Microarray analysis of New Green Cocoon associated genes in silkworm, Bombyx mori

Green cocoons in silkworm, Bombyx mori, are caused by flavonoids accumulation in the silk proteins, fibroin and sericin. Despite the economic value of natural green cocoon and medical value of flavonoids, there is limited understanding of the molecular mechanism regulating flavonoids uptake in silkworm, which is tightly associated with the trait of green cocoon. The purpose of this study is to perform a comprehensive analysis to understand the molecular mechanisms of flavonoids uptake in silkworm based on microarray analyses. The study subject was the New Green Cocoon from the silkworm strains, G200 and N100, a new spontaneous dominant green cocoon trait identified in the 2000s. The genes regulating this trait are independent of other green cocoon genes previously reported. Genome-wide gene expression was compared between the New Green Cocoon producing silkworm strains, G200 and NI00, and the control sample, which is the white cocoon producing strain 872B. Among these strains, N100 and 872B are near-isogenic lines. The results showed that 130 genes have consistently changing expression patterns in the green cocoon strains when compared with the white cocoon strain. Among these, we focused on the genes related to flavonoids metabolism and absorption, such as sugar trans- porter genes and UDP-glucosyltransferase genes. Based on our findings, we propose the potential mechanisms for flavonoids absorption and metabolism in silkworm. Our results imply that silkworm might be used as an underlying model for flavonoids in pharmaceutical research.

Comparison between Bilateral and Unilateral Sudden Sensorineural Hearing Loss

Background：Bilateral sudden sensorineural hearing loss （BSSHL） is rare and assumed to be a different clinical entity compared to unilateral SSHL （USSHL）.This study examined the differences between the idiopathic BSSHL and ***：Forty-six sequential BSSHL patients （Se-BSSHL） and 68 simultaneous BSSHL （Si-BSSHL） were consecutively admitted between June 2008 and December *** sets of patients served as control groups：（1） USSHL patients with healthy contralateral ear and （2） USSHL patients with contralateral preexisting hearing loss （USSHLwCHL）.We retrospectively analyzed differences among four cohorts using analysis of variance,Kruskal-Wallis test,Welch＇s t-test,and Chi-square test as appropriate before and after propensity score matching （PSM） based on age,gender,and body mass index （BMI）.Results：The prevalence of idiopathic BSSHL was 8.6% （114/1329） among the total SSHL *** the total cohort,USSHL patients tended to be younger,female,and tended to have lower BMI,renal parameters,and total cholesterol in addition to higher high-density lipoprotein compared to the other three *** routine blood indicators,some coagulation markers,and immunoglobulin M （H =13.4,P =0.004） were significantly different among the study *** PSM,the major significant differences were found in audiometric ***-BSSHL and Se-BSSHL patients demonstrated similar hearing thresholds as USSHL but were significantly better than the USSHLwCHL patients across most frequencies before and after treatment （H =30.0,P 〈 0.001 for initial hearing and H =12.0,P =0.007 for final hearing）.Moreover,the BSSHL patients showed different hearing loss distribution patterns （more descending type,x2 =33.8,P =0.001） with less hearing gain （H =17.5,P 〈 0.001） compared to the USSHL ***：Idiopathic BSSHL is a relatively rare subtype of SSHL with a higher rate of descending audiogram type and inferior hearing outcome rather than being classified as a completely

Macrophage-inducible C-type lectin is associated with anti-cyclic citrullinated peptide antibodies-positive rheumatoid arthritis in men

