Osteosarcoma(OS)is one of the most common malignant tumors in children and young adults.As chemotherapy and other therapies are limited by low therapeutic efficiency,severe side effects and single therapeutic function,it is of high value to develop innovative therapies for precise and efficient treatment of OS.Herein,natural photo-synthetic microalgae(C.vulgaris,CV)were utilized as carriers for the chemotherapeutic agent doxorubicin(DOX)to create a multifunctional therapeutic platform(CV@DOX)for the photo-modulation of the tumor microenvi-ronment(TME)and synergistic chemo-photodynamic therapy of osteosarcoma.CV@DOX exhibited rapid drug release behavior in the acidic TME,improving the efficiency of chemotherapy against tumors and reducing side effects on other normal tissues.Under 650 nm laser irradiation,CV@DOX demonstrated the ability to effectively generate oxygen to alleviate tumor hypoxia and utilize the photosensitizing properties of chlorophyll in CV to produce an increased amount of reactive oxygen species(ROS),thereby enhancing photodynamic therapy(PDT).CV@DOX-mediated synergistic chemo-photodynamic therapy demonstrated efficacy in halting tumor progression in an orthotopic osteosarcoma mouse model by promoting tumor cell apoptosis,inhibiting tumor proliferation and angiogenesis.Moreover,chlorophyll-assisted fluorescence imaging enabled monitoring of the distribution of CV@DOX in osteosarcoma after administration.Finally,CV@DOX did not cause significant hematological and tissue toxicity,and prevented DOX-induced cardiotoxicity,showing good in vivo biocompatibility.Overall,this work presents a novel TME-responsive and TME-modulating platform for imaging-guided multimodal osteosar-coma treatment.

Central nervous injury risk factors after endovascular repair of a thoracic aortic aneurysm with type B aortic dissection

Aortic dissection is the deadliest disease of the cardiovascular system.Type B aortic dissection accounts for 30%-60%of aortic dissections and is mainly treated by endovascular repair of thoracic endovascular aneurysm repair(TEVAR).However,patients are prone to various complications after surgery,with central nervous system injury being the most common,which seriously affects their prognosis and increases the risk of disability and death.Therefore,exploring the risk factors of central nervous system injury after TEVAR can provide a basis for its prevention and control.AIM To investigate the risk factors for central nervous system injury after the repair of a thoracic endovascular aneurysm with type B aortic dissection.METHODS We enrolled 306 patients with type B aortic dissection who underwent TEVAR at our hospital between December 2019 and October 2022.The patients were categorized into injury(n=159)and non-injury(n=147)groups based on central nervous system injury following surgery.The risk factors for central nervous system injury after TEVAR for type B aortic dissection were screened by comparing the two groups.Multivariate logistic regression analysis was performed.RESULTS The Association between age,history of hypertension,blood pH value,surgery,mechanical ventilation,intensive care unit stay,postoperative recovery times on the first day after surgery,and arterial partial pressure of oxygen on the first day after surgery differed substantially(P<0.05).Multivariate logistic regression analysis indicated that age,surgery time,history of hypertension,duration of mechanical ventilation,and intensive care unit stay were independent risk factors for central nervous system injury after TEVAR of type B aortic dissection(P<0.05).CONCLUSION For high-risk patients with central nervous system injury after TEVAR of type B aortic dissection,early intervention measures should be implemented to lower the risk of neurological discomfort following surgery in high-risk patients with central nervous system injury after TEVAR f

Hydrochemistry predominantly shapes the unique nitrogen-fixing bacterial communities in Tibetan hot springs

The Tibetan Plateau has a large number of hot springs with varying temperatures and hydrochemistry， high elevation， and limited nitrogenous nutrition. Nitrogen-fixing bacteria (NFB) can fix N2to form ammonia and thus provide bioavailable nitrogen. However， there is limited knowledge about the distribution of NFB and its influencing factors in Tibetan hot springs. Here， we measured hydrochemical variables of the hot springs with a wide temperature range (32–77 ℃) in the Qucai and Daggyai geothermal zones on the Tibetan Plateau and investigated the composition of NFB using high-throughput sequencing of 16S rRNA and nifH genes. The Cl-/SO42-ratio in Qucai hot springs was higher than that in Daggyai， indicating that Qucai and Daggyai hot springs were more affected by the supply of liquid and gaseous phases， respectively. The NFB communities consisted predominantly of Nirtospirae， Chloroflexi， Deltaproteobacteria and an unidentified clade， with the last three acting as the main NFB with over 42% of the communities (the proportions are significantly larger than those found in hot springs of other geothermal regions). This demonstrates the uniqueness of NFB communities in Tibetan hot springs. NFB richness was limited by temperature in the studied Tibetan hot springs and was significantly lower than in low-elevation geothermal regions. The NFB community was predominantly affected by hydrochemistry， in contrast to the entire prokaryotic community， which was primarily influenced by temperature. This study expands our current understanding of NFB distribution and diversity as well as biogeochemical process in geothermal spring environments.

