Seed-borne bacterial pathogens cause severe yield loss and biotoxin contamination in rice,leading to increasing concern on the global food supply and environmental *** native microbes play an important role in defending against diseases,but their actions are often influ-enced by the chemical fungicides applied in the ***,Bacillus licheniformis mmj was isolated from rice spikelet,which uniquely showed not only fungicide-responsiveness but also broad-spectrum antimicrobial activity against major rice bacterial pathogens including Xanthomonas oryzae ***,Burkholderia plantari and Burkholderia *** understand the hallmark underlying the environmental adaptation and anti-microbial activity of *** mmj,the genome sequence was determined by SMRT and subjected to bioinformatics *** sequence analysis enabled the identification of a set of antimicrobial-resistance and antibacterial activity genes together with an array of harsh environment-adaptive ***,*** mmj metabolites were analyzed with gas chromatography coupled to triple quadrupole mass spectrometry,and the volatile components that were linked with the antimicrobial activity were preliminarily ***,the present findings reveal the genomic and metabolic landscapes underlying fungicide-responsive ***,which offers a new opportunity to design harsh environment-adaptive biopesticides to cope with prevalent bacterial phytopathogens.

Using different endothelial/smooth muscle cell co-culture modes to simulate the intimal structure of blood vessels, the water filtration rate and the infiltration/accumulation of LDL of the cultured cell layers were studied. The three cell culture modes of the study were: (i) The endothelial cell monolayer (EC/Φ); (ii) endothelial cells directly co-cultured on the smooth muscle cell monolayer (EC-SMC); (iii) endothelial cells and smooth muscle cells cultured on different sides of a Millicell-CM membrane (EC/SMC). It was found that under the same condition, the water filtration rate was the lowest for the EC/SMC mode and the highest for the EC/Φ mode, while the infiltration/accumulation of DiI-LDLs was the lowest in the EC/Φ mode and the highest in the EC-SMC mode. It was also found that DiI-LDL infiltration/accumulation in the cultured cell layers increased with the increasing water filtration rate. The results from the in vitro model study therefore suggest that the infiltration/accumulation of the lipids within the arterial wall is positively correlated with concentration polarization of atherogenic lipids, and the integrity of the endothelium plays an important role in the penetration and accumulation of atherogenic lipids in blood vessel walls.

In this study,vehicle exhaust particles were collected three locations (the middle,entrance and outside) of the Dapu Road tunnel in downtown Shanghai,and the particle samples were characterized using SEM,XAFS,ICP-MS and M?ssbauer spectrometry,and the oxidative damage was assessed by plasmid *** iron-containing particles are found from vehicle exhaust,and iron oxide is the major species in all *** concentration in the particles inside the tunnel is higher than that the outside *** iron particles inside the tunnel have higher proportion of water-soluble fraction in hydrous iron sulfate form or smaller ***2 is the main soluble fraction in zinc-containing particles,while higher percentage of insoluble fraction existed in the particles outside of *** species of lead-containing particles are PbSO4,Pb3(PO4)2 and *** addition,the soluble fraction of other transition metallic elements as Ti and V is higher in the particles inside the tunnel than that outside the *** plasmid DNA assay results indicate that the particles from vehicle exhaust have a stronger oxidative damage and inflammation than that from outside of the tunnel.

OBJECTIVE: To investigate the efficacy of Yuzhizi(Fructus Akebiae Quinatae) seed extract(FAQSE) on inhibiting the proliferation of hepatocellular carcinoma(HCC) cells in vitro and to explore the anti-HCC action mechanism of FAQSE. METHODS: Human HCC Hep G2 and Huh7 cells were used to investigate the anti-HCC effect of FAQSE in vitro. 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide(MTT) method was used to measure cell viability. Affymetrix microarray was adopted to detect the expression of transcriptome. The differentially expressed genes(DEGs) of each cell line were identified. For coDEGs of both cell lines, the gene ontology(GO) and Kyoto Encyclopedia of Genes and Genomes(KEGG) pathway were enriched using the Database for Annotation, Visualization and Integrated Discovery(DAVID), and the network analysis of protein-protein interaction(PPI) was mapped using the Retrieval of Interacting Genes/Proteins(STRING) and Cytoscape software. Some important genes in the PPI network of coDEGs were selected to verify by quantitative real-time reverse transcription-polymerase chain reaction, Western blot and enzyme-linked immunosorbent assay. RESULTS: FAQSE decreased the viability of Hep G2 and Huh7 cells. There were 211 co-upregulated and 86 codownregualted genes in both cell lines after FAQSE treatment. The enriched GO terms of co-upregulated DEGs were primarily involved cell-cell adhesion, viral process, transcription initiation from RNA polymerase II promoter, positive regulation of transcription from RNA polymerase II promoter and actin cytoskeleton organization. The GO terms of co-downregulated DEGs were mainly enriched in the processes of SRP-dependent cotranslational protein targeting to membrane, viral transcription, nuclear-transcribed m RNA catabolic process, nonsense-mediated decay, translational initiation and r RNA processing.