Background Macrophage-inducible C-type lectin （MINCLE） is an important member of C-type lectin superfamily, which has been shown evidence for susceptibility to arthritis in animal models. We aimed to investigate the possible association of MINCLE with rheumatoid arthritis （RA） susceptibility in Chinese Han population. Methods Haplotypes from HapMap database （Chinese Han Beijing, CHB） were used to select tag-single nucleotide polymorphism （SNP）（r2=0.8） residing in MINCLE gene. A total of 563 patients with RA and 404 healthy controls were TagMan genotyped for SNP rs10841845. Association analyses were performed on the whole data set and on RA subsets based on gender difference and the status of anti-cyclic citrullinated peptide （anti-CCP） antibody in RA patients. Association statistics were calculated by age and sex adjusted logistic regression. Results Overall, MINCLE SNP rs10841845 was not associated with susceptibility to RA. However, following anti-CCP stratification, rs10841845 GG genotypes conferred a significantly protective effects against anti-CCP-positive RA （OR 0.65, 95% CI 0.430-0.995, P=0.048）. Following gender stratification, SNP rs10841845 G allele appeared to insert its RA protective effect only in male patients, both at allele level （G vs. A OR 0.66, 95% CI 0.46-0.93, P=0.018） and at genotype level （GG vs. AA±AG, OR 0.429, 95% CI 0.20-0.95, P=0.036）. Notably, the male RA protective effect of rs10841845 G allele was only seen in anti-CCP-positive RA （G vs. A： OR 0.64, 95% CI 0.43-0.96, P=0.029; GG vs. AA＋AG： OR 0.375, 95% CI 0.14-0.94, P=0.038）. Furthermore, we observed a significant reduction of Disease Activity Score （DAS） 28 score （3.91±0.70 vs. 5.66±0.31, P=-0.022） and serum C-reactive protein levels （31.64±24.13 vs. 91.80±12.02, P=0.012） in male anti-CCP-positive RA patients carrying rs10841845 GG genotype, compared with patients carrying AA±AG genotypes. Emergency Response Plan (ERplan for short) plays an important role in carrying out emergency rescue effectively during the emergency response process. Unfortunately, the fact that ERplans are generally stored in form of text exposed some problems such as disunited standard, poor operability and so on, which affected the efficiency of emergency response, especially for cooperation of emergency response. In order to solve the deficiencies of text-based ERplan, the paper presents a feasible approach to digitalize the ERplan using workflow technology. For developing the ERplan workflow designer, the special features of ERplan are illustrated and modeling of business process of Tianjin overall ERplan with Petri Net was described in detail. Finally, the prototype of the emergency response cooperation platform based on ERplan workflow was developed to support the cooperation of inter-organizational for emergency response. The results showed that ERplan workflow provided a feasible solution for improving the performance of emergency response process.

Efficient electroreduction of nitrate via enriched active phases on copper-cobalt oxides

Electrochemical conversion of nitrate(NO_(3)~-) to ammonia(NH_(3)) can target two birds with one stone well, in NO_(3)^(-)-containing sewage remediation and sustainable NH_(3) production. However, single metalbased catalysts are difficult to drive high-efficient NO_(3)~- removal due to the multi-electron transfer ***, we present a tandem catalyst with simple structure, Cu-Co binary metal oxides(Cu-Co-O), by engineering intermediate phases as catalytic active species for NO_(3)~- conversion. Electrochemical evaluation,X-ray photoelectron spectroscopy, and in situ Raman spectra together suggest that the newly-generated Cu-based phases was prone to NO_(3)~- to NO_(2)~- conversion, then NO_(2)~- was reduced to NH_(3) on Co-based species. At an applied potential of -1.1 V vs. saturated calomel electrode, the Cu-Co-O catalyst achieved NO_(3)~- -N removal of 90% and NH_(3) faradaic efficiency of 81% for 120 min in 100 m L of 50 mg/L NO_(3)~- -N,consuming only 0.69 k Wh/mol in a two-electrode system. This study provides a facile and efficient engineering strategy for developing high-performance catalysts for electrocatalytic nitrate conversion.

The Bozhong19-6(BZ19-6)condensate gas reservoirs,located in the southwestern Bozhong sub-basin,Bohai Bay Basin,China,were paleo-oil reservoirs in the geological past and have undergone at least three successive hydrocarbon charging *** hydrocarbon migration and accumulation process of“early oil and late gas”has occurred in the current *** the end of the sedimentation of the Guantao Formation(N_(1)g,∼12 Ma),the reservoirs began to fill with first stage low-moderate mature crude *** the late stage of the Lower Minghuazhen Formation(N_(1)ml)(∼6.7 Ma),the reservoirs were largely charged with second stage high mature crude *** the deposition of the upper Minghuazhen Formation(N_(2)m^(u),∼5.1 Ma),the paleo-oil reservoirs were transformed into shallow Neogene reservoirs due to the reactivation of basement *** the late stage of the N_(2)m^(u)to the present day(∼2.8–0 Ma),the reservoirs were rapidly filled by natural gas within a short *** addition,analysis of the formation of the reservoir bitumen and the conspicuous loss of the lower molecular weight n-alkanes in the crude oil reveal that the injection of a large amount of gas in the late stage caused gas flushing of the early charged oil.

