Magnetic resonance imaging of knees:a novel approach to predict recombinant human growth hormone therapy response in short-stature children in late puberty

Background There is no appropriate tool to predict recombinant human growth hormone(rhGH)response before therapy initiation in short-stature children in late *** current study aimed to explore the associations between magnetic resonance imaging(MRI)stages of the knee growth plates and rhGH response in short-stature children in late *** In this prospective cohort study,short-stature children in late puberty were treated with rhGH and followed up for 6 *** proposed a novel knee MRI staging system according to the growth plate states of distal femurs or proximal tibias and divided the participants into three groups:unclosed growth plate group,marginally closed growth plate group,and nearly closed growth plate *** primary outcomes were height gain and growth velocity(GV),which were assessed three months *** Fifty participants were enrolled,including 23 boys and 27 *** and height gain after 6 months of rhGH therapy decreased successively in the three groups with an increased degree of growth plate fusion,especially when grouped by proximal tibias(GV_(1-3 mon)from 9.38 to 6.08 to 4.56 cm/year,GV_(4-6 mon)from 6.75 to 4.92 to 3.25 cm/year,and height gain from 4.03 to 2.75 to 1.95 cm,all P<0.001).Moreover,the MRI stages of growth plates independently served as a significant variable for GV and height gain after therapy,especially when grouped by proximal tibias(all P<0.01).Conclusion The MRI staging method is expected to be an effective tool for predicting rhGH response before therapy initiation in short-stature children in late puberty.

Investigation of groundwater characteristics and its influence on Landslides in Heifangtai Plateau using comprehensive geophysical methods

The occurrence of landslides in Heifangtai plateau is primarily caused by the rise in water levels due to irrigation. To accurately understand the distribution of groundwater and its impact on the landslide hazard, a combination of Electrical Resistivity Tomography(ERT), Induced Polarization(IP) and Surface Nuclear Magnetic Resonance(SNMR) methods were used in this study. By conducting a comprehensive analysis, the characteristics of water-bearing structure in vertical and groundwater distribution in horizontal were detected;and the influence of the groundwater on plateau and landslides was also identified. The results indicate that the groundwater occurs in the loess aquifer with a three-layer structure in ***, the aquifer has a unified water table over the plateau, with a low water level in the north and high one in the south. The high resistivity bedrock uplift belt in the middle of the plateau forms a watershed,with the north side of the uplift belt being a relatively stable slope area with stable water content and fewer geological disasters. In contrast, the south side of the uplift belt is a disaster-prone region with vertical fissures well developed in the loess aquifers. The southern landslides are characterized by the interphase distribution of high and low electrical resistivity. The infiltration and discharge of groundwater result in the formation of a collapse belt in the low resistivity water-bearing structure of landslide, which causes the entire block with high resistivity and stable bedrock to slide. There was a newly formed landslide in a larger range at the landslide' s trailing edge. This study provides a scientific basis for the study of landslides mechanisms and disaster prevention by identifying the distribution characteristics of groundwater and analyzing its influence from a geophysical perspective in Heifangtai.

Public Auditing for Encrypted Data with Client-Side Deduplication in Cloud Storage

Storage auditing and client-side deduplication techniques have been proposed to assure data integrity and improve storage efficiency, respectively. Recently, a few schemes start to consider these two different aspects together. However, these schemes either only support plaintext data file or have been proved insecure. In this paper, we propose a public auditing scheme for cloud storage systems, in which deduplication of encrypted data and data integrity checking can be achieved within the same framework. The cloud server can correctly check the ownership for new owners and the auditor can correctly check the integrity of deduplicated data. Our scheme supports deduplication of encrypted data by using the method of proxy re-encryption and also achieves deduplication of data tags by aggregating the tags from different owners. The analysis and experiment results show that our scheme is provably secure and efficient.

Context-Aware Adaptive Routing for Opportunistic Network

Opportunistic networking-forwarding messages in a disconnected mobile ad hoc network via any encountered nodes offers a new mechanism for exploiting the mobile devices that many users already carry. However, forwarding messages in such a network is trapped by many particular challenges, and some protocols have contributed to solve them partly. In this paper, we propose a Context-Aware Adaptive opportunistic Routing algorithm（CAAR）. The algorithm firstly predicts the approximate location and orientation of the destination node by using its movement key positions and historical communication records, and then calculates the best neighbor for the next hop by using location and velocity of neighbors. In the unpredictable cases, forwarding messages will be delivered to the more capable forwarding nodes or wait for another transmission while the capable node does not exist in the neighborhood. The proposed algorithm takes the movement pattern into consideration and can adapt different network topologies and movements. The experiment results show that the proposed routing algorithm outperforms the epidemic forwarding（EF） and the prophet forwarding（PF） in packet delivery ratio while ensuring low bandwidth overhead.

