Parkinson’s disease(PD)is a common neurodegenerative disorder with no cure.Astragalus membranaceus is used in Chinese culture as a food supplement to boost immunity.The present study aimed to explore the neuroprotective effects of total flavonoids extracted from A.membranaceus(TFA)and their protective mechanisms.TFA offered neuroprotection against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)in the mouse model of Parkinsonism,by improving behavior performance in the gait analysis and pole test,and inhibiting the decline of tyrosine hydroxylase(TH)positive neurons and TH protein expression in substantia nigra of mice.TFA also prevented 1-methyl-4-phenylpyridinium(MPP+)induced neurotoxicity in SHSY5Y cells,by increasing GSH and GSH/GSSG ratio,and reducing reactive oxygen species.In addition,the neuroprotective effects of TFA were associated with its ability to restore MPTP/MPP+induced downregulation of SLC7A11 and glutathione peroxidase 4(GPX-4).In conclusion,we demonstrated that TFA exerted significant neuroprotection against MPTP/MPP+induced neurodegeneration by inhibiting ferroptosis through the regulation of SLC7A11/GPX-4 axis,suggesting the use of TFA as a possible food supplement in the prevention of PD.

Organic carbon isotope and pollen evidence for mangrove development and response to human activity in Guangxi(Southwest China) over the last 140 years

Mangrove degradation must reduce carbon sequestration in recent years, thereby aggravating global warming.Thus, short-term impacts of human activity on mangrove ecosystems are cause for concern from local governments and scientists. Mangroves sediments can provide detailed records of mangrove species variation in the last one hundred years, based on detailed 210 Pb data. The study traced the history of mangrove development and its response to environmental change over the last 140 years in two mangrove swamps of Guangxi, Southwest China. Average sedimentation rates were calculated to be 0.48 cm/a and 0.56 cm/a in the Yingluo Bay and the Maowei Sea, respectively. Chemical indicators（δ13Corg and C：N） were utilized to trace the contribution of mangrove-derived organic matter（MOM） using a ternary mixing model. Simultaneous use of mangrove pollen can help to supplement some of these limitations in diagenetic/overlap of isotopic signatures. We found that vertical distribution of MOM was consistent with mangrove pollen, which could provide similar information for tracing mangrove ecosystems. Therefore, mangrove development was reconstructed and divided into three stages： flourishing, degradation and re-flourishing/re-degradation period. The significant degradation, found in the period of 1968–1998 and 1907–2007 in the Yingluo Bay and the Maowei Sea, respectively, corresponding to a rapid increase of reclamation area and seawall length, rather than climate change as recorded in the region.

Sedimentary Records of Mangrove Evolution During the Past one Hundred Years Based on Stable Carbon Isotope and Pollen Evidences in Maowei, SW China

Mangroves accumulate sedimentary sequences, where cores can provide historical records of mangrove evolution with past climate change and human activity. The study traced the history of mangrove evolution during the past one hundred years in a mangrove swamp of Maowei Sea, SW China. The sedimentation rates(0.38-0.95 cm yr^(-1)) were calculated on the basis of ln(^(210)Pb_(xs)/Al) and mass depth in the core sediments. Chemical tracers, such as δ^(13)C_(org) and C:N values, were utilized to trace the contribution of mangrove-derived organic matter using a ternary mixing model. Because of potential diagenetic alteration and / or overlap in the isotopic signatures of different components, simultaneous use of mangrove pollen diagrams can help to supplement some of these limitations. Combined with mangrove pollen, mangrove evolution was reconstructed and could be divided into three stages: flourishment(1886-1905 AD), slight degradation(1905-1949 AD) and rapid degradation period(1949-2007 AD), which was consistent with previous reports. The reclamation of mangrove swamps to shrimp ponds was the major reason for rapid degradation of mangrove ecosystems in recent years, rather than climate change in the region.

^(210)Pb Chronology and Trace Metal Geochemistry in the Intertidal Sediment of Qinjiang River Estuary, China

Historical records of metal inputs were studied by using a sediment core collected from a sand-rich mudflat in the Qinjiang River estuary, China. 210Pb chronology was used to reconstruct the fluxes of Hg, Cu, Pb, Zn, Cd, Cr and As to the core site during the last 86 years. Based on the constant initial concentration model, the sedimentation rates are 1.18 cm year-1 in the top 30 cm sandy layer and 0.92 cm year-1 in the muddy bottom layer. To compensate for grain-size and mineralogy effects on metal concentra-tions, aluminum was used as the normalizing element. The enrichment factors (EF) indicate that the natural inputs had prevailed up to the early 1980s. After this period, the intensity of human activities has resulted in continual increasing trend of metals towards the surface. Recent sediment samples from the Qinjiang River estuary are found moderately enriched by Cd (EF>1.5) and slightly enriched by other metals (EF<1.5). Considering that the drainage area of the Qinjiang River is mostly agricultural land, the increased Cd may be due to the usage of fertilizers and pesticides in agricultural activities and the combustion of fossil fuels.