Expression pattern and tissue localization of the class B scavenger receptor BmSCRBQ4 in Bombyx moil

Class B scavenger receptors （SR-Bs） are cell surface glycoproteins involved in various physiological processes in vivo, including the transport and metabolism of lipids, binding and phagoeytosis of xenobiotics, and signaling. But little information is available about silkworm SR-Bs; it is necessary to study these SR-Bs for revealing their function. In this study, we cloned the full-length coding sequence of BrnSCRBQ4, a SR-B gene from the silkworm Bombyx mori L. We found that the BmSCRBQ4 gene consists of nine exons and eight introns, with an open reading frame of 1371 bp encoding 456 amino acids. Gene expression studies determined that BmSCRBQ4 messenger RNA （mRNA） was expressed in unfertilized eggs, during embryonic development and throughout the majority of the larval period. Expression of mRNA was detected in the mid gut, middle silk gland, posterior silk gland, head, integumentum, fat body, testes and the ovaries of the larval B. mori Dazao strain, as well as in the silkworm cell lines BmN and BmE. Protein expression studies found BmSCRBQ4 protein was expressed only in the testes, fat body and middle silk gland of larvae, as well as in the silkworm cell lines BmN and BmE. The BmSCRBQ4 protein showed variability in banding patterns in different tissues and cells when analyzed by Western blotting. Immunohistochemical staining showed that the BmSCRBQ4 protein localizes to the constitutive membranes or cellular membranes of these tissues. These results indicated that BmSCRBQ4 gene may play some physiologically relevant roles at the cell surface in each tissue.

Impact of atmospheric circulations on droughts and drought propagation over China

The GRACE satellite mission provides a new approach for monitoring,tracking,and assessing drought conditions by detecting changes in Earth's gravitational fields and inversing signals of terrestrial water storage *** studies of terrestrial water storage anomalies related droughts paid rare attention to the behind atmospheric physical mechanisms,nor quantified the risk propagation patterns between terrestrial water storage deficits and hydrological or agricultural *** this study,we first extract terrestrial water storage(TWS) droughts,hydrological droughts,and agricultural droughts by using multiple variables including TWS from the GRACE/GRACE-FO satellites,runoff and soil moisture from the ERA5-Land reanalysis *** then identify key atmospheric and oceanic oscillation indices affecting water deficits by employing machine learning *** characterize the joint distributions between drought duration and severity by using the Copula function and quantify the risk propagation of hydrological and agricultural droughts to TWS *** results show that:(1) From 2002 to 2021,there is a significant decrasing trend of TWS in China;the WPIO group atmospheric circulation indices(sea surface temperature index within the Western Pacific and Indian Ocean) contributes the most to ***,the sea surface temperature anomalies within the NINO 3.4 region as well as the Western Pacific Warm Pool area index show significantly positive correlation relationships with TWS in southern China;(2) The northwestern China,the Yangtze River basin,and the downstream of the Pearl River basin are the hotspots of TWS ***,the hotspots of hydrological droughts locate in northwestern China and the hotspots of agricultural droughts locate in South China and eastern Tibetan Plateau;(3) The elastic coefficients of hydrological droughts propagating to TWS droughts are higher than those of agricultural droughts,indicating that the TWS droughts

现有城轨车辆车体称重调簧模型忽略了车体的非均匀弹性变形、弹簧刚度的非线性等问题。针对该问题,文章首先建立了基于最小二乘法拟合二系支撑位刚度的改进称重调簧模型,该模型相较于传统称重调簧模型更贴合车体的实际负载情况;利用改进白骨顶鸡(COOT)算法对称重调簧算法进行多目标改造和搜索机制优化,相较于模拟退火(SA)算法、粒子群优化(PSO)算法、遗传算法(GA),改进COOT算法使稳定收敛载荷偏差分别降低了3.2 N、2.1 N和7.4 N,收敛速度分别提高了3.5%、6.8%和18.0%,具有更好的精度、效率和鲁棒性;基于C#和MATLAB搭建了整套称重调簧试验系统,并进行模拟试验及实车验证,证明了改进COOT算法及称重调簧系统的准确性和稳定性。