In the past few years,many groups have focused on the research and development of GaN-based ultraviolet laser diodes(UV LDs).Great progresses have been achieved even though many challenges exist.In this article,we analyze the challenges of developing GaN-based ultraviolet laser diodes,and the approaches to improve the performance of ultraviolet laser diode are reviewed.With these techniques,room temperature(RT)pulsed oscillation of AlGaN UVA(ultraviolet A)LD has been realized,with a lasing wavelength of 357.9 nm.Combining with the suppression of thermal effect,the high output power of 3.8 W UV LD with a lasing wavelength of 386.5 nm was also fabricated.

Multimorbidity and mortality among older patients with coronary heart disease in Shenzhen,China

BACKGROUND The current understanding of the magnitude and consequences of multimorbidity in Chinese older adults with coronary heart disease(CHD)is insufficient.We aimed to assess the association and population-attributable fractions(PAFs)between multimorbidity and mortality among hospitalized older patients who were diagnosed with CHD in Shenzhen,China.METHODS We conducted a retrospective cohort study of older Chinese patients(aged≥65 years)who were diagnosed with CHD.Cox proportional hazards models were used to estimate the associations between multimorbidity and all-cause and cardiovascular disease(CVD)mortality.We also calculated the PAFs.RESULTS The study comprised 76,455 older hospitalized patients who were diagnosed with CHD between January 1,2016,and August 31,2022.Among them,70,217(91.9%)had multimorbidity,defined as the presence of at least one of the predefined 14 chronic conditions.Those with cancer,hemorrhagic stroke and chronic liver disease had the worst overall death risk,with adjusted HRs(95%CIs)of 4.05(3.77,4.38),2.22(1.94,2.53),and 1.85(1.63,2.11),respectively.For CVD mortality,the highest risk was observed for hemorrhagic stroke,ischemic stroke,and chronic kidney disease;the corresponding adjusted HRs(95%CIs)were 3.24(2.77,3.79),1.91(1.79,2.04),and 1.81(1.64,1.99),respectively.All-cause mortality was mostly attributable to cancer,heart failure and ischemic stroke,with PAFs of 11.8,10.2,and 9.1,respectively.As for CVD mortality,the leading PAFs were heart failure,ischemic stroke and diabetes;the corresponding PAFs were 18.0,15.7,and 6.1,respectively.CONCLUSIONS Multimorbidity was common and had a significant impact on mortality among older patients with CHD in Shenzhen,China.Cancer,heart failure,ischemic stroke and diabetes are the primary contributors to PAFs.Therefore,prioritizing improved treatment and management of these comorbidities is essential for the survival prognosis of CHD patients from a holistic public health perspective.

The Spatiotemporal Distribution Characteristics of Cloud Types and Phases in the Arctic Based on CloudSat and CALIPSO Cloud Classification Products

The cloud type product 2B-CLDCLASS-LIDAR based on CloudSat and CALIPSO from June 2006 to May 2017 is used to examine the temporal and spatial distribution characteristics and interannual variability of eight cloud types(high cloud, altostratus, altocumulus, stratus, stratocumulus, cumulus, nimbostratus, and deep convection) and three phases(ice,mixed, and water) in the Arctic. Possible reasons for the observed interannual variability are also discussed. The main conclusions are as follows:(1) More water clouds occur on the Atlantic side, and more ice clouds occur over continents.(2)The average spatial and seasonal distributions of cloud types show three patterns: high clouds and most cumuliform clouds are concentrated in low-latitude locations and peak in summer;altostratus and nimbostratus are concentrated over and around continents and are less abundant in summer;stratocumulus and stratus are concentrated near the inner Arctic and peak during spring and autumn.(3) Regional averaged interannual frequencies of ice clouds and altostratus clouds significantly decrease, while those of water clouds, altocumulus, and cumulus clouds increase significantly.(4) Significant features of the linear trends of cloud frequencies are mainly located over ocean areas.(5) The monthly water cloud frequency anomalies are positively correlated with air temperature in most of the troposphere, while those for ice clouds are negatively correlated.(6) The decrease in altostratus clouds is associated with the weakening of the Arctic front due to Arctic warming, while increased water vapor transport into the Arctic and higher atmospheric instability lead to more cumulus and altocumulus clouds.

