Genomics of hepatitis B virus-related hepatocellular carcinoma and adjacent noncancerous tissues with cDNA microarray

Background Hepatocellular carcinoma （HCC） is a common primary cancer frequently associated with hepatitis B virus （HBV） infection. However, whether these identified genes are particularly associated with HBV-related HCC remains unknown. The aim of this study was to investigate the differential gene expression between HBV-related HCC tissues and adjacent noncancerous tissues. Methods cDNA microarray was used to detect the differential gene expression profile in the HBV-related HCC tissues and adjacent noncancerous tissues, and reverse transcription-polymerase chain reaction （RT-PCR） was performed to verify the differential expression of candidate genes obtained from cDNA microarray experiment. Results In this study, 1369 genes or expressed sequence tags （ESTs） including 121 genes or ESTs with at least two-fold expression alterations between cancerous and noncancerous tissues were identified. Special AT-rich sequence binding protein 1 （SATB-1） expression was positive in 73% （16/22） of cancerous tissues and negative （0/22） in all noncancerous tissues of HBV-related HCC patients. Transmembrane 4 superfamily member 1 （TM4SF-1） expression was positive in 86% （19/22） of cancerous tissues and negative （0/22） in all noncancerous tissues. Suppression of tumorigenicity 14 （ST-14） expression was positive in 73% （16/22） of noncancerous tissues in patients with HBV-related HCC and negative in all HCC tissues （0/22）. Conclusion This study provided the gene expression profile of HBV-related HCC and presented differential expression patterns of SATB-1, TM4SF-1 and ST-14 between cancerous and noncancerous tissues in patients with HBV-related HCC.

Background Increasing prevalence of Staphylococcus aureus （S. aureus）, particularly methicillin-resistant S. aureus （MRSA） has been reported in China. In this study, we investigated the drug resistance characteristic, genetic background, and molecular epidemiological characteristic of S. aureus in Changsha. Methods Between January 2006 and December 2008, 293 clinical isolates of S. aureus were collected from 11 hospitals in Changsha and identified by the Vitek-2 system. All the isolates were verified as MRSA by PCR amplification of both femA and mecA genes. K-B disk method was used to test drug sensitivity of S. aureus to antibiotics. Pulsed-field gel electrophoresis （PFGE） was performed for genotypic and homologous analysis of 115 isolates randomly selected from the original 293 clinical S. aureus isolates. Results S. aureus was highly resistant to penicillin, ampicillin, erythromycin, and clindamycin with resistant rates of 96.6%, 96.6%, 77.1%, and 67.2% respectively. All the isolates were susceptible to tecoplanin, vancomycin, and linezolid. MRSA accounted for 64.8% （190/293） of all the S. aureus strains. The 115 S. aureus isolates were clustered into 39 PFGE types by PFGE typing, with 13 predominant patterns （designated types A to M） accounting for 89 isolates. The most prevalent PFGE type was type A （n=56, 48.7%） and 100.0% of type A strains were MRSA. PFGE type A included 13 subtypes, and the most prevalent subtype was subtype A1 （46.4%, 26/56）. Strains with PFGE type A were isolated from eight hospitals （8/11）, and both subtypes A1 and A4 strains were isolated in a university hospital. Conclusions Clinical isolates of S. aureus in Changsha were resistant to multiple traditional antibiotics. There was an outbreak of PFGE type A MRSA in this area and the A1 subtype was the predominant epidemic clone. Dissemination of the same clone was an important reason for the wide spread of MRSA.

Experimental Study on Wire Melting Control Ability of Twin-Body Plasma Arc

Understanding the surface restructuring, including atomic rearrangement and chemical composition adjustments,is urgent to accelerate electrocatalytic reactions. Herein, we focus on the surface restructuring and active species for phosphide especially Ni2P during the water splitting process by combination of in-situ Raman and ex-situ X-ray photoelectron spectroscopy. Results confirm that NiOOH and NiOO* are formed during OER, while Niδ+-Hadactive sites are formed during HER. In situ-formed NiOOH/Ni2P@C and Niδ+/Ni2P@C show enhanced water splitting（1.514 V at 10 mA/cm2） and NO3RR(86% and 668 μmol h-1cm-2for NH3Faraday efficiency and yield)activities. In-depth investigation of the surface dynamic changes can provide guidance for establishing the relation of structure-electrocatalytic activity.