Sediment transport in the Luanhe River delta:grain size trend analysis

Sediment grain size in the deltaic environment of the Luanhe River(LR),Liaoning,China,contains sediment transport pathway information useful in elucidating the shoreline change and fluvialmarine interaction.In this study,we utilized numerical partitioning of the sedimentary components and geostatistical grain size trend analysis(GSTA)to define the sediment transport pattern in the Luanhe River delta(LRD)and interpolated the sediment transport pattern using content changes of end numbers(EM).EM1(the mean grain size 7.12Ф,fine silt),EM2(2.37Ф,fine sand),and EM3(1.27Ф,medium sand)components were identified by the numerical partitioning by GSTA.Kriging interpolation method was used to interpolate the parameters of the grain size for the regular grid,and the interpolation radius was 0.015 decimal degree.We chose 0.09 decimal degree as the characteristic distance for GSTA in the semivariogram model using the geostatistical method.The FB(-)case(finer,better sorted and more negatively skewed)was adopted in GSTA for its satisfaction in the Global Moran’s I test.The result of the GSTA shows that the sediments in the south barriers(SBs)were transported to the southwest of the study area.The sediments in the north,in the SE direction of sediment transport trend from the river mouth,indicated that the sediments in the north of the study area were transported from the LR to the northern beaches,and to the south and east of the study area.The sediment transport trend that simplified by GSTA as the FB(-)case was approved by the content changes of sedimentary components(i.e.EM1,EM2,and EM3).In addition,the turbulent jet diffusion pattern indicated that the coarse sediments(EM3)were delivered by LR during the flood season,and the EM2 and EM1 were from wave and tide,respectively.

通过巢式简并PCR和基因组步行技术从香菇(Lentinula edodes)单核体中克隆到乳清酸核苷-5′-单磷酸脱羧酶(orotidine-5'-monophosphate decarboxylase,OMPDC)的编码基因le-pyrG,长度为946 bp.经生物信息学分析,香菇le-pyrG基因含有2个长度分别为67 bp和51 bp的内含子,该基因可以编码1个含有275个氨基酸残基的蛋白质序列,该序列经比对分析显示与裂褶菌(Schizophyllum commne)和灰盖鬼伞(Coprinus cinereus) OMPDC序列的相同性分别为73%和70%,相似性分别为84% 和83%.这是首次报道从栽培食用真菌中克隆出乳清酸核苷-5′-单磷酸脱羧酶编码基因的全序列.

Polysulfide regulation by defect-modulated Ta_(3)N_(5-x) electrocatalyst toward superior room-temperature sodium-sulfur batteries

Resolving low sulfur reaction activity and severe polysulfide dissolution remains challenging in metalsulfur batteries.Motivated by a theoretical prediction,herein,we strategically propose nitrogenvacancy tantalum nitride(Ta3N5-x)impregnated inside the interconnected nanopores of nitrogendecorated carbon matrix as a new electrocatalyst for regulating sulfur redox reactions in roomtemperature sodium-sulfur batteries.Through a pore-constriction mechanism,the nitrogen vacancies are controllably constructed during the nucleation of Ta3N5-x.The defect manipulation on the local environment enables well-regulated Ta 5d-orbital energy level,not only modulating band structure toward enhanced intrinsic conductivity of Ta-based materials,but also promoting polysulfide stabilization and achieving bifunctional catalytic capability toward completely reversible polysulfide conversion.Moreover,the interconnected continuous Ta3N5-x-in-pore structure facilitates electron and sodium-ion transport and accommodates volume expansion of sulfur species while suppressing their shuttle behavior.Due to these attributes,the as-developed Ta3N5-x-based electrode achieves superior rate capability of 730 mAh g-1 at 3.35 A g-1,long-term cycling stability over 2000 cycles,and high areal capacity over 6 mAh cm-2 under high sulfur loading of 6.2 mg cm-2.This work not only presents a new sulfur electrocatalyst candidate for metal-sulfur batteries,but also sheds light on the controllable material design of defect structure in hopes of inspiring new ideas and directions for future research.

Sand barrier morphological evolution based on time series remote sensing images:a case study of Anhaiao,Pingtan

The morphological evolution of the sand barrier in the Anhaiao coastal zone of Pingtan from 1996 to 2018 was studied.Tidal correction was used to refine the location of the coastline.A standard deviation ellipse method was applied to further analyze the movement of the barrier head with the axis and rotation angle.A natural neighbor interpolation(NNI)method was carried out to calculate the terrain of the intertidal area,and the erosion and deposition characteristics were illustrated based on the terrain.The results showed that the northern part of the sand barrier facing the lagoon area was deposited over the whole studied period,while erosion has always occurred in the southern part of the sand barrier facing the open sea.The erosion and deposition were slightly different on both sides of the barrier head due to hydrodynamic turbulence.The middle sand barrier moved 102.60 m away from its original location in 1996,and the end of the barrier moved 65.45 m.The head of the sand barrier continued moving 379 m to the northwest.Consequently,the preliminary morphological evolution of the sand barrier corresponding to the distance and direction of movement was detected.

The evolving views of hematopoiesis:from embryo to adulthood and from in vivo to in vitro

The hematopoietic system composed of hematopoietic stem and progenitor cells(HSPCs)and their differentiated lineages serves as an ideal model to uncover generic principles of cell fate transitions.From gastrulation onwards,there successively emerge primitive hematopoiesis(that produces specialized he-matopoietic cells),pro-definitive hematopoiesis(that produces lineage-restricted progenitor cells),and definitive hematopoiesis(that produces multipotent HSPCs).These nascent lineages develop in several transient hematopoietic sites and finally colonize into lifelong hematopoietic sites.The development and maintenance of hematopoietic lineages are orchestrated by cell-intrinsic gene regulatory networks and cell-extrinsic microenvironmental cues.Owing to the progressive methodology(e.g.,high-throughput lineage tracing and single-cell functional and omics analyses),our understanding of the developmental origin of hematopoietic lineages and functional properties of certain hematopoietic organs has been updated;meanwhile,new paradigms to characterize rare cell types,cell heterogeneity and its causes,and comprehensive regulatory landscapes have been provided.Here,we review the evolving views of HSPC biology during developmental and postnatal hematopoiesis.Moreover,we discuss recent advances in the in vitro induction and expansion of HSPCs,with a focus on the implications for clinical applications.