Modeling Study of Fruit Morphological Formation in Melon

Modeling of fruit morphological formation in melon is important for realizing virtual and digital plant *** objective of this study was to characterize the changes in patterns of fruit growth characters during plant *** cultivar experiments,a high-resolution wireless vision sensor network has been developed to realize non-contact automatic uninterrupted measurement of the fruit shape micro-change （fruit size,color,and net）.Results showed that the fruit swelling process （vertical and horizontal diameters） exhibited a slow-rapid-slow pattern,which could be well described with a logistic curve against growing degree days （GDD）;fruit color changes based on the RGB values could be represented by quadratic relationship to cumulative GDD;the fruit net changes over growth progress could be partitioned into three phases according to the time *** first phase was from 1 to 30 days after pollination （DAP）,in which the vertical stripe appeared at fruit middle part and the horizontal stripe at fruit petiole and hilum part as well;the second phase was from 30 to 40 DAP,the horizontal stripe occurred at fruit middle part and the net was formed;the third phase was the process started from 40 DAP,the netted breadth and thickness were gradually *** model was validated with the independent data from the experiment,and the mean RMSE （root mean square error） of fruit were 0.36 and 0.28 cm for vertical and horizontal diameters,11.9 for fruit color,and 0.45 cm for stripe length and diameter at varied GDD,*** work is beneficial to a reliable foundation for study the relationship between morphological formation and physiological change of the melon fruit internally and then realize the intelligent precision management to improve the yield and quality of greenhouse melon production.

WWOX induces apoptosis and inhibits proliferation of human hepatoma cell line SMMC-7721

AIM: To investigate the effects of the WWOX gene on the human hepatic carcinoma cell line SMMC-7721. METHODS: Full-length WWOX cDNA was amplified from normal human liver tissues. Full-length cDNA was subcloned into pEGFP-N1, a eukaryotic expression vector. After introduction of the WWOX gene into cancer cells using liposomes, the WWOX protein level in the cells was detected through Western blotting. Cell growth rates were assessed by methyl thiazolyl tetrazolium (MTT) and colony formation assays. Cell cycle progression and cell apoptosis were measured by flow cytometry. The phosphorylated protein kinase B (AKT) and activated fragments of caspase-9 and caspase-3 were examined by Western blotting analysis. RESULTS: WWOX significantly inhibited cell proliferation, as evaluated by the MTT and colony formation assays. Cells transfected with WWOX showed significantly higher apoptosis ratios when compared with cells transfected with a mock plasmid, and overexpression of WWOX delayed cell cycle progression from G1 to S phase, as measured by flow cytometry. An increase in apoptosis was also indicated by a remarkable activation of caspase-9 and caspase-3 and a dephosphorylation of AKT (Thr308 and Ser473) measured with Western blotting analysis. CONCLUSION: Overexpression of WWOX induces apoptosis and inhibits proliferation of the human hepatic carcinoma cell line SMMC-7721.

Dynamics of Nitrogen Speciation in Horticultural Soils in Suburbs of Shanghai,China

Dissolved organic nitrogen （DON） represents a significant pool of soluble nitrogen （N） in soil ecosystems. Soil samples under three different horticultural management practices were collected from the Xiaxiyang Organic Vegetable and Fruit Farm, Shanghai, China, to investigate the dynamics of N speciation during 2 months of aerobic incubation, to compare the effects of different soils on the mineralization of ^14C-labeled amino acids and peptides, and to determine which of the pathways in the decomposition and subsequent ammonification and nitrification of organic N represented a significant blockage in soil N supply. The dynamics of N speciation was found to be significantly affected by mineralization and immobilization. DON, total free amino acids, and NH^＋-N were maintained at very low levels and did not accumulate, whereas NO3^--N gradually accumulated in these soils. The conversion of insoluble organic N to low-molecular-weight （LMW） DON represented a main constraint to N supply, while conversions of LMW DON to NH4^＋-N and NH4^＋-N to NO3^--N did not. Free amino acids and peptides were rapidly mineralized in the soils by the microbial community and consequently did not accumulate in soil. Turnover rates of the additional amino acids and peptides were soil-dependent and generally followed the order of organic soil 〉 transitional soil 〉 conventional soil. The turnover of high-molecular-weight DON was very slow and represented the major DON loss. Further studies are needed to investigate the pathways and bottlenecks of organic N degradation.

Effects of RNAi Silencing of SSIII Gene on Phosphorus Content and Characteristics of Starch in Potato Tubers

The sense and antisense fragments of the soluble starch synthase （SSII1） gene and the intron fragment of somatic embryogenesis receptor-like kinase （SERK1） gene were cloned from potato using PCR techniques. The RNAi plant expression vectors pBI-SSIII-RNAi and pBIC-SSIII-RNAi were constructed which containing fusion fragment of ＂sense fragment-intron-antisense fragment＂ driven by the constitutive expression promoter CaMV 35S and the tuber-specific expression promoter CIPP, respectively. The putative transgenic plants of potato cultivars Kexin-1 and Kexin-4 were obtained using Agrobacterium-mediated transformation method. PCR assay showed that the interference fragment of SSlll gene was integrated into potato genome. The RT-PCR analysis showed that the expression of SSlll gene was repressed apparently on the transcription level. Starch granules of the transgenic potato plants were different in morphology and became cracked in starch granule centre compared with the non-transgenic control plants. The amylose content of starch was increased by 2.68-29.05%, amylopectin to amylose ratio of starch had declined significantly, and the phosphorus content of the starch of the transgenic plants was reduced 9.94-58.36% compared with control plants. The results could provide certain foundation for improvement of potato starch quality.
