics-guided machine learning as exemplified using“21·7”extreme rainfall event in Henan

As a natural disaster,extreme precipitation is among the most destructive and influential,but predicting its occurrence and evolution accurately is very challenging because of its rarity and *** the example of the“21·7”extreme precipitation event(17–21 July 2021)in Henan Province,this study explores the potential of using physics-guided machine learning to improve the accuracy of forecasting the intensity and location of extreme *** physics-guided ways of embedding physical features,fusing physical model forecasts and revised loss function are used,i.e.,(1)analyzing the anomalous circulation and thermodynamical factors,(2)analyzing the multi-model forecast bias and the associated underlying reasons for it,and(3)using professional forecasting knowledge to design the loss function,and the corresponding results are used as input for machine learning to improve the forecasting *** results indicate that by learning the relationship between anomalous physical features and heavy precipitation,the forecasting of precipitation intensity is improved significantly,but the location is rarely adjusted and more false alarms *** reasons for this are as *** anomalous features used here mainly contain information about large-scale systems and factors which are consistent with the model precipitation deviation;moreover,the samples of extreme precipitation are sparse and so the algorithm used here is ***,by combining“good and different”multi models with machine learning,the advantages of each model are extracted and then the location of the precipitation center in the forecast is improved ***,by combining the appropriate anomalous features with multi-model fusion,an integrated improvement of the forecast of the rainfall intensity and location is ***,this study is a novel exploration to improve the refined forecasting of heavy precipitation with extreme intensity and high variability,

Precise immobilization of metal single atoms into a porphyrinic metal-organic framework for an efficient alkene hydrosilylation

Alkene hydrosilylation is one of the most concise and atom-economical methods to synthesize organosilicon ***,we reported the precise immobilization of metal single atoms(M-SAs;M=Ru,Rh,Ir,Pd,Pt,and Au)into a porphyrinic metalorganic framework(MOF)of PCN-222(PCN=porous coordination network),and then applied the resultant MOF composites of M-SAs@PCN-222 to alkene *** solvent-free conditions,Pt-SAs@PCN-222 displayed an especially high catalytic efficiency with the turnover frequency up to 119 s−1 and the maximum turnover number of 906,250 at room *** and theoretical studies revealed that there existed strong interactions between Pt-SAs@PCN-222 and the substrates,which helped to condense the substrates in the cavities of the porous *** density functional theory calculations and molecular dynamics simulations disclosed that PCN-222 could transfer electrons to Pt-SAs to enhance the silane oxidative addition and drive the reaction to proceed smoothly via Chalk–Harrod pathway.

Differences in insomnia-related self-reported outcomes among elderly hospitalized patients

Insomnia is among the most common sleep disorders *** in older adults is a social and public health *** affects the physical and mental health of elderly hospitalized patients and can aggravate or induce physical *** subjective feelings and providing reasonable and standardized care for elderly hospitalized patients with insomnia are urgent *** To explore the differences in self-reported outcomes associated with insomnia among elderly hospitalized *** One hundred patients admitted to the geriatric unit of our hospital between June 2021 and December 2021 were included in this ***-reported symptoms were assessed using the Athens Insomnia Scale(AIS),Generalized Anxiety Disorder Scale-7(GAD-7),Geriatric Depression Scale-15(GDS-15),Memorial University of Newfoundland Scale of Happiness(MUNSH),Barthel Index Evaluation(BI),Morse Fall Scale(MFS),Mini-Mental State Examination,and the Short Form 36 Health Survey Questionnaire(SF-36).Correlation coefficients were used to analyze the correlation between sleep quality and self-reported *** of insomnia was analyzed using Logistic regression *** Nineteen patients with AIS≥6 were included in the insomnia group,and the incidence of insomnia was 19%(19/100).The remaining 81 patients were assigned to the non-insomnia *** were significant differences between the two groups in the GDA-7,GDS-15,MUNSH,BI,MFS,and SF-36 items(P<0.05).Patients in the insomnia group were more likely to experience anxiety,depression,and other mental illnesses,as well as difficulties with everyday tasks and a greater risk of falling(P<0.05).Subjective well-being and quality of life were poorer in the insomnia group than in the control *** AIS scores positively correlated with the GAD-7,GDS-15,and MFS scores in elderly hospitalized patients with insomnia(P<0.05).Logistic regression analysis showed that GDS-15≥5 was an independent risk factor for ins

超表面由亚波长尺度排列的人工原子阵列组成,在调控光场相位、振幅、偏振等方面具有巨大优势.受离散采样原理和周期性假设的限制,传统正向设计方法不可避免地存在设计误差,容易导致器件性能下降.本文采用基于伴随的多目标拓扑优化方法,逆向设计了一种具有大偏折角度、偏振不敏感特性的自由形状波分复用超光栅.仿真结果表明,相比于离散规则结构,拓扑优化的波分复用超光栅具有更优越的偏振不敏感性能.此外,该结构对510 nm入射光的偏折角度可达70.8°,其绝对偏折效率高达48%;对于852 nm入射光,其透射效率为98%.在此基础上,通过使用随机初始结构可将绝对偏折效率优化至70%以上.本文设计的自由形状波分复用超光栅具有偏折角度大、效率高和空间串扰低等优点,在光通信、微纳光场调控、基于里德堡原子的微波测量等领域具有潜在应用前景.
