Characterization of Polysaccharides Extracted from a Cultivated Brown Alga Costaria costata During the Harvest Period

In this study, variations in composition and properties of polysaccharides isolated from brown algae Costaria costata were analyzed. The algae were collected from May to July of the harvest period. The carbohydrates alginate and fucoidan were extracted with selected solvents. 1H Nuclear magnetic resonance and rheology were used to investigate the monomer composition and rheological characteristics of alginate. Gas chromatography and Fourier transform-infrared spectroscopy were employed to investigate the compositional properties of purified fucoidan. The results indicated that the composition and properties of alginate and fucoidan varied during the life of this alga. The alginate from the alga harvested in May and June had a higher molecular weight, viscosity, and proportion of mannuronic acid, whereas that harvested in July had a lower molecular weight and viscosity but a higher proportion of guluroinc acid. The alginate from C. costata had a higher molecular weight and a different mannuronic acid： guluronic acid ratios compared with other algae; thus, it could be used in the chemical, food, cosmetics, and pharmaceutical industries. fucoidan content reached the maximum in June. Substantial changes in the molecular weight distribution, monosaccharide composition, and sulfate content occurred simultaneously. The fraction of fucose in the polysaccharides decreased significantly from June to July, whereas that of mannose increased. This alga can be harvested during different growth periods to obtain fucoidans and alginates with different compositions and, therefore, with different biological properties.

Carbohydrases secreted by marine Vibrio sp. YKW-34 with strong Laminaria cell wall degrading ability were screened, and among them alginate lyase was found to be dominant. The effects of medium composition and culturing condition on the produc- tion of alginate lyase by marine Vibrio sp. YKW-34 in flask were investigated in this study. In the culture medium of marine broth, no alginate lyase was produced. The activity of the alginate lyase, after being induced, reached 5 UmL–1. The best inoculum volume and inoculum age were 10% and 12 h, respectively. The optimal temperature for alginate lyase production was 25℃. The fermentation medium was composed of 0.5% of Laminaria powder and 0.2% of KNO3 with an initial acidity of pH 8.0. Alginate could induce alginate lyase production but not as efficiently as Laminaria powder did. The addition of fucoidan, cellulose and glucose had nega- tive effect on the alginate lyase production. Other kinds of nitrogen sources, such as yeast extract, beef extract and peptone, had posi- tive effect on the growth of the microorganism and negative effect on alginate lyase production. In addition, the time course of algi- nate lyase production under the optimized condition was described. The optimal harvest time was 48 h.

Over the past century or more,traditional uses of seaweeds began and initially comprised the simple collection,sorting and drying of seaweeds harvested from the wild,or cast upon the beach,for use as soil“manure/fertilizer/improver”.Some macroalgae were used for unprocessed food purposes and subsequently harvested and cultivated biomasses of selected brown and red seaweeds becameindustrially processed to yield alginates,mannitol,iodine,agar,agarose,carrageenan,and more recently biostimulants,*** from various seaweeds are very diverse and are commonly applied in a range that includes textiles and dying,food/feed and supplements and specialized medical applications.

Polypehnol is an important,potentially bioactive component of Sargassum *** this study,ultrasonic assisted extraction of polyphenol-rich substances was performed using a 38%ethanol solution at a solid:liquid ratio of 1:30 at 68℃ for 32min,determined by single-factor and response surface methodology(RSM)*** content of polyphenol was 5.66mg/g in the crude *** extraction showed that the polyphenol mainly distributed in ethyl acetate(SKEE)and water phases(SKEW).The anti-oxidation test by electron spin resonance(ESR)spectrum showed that the SKEE had the strongest scavenging activity on DPPH(1,1-diphenyl-2-picrylhydrazyl)and alkyl *** was shown noncytotoxic but could inhibit the generation of cellular ROS,showing protective effects in H2O2 and AAPHinduced Vero cells and UV-B irradiated HaCaT *** also signifi cantly inhibited the release of NO of LPS-induced RAW 264.7 ***,the polyphenol-rich extracts in ethanol and ethyl acetate showed excellent anti-oxidant and anti-infl ammatory activities,which is beneficial to the development of high-value bio-substances.

The Chinese Space Station Telescope(CSST)is a cutting-edge two-meter astronomical space telescope currently under *** primary Survey Camera(SC)is designed to conduct large-area imaging sky surveys using a sophisticated seven-band photometric *** resulting data will provide unprecedented data for studying the structure and stellar populations of the Milky *** support the CSST development and scientific projects related to its survey data,we generate the first comprehensive Milky Way stellar mock catalogue for the CSST SC photometric system using the TRILEGAL stellar population synthesis *** catalogue includes approximately 12.6 billion stars,covering a wide range of stellar parameters,photometry,astrometry,and kinematics,with magnitude reaching down to g=27.5 mag in the AB magnitude *** catalogue represents our benchmark understanding of the stellar populations in the Milky Way,enabling a direct comparison with the future CSST survey ***,it sheds light on faint stars hidden from current sky *** crowding limit analysis based on this catalogue provides compelling evidence for the extension of the CSST Optical Survey(OS)to cover low Galactic latitude *** strategic extension of the CSST-OS coverage,combined with this comprehensive mock catalogue,will enable transformative science with the CSST.

The use of synthetic anabolic steroid methyltestosterone (MT) as growth promoter is prohibited in China. Vali-dations of analytical methods for MT residue in food and the results obtained have become indispensable. The high perfor-mance liquid chromatography (HPLC) method for the detection of MT with liquid-liquid extraction by trichloromethane-methanol in carp muscle tissue was preliminarily validated with reference to the following parameters: recovery (accuracy)at the 1, 5 and 10mg kg-1 level, between-run and within-run CV values (repeatability, also called relative standard deviation(RSD)) and limit of detection. The recoveries were above 80% and the between-run and within-run CV values below 10%for muscle tissue. The limit of detection was 0.05 mg kg-1.

Rheological Properties of Five Plant Gums

The rheological properties of five gum solutions (Gum Shiraz, Karaya Gum, Ghatti Gum, Arabic Gum and Gum Tragacanth) have been examined. The five mucilage solutions exhibited non-Newtonian, shear-thinning with thixotropy at higher concentrations (0.5% - 1%, m/v). According to the change of viscosity with increasing temperature, all five gums could be defined as three types: gum tragacanth and gum shiraz have a good temperature stability;karaya gum is affected by temperature obviously;the remaining two gums have a general stability. The viscosity of mucilage solutions (Arabic Gum, Ghatti Gum and Karaya Gum) had an apparent dependence on temperature. The parameter n of Shiraz Gum is the minimum, which means it has the best flow properties. The flow activation energy of karaya gum is 2.683 kcal which is the highest than other gums so that it has a great influence on temperature. The gum solutions’ viscosity had different sensitive degree over tested pH range (pH 2 - 10). Shiraz gum and arabic gum possessed better acid-proof and alkali-proof respectively. The addition of salts (NaCl and CaCl2) led to the reduction of viscosity, which was more sensitive to Ca2+ than to Na+. Both storage modulus G’ and loss modulus G” of all five gums had a dependence on temperature and frequency. These results are potentially useful for the application in the field of food processing.