The twin-body plasma arc has the decoupling control ability of heat transfer and mass transfer,which is beneficial to shape and property control in wire arc additive *** this paper,with the wire feeding speed as a characteristic quantity,the wire melting control ability of twin-body plasma arc was studied by adjusting the current separation ratio(under the condition of a constant total current),the wire current/main current and the position of the wire in the arc axial *** results showed that under the premise that the total current remains unchanged(100 A),as the current separation ratio increased,the middle and minimum melting amounts increased approximately synchronously under the effect of anode effect power,the first melting mass range remained constant;the maximum melting amount increased twice as fast as the middle melting amount under the effect of the wire feeding speed,and the second melting mass range was *** the wire current increased,the anode effect power and the plasma arc power were both factors causing the increase in the wire melting amount;however,when the main current increased,the plasma arc power was the only factor causing the increase in the wire melting *** average wire melting increment caused by the anode effect power was approximately 2.7 times that caused by the plasma arc *** minimum melting amount was not affected by the wire-torch distance under any current separation ratio *** the current separation ratio increased and reached a threshold,the middle melting amount remained constant with increasing wire-torch *** the current separation ratio continued to increase and reached the next threshold,the maximum melting amount remained constant with the increasing wire-torch *** effect of the wire-torch distance on the wire melting amount reduced with the increase in the current separation *** this study,the decoupling mechanism and ability of this innovative

Tungsten-based materials are one of the most important candidates for the plasma facing materials（PFMs） of future fusion reactors,and it is necessary to study the properties of tungsten-based materials under the service conditions of fusion devices to ensure the long-term steady-state operation of future fusion *** this paper,different tungsten-based materials made by the same rotary swaging process are selected to simulate the transient thermal loads on the PFMs of a fusion reactor using an electron beam,and thermal load experiments are carried out by changing the power density of a loading pulse and the pulse *** is found that different tungsten-based materials made by the same process have large differences in thermal shock resistance,and the doped tungsten materials（W-K and W-La2O3） are significantly better than the pure tungsten materials with the increase in thermal shock power density and the number of cycles,especially the W-K materials,which have excellent performance.

For the functional partially linear models including flexible nonparametric part and functional linear part,the estimators of the nonlinear function and the slope function have been studied in existing *** to test the correlation between response and explanatory variables,however,still seems to be ***,a test procedure for testing the linearity in the functional partially linear models will be proposed in this paper.A test statistic is constructed based on the existing estimators of the nonlinear and the slope ***,we prove that the approximately asymptotic distribution of the proposed statistic is a chi-squared distribution under some regularity ***,some simulation studies and a real data application are presented to demonstrate the performance of the proposed test statistic.

Persistent mixing bursts in the equatorial Pacific thermocline induced by persistent equatorial waves

A recent study by Liu et al.(2020)suggested that due to the saturation of equatorially trapped planetary waves with different dynamical types,temporal periods,meridional and baroclinic modes,complex layer structures of vertical velocity shear and hence turbulent mixing could frequently occur in the thermocline of the eastern equatorial *** investigated the occurrence of the interior turbulent mixing as indicated by shear instabilities,above the Equatorial Undercurrent(EUC)core at three equatorial sites along 140°W,170°W,and 165°E,respectively,based mainly on data from the Tropical Atmosphere and Ocean(TAO)mooring *** found that turbulent mixing bursts persisted in the thermocline of all three ***,the interior turbulent mixing layers(ITMLs)could occur in probability of approximately 68%,53%,and 48%at the three sites,*** overall occurrence probability shows obvious and similar biannual variations at 140°W and 170°W,which is higher in boreal from late summer to winter and lower in ***,the ITMLs are primarily located above the EUC core and prevail in deeper(shallower)layers from late summer to winter(spring).Most ITMLs(70%)lasted for hours to 3 days,and a few of them(15%)for more than 7 *** thicknesses of ITMLs were concentrated between 15 and 55 *** 165°E,the vertical distribution of ITML occurrence probability was different from that at 140°W and 170°W,as it did not show a preference for depths;the durations of ITMLs are short(also from hours to several days)and their thicknesses were between 5 and 25 *** properties,particularly the high occurrence probability,and short durations demonstrated the persistence of thermocline mixing in the western to eastern equatorial Pacific thermocline and confirmed the generation mechanism by persistent equatorial waves as well.
