海南中北部地区牛体表蜱种鉴定及蜱携带无形体属病原体情况调查研究

目的:研究海南牛体表蜱虫的种类及蜱携带无形体属病原体的情况。方法:本研究选取海南中北部9个县市为样本采集点,从牛体表采集蜱虫样本。通过形态学观察和分子生物学技术进行蜱种鉴定,并以16S rRNA、groEL和gltA为靶基因,利用PCR方法检测蜱虫携带无形体属病原体情况。并进一步采用系统发育分析方法,揭示海南牛体表蜱虫携带的无形体属病原体种间关系。结果:本研究共采集335只蜱虫,包括血红扇头蜱(Rhipicephalus sanguineus)44只、微小扇头蜱(Rhipicephalus microplus)291只。PCR检测结果表明,该地区蜱中存在候选博莱无形体(Candidatus Anaplasma boleense)、边缘无形体(Anaplasma marginale),阳性率分别为0.60%(2/335),0.90%(3/335)。系统发育分析结果表明,本实验获得的无形体与已知的无形体同源性较高。结论:海南中北部地区牛体表的优势蜱种为微小扇头蜱,并且蜱虫携带无形体属病原体,本研究为该地区无形体病的预防和治疗提供了重要的数据支撑。

Fibroblast growth factor(FGF) is found throughout multicellular organisms; however, fgf homologs(vfgf) have only been identified among viruses in lepidopteran baculoviruses. The function of v FGFs from Group I alphabaculoviruses, including Autographa californica multiple nucleopolyhedrovirus(Ac MNPV) and Bombyx mori nucleopolyhedrovirus(Bm NPV), involves accelerated killing of infected larvae by both viruses. The v FGF of Group II alphabaculovirus is structurally different from that of Group I alphabaculovirus, with a larger C-terminal region and additional N-linked glycosylation sites. In this study, we characterized the Group II alphabaculovirus v FGF of Helicoverpa armigera single nucleopolyhedrovirus(Hear NPV). The transcription and expression of vfgf was detected at 3 h and 16 h post-infection in Hear NPV-infected cells. To further study v FGF function, we constructed vfgf-knockout and-repaired Hear NPV bacmids and investigated their affect in both cultured cells and insects. Deletion of vfgf had no effect on budded-virus production or viral DNA replication in cultured Hz AM1 cells. However, bioassays showed that Hear NPV vfgf deletion significantly increased the median lethal dose and delayed the median lethal time by ~12 h in the host insect when the virus was delivered orally. These results suggested that v FGF is an important virulent factor for HearN PV infection and propagation in vivo.

Developing a cotton fabric sensing layer with good waterproofness and breathability via a low-cost and eco-friendly method is increasingly important for the construction of comfortable and wearable electronic ***,a waterproof and breathable cotton fabric composite decorated by reduced graphene oxide(rGO)and carbon nanotube(CNT),Cotton/rGO/CNT,is reported by a facile solution infiltration method,and we adopt such Cotton/rGO/CNT composite to develop a layer-by-layer structured multifunctional flexible sensor,enabling the high-sensitivity detection of pressure and temperature ***,the multifunctional flexible sensor exhibits a high response toward tiny pressure,demonstrating salient superiority in the continuous and reliable monitoring of human physiological *** temperature sensing,a good linear response for the temperatures ranging from 28 to 40℃ is achieved by the multifunctional flexible sensor and gives rise to be successfully applied to the non-contact real-time monitoring of human respiration ***,an array consisting of multifunctional flexible sensors further demonstrates its feasibility in perceiving and mapping the pressure and temperature information of contact *** work provides a feasible strategy for designing cotton-based sensing layers that can effectively resist liquid water penetration and allow water vapor transmission,and offers reasonable insight for constructing comfort and multifunctional wearable electronics.

The multimode fiber(MMF)has great potential to transmit high-resolution images with less invasive methods in endoscopy due to its large number of spatial modes and small core ***,spatial modes crosstalk will inevitably occur in MMFs,which makes the received images become speckles.A conditional generative adversarial network(GAN)composed of a generator and a discriminator was utilized to reconstruct the received *** conduct an MMF imaging experimental system of transmitting over 1 m MMF with a 50μm *** with the conventional method of U-net,this conditional GAN could reconstruct images with fewer training datasets to achieve the same performance and shows higher feature extraction capability.

Inflammatory bowel disease(IBD)is a serious disorder,and exploration of active compounds to treat it is *** acidic polysaccharide named SUSP-4 was purified from Selaginella uncinata(Desv.)Spring,which contained galacturonic acid,galactose,xylose,arabinose,and rhamnose with the main chain structure of→4)-α-d-GalAp-(1→and→6)-β-d-Galp-(1→and the branched structure of→5)-α-l-Araf-(1→.Animal experiments showed that compared with Model group,SUSP-4 significantly improved body weight status,disease activity index(DAI),colonic shortening,and histopathological damage,and elevated occludin and zonula occludens protein 1(ZO-1)expression in mice induced by dextran sulfate sodium salt(DSS).16S ribosomal RNA(rRNA)sequencing indicated that SUSP-4 markedly downregulated the level of Akkermansia and *** results confirmed that SUSP-4 obviously elevated thiamine levels compared with Model mice by adjusting thiamine metabolism,which was further confirmed by a targeted metabolism *** transplantation experiments showed that SUSP-4 exerted an anti-IBD effect by altering the intestinal flora in mice.A mechanistic study showed that SUSP-4 markedly inhibited macrophage activation by decreasing the levels of phospho-nuclear factor kappa-B(p-NF-κB)and cyclooxygenase-2(COX-2)and elevating NF-E2-related factor 2(Nrf2)levels compared with Model *** conclusion,SUSP-4 affected thiamine metabolism by regulating Akkermania and inhibited macrophage activation to adjust NF-κB/Nrf2/COX-2-mediated inflammation and oxidative stress against *** is the first time that plant polysaccharides have been shown to affect thiamine metabolism against IBD,showing great potential for in-depth research and development applications.