Vaccines have been the primary remedy in the global fight against coronavirus disease 2019（COVID-19）.The receptor-binding domain（RBD） of the spike protein, a critical viral immunogen, is affected by the heterogeneity of its glycan structures and relatively low immunogenicity. Here, we describe a scalable synthetic platform that enables the precise synthesis of homogeneously glycosylated RBD, facilitating the elucidation of carbohydrate structure–function relationships. Five homogeneously glycosylated RBDs bearing biantennary glycans were prepared, three of which were conjugated to T-helper epitope(Tpep) from tetanus toxoid to improve their weak immune response. Relative to natural HEK293-derived RBD, synthetic RBDs with biantennary N-glycan elicited a higher level of neutralising antibodies against SARS-CoV-2 in mice. Furthermore, RBDs containing Tpepelicited significant immune responses in transgenic mice expressing human angiotensin-converting enzyme 2. Our collective data suggest that trimming the N-glycans and Tpepconjugation of RBD could potentially serve as an effective strategy for developing subunit vaccines providing efficient protection.

Cucurbit[6]uril functionalized gold nanoparticles and electrode for the detection of metformin drug

Based on the host-guest molecular recognition capability of cucurbit[6]uril(CB[6])modified on the gold surface,sensitive spectrophotometric and electrochemical methods for the detection of metformin(MET)have been developed.The molecular recognition between cucurbit[7]uril(CB[7])or CB[6]and MET is initially demonstrated and the related recognition mechanism is further deliberated.First,CB[6]-modified gold nanoparticles(AuNPs/CB[6])were synthesized and then characterized by ultraviolet visible light spectrum(UV–vis)and transmission electron microscopy(TEM).The aggregation of Au NPs/CB[6]prompted by MET triggered changes of color and the absorption spectrum,that explored for the visual identification and spectrophotometric determination of MET.Under the optimized detection conditions,the UV–vis spectrometry had a good linear relationship in the range of 6–700μmol/L,and the detection limit was 2μmol/L.In addition,a single-layer CB[6]-modified gold electrode(GE-CB[6])detection system for MET was constructed.As the concentration of MET in the solution continues to increase,the charge transfer resistance(Rct)in the Nyquist diagram of the electrochemical impedance method(EIS)continues to increase.In the concentration range from 10 pmol/L to 20 nmol/L,the logarithm of the MET concentration has a good linear relationship with Rct,and the detection limit of this method is 1.35 pmol/L.Both methods have good concentration sensitivity to MET in different concentration ranges,providing a powerful tool for the detection of MET.

Dark Matter Search in Dwarf Irregular Galaxies with Ten Years of Data from the Ice Cube Neutrino Observatory

Dwarf irregular galaxies(dIrrs),as rotationally supported systems,have more reliable J-factor measurements than dwarf spheroidal galaxies and have received attention as targets for dark matter detection in recent years.In this paper,we use 10 yr of IceCube muon-track data and an unbinned maximum-likelihood-ratio method to search for neutrino signals beyond the background from the directions of seven dIrrs,aiming to detect neutrinos produced by heavy annihilation dark matter.We do not detect any significant signal.Based on such null results,we calculate the upper limits on the velocity-averaged annihilation cross section for 1 TeV–10 PeV dark matter.Our limits,although weaker than the strictest constraints in the literature in this mass range,are also a good complement to the existing results considering the more reliable J-factor measurements of dIrrs.

Mast cell degranulation-triggered by SARS-CoV-2 induces tracheal-bronchial epithelial inflammation and injury

SARS-CoV-2 infection-induced hyper-inflammation is a key pathogenic factor of COVID-19.Our research,along with others',has demonstrated that mast cells(MCs)play a vital role in the initiation of hyper-inflammation caused by SARS-CoV-2.In previous study,we observed that SARS-CoV-2 infection induced the accumulation of MCs in the peri-bronchus and bronchioalveolar-duct junction in humanized mice.Additionally,we found that MC degranulation triggered by the spike protein resulted in inflammation in alveolar epithelial cells and capillary endothelial cells,leading to subsequent lung injury.The trachea and bronchus are the routes for SARS-CoV-2 transmission after virus inhalation,and inflammation in these regions could promote viral spread.MCs are widely distributed throughout the respiratory tract.Thus,in this study,we investigated the role of MCs and their degranulation in the development of inflammation in tracheal-bronchial epithelium.Histological analyses showed the accumulation and degranulation of MCs in the peri-trachea of humanized mice infected with SARS-CoV-2.MC degranulation caused lesions in trachea,and the formation of papillary hyperplasia was observed.Through transcriptome analysis in bronchial epithelial cells,we found that MC degranulation significantly altered multiple cellular signaling,particularly,leading to upregulated immune responses and inflammation.The administration of ebastine or loratadine effectively suppressed the induction of inflammatory factors in bronchial epithelial cells and alleviated tracheal injury in mice.Taken together,our findings confirm the essential role of MC degranulation in SARS-CoV-2-induced hyper-inflammation and the subsequent tissue lesions.Furthermore,our results support the use of ebastine or loratadine to inhibit SARS-CoV-2-triggered degranulation,thereby preventing tissue damage caused by hyper-inflammation.
