Prognostic significance of vascular endothelial growth factor polymorphisms in colorectal cancer patients

AIM To investigate the associations of the genetic polymor-phisms of vascular endothelial growth factor A(VEGf-A)-1498C>T and-634G>C, with the survival of patients with colorectal cancer(CRC). METHODS A prospective cohort consisting of 131 Brazilians patients consecutively operated on with a curative intention as a result of sporadic colorectal carcinoma was studied. DNA was extracted from peripheral blood and its amplification and allelic discrimination for each genetic polymorphism was performed using the technique of polymerase chain reaction(PCR) in real-time. The real-time PCR technique was used to identify the VEGf-A-1498C>T(rs833031) and-634G>C(rs2010963) polymorphisms. Genotyping was validated for VEGf-A-1498C>T polymorphism in 129 patients and for VEGf-A-634G>C polymorphism in 118 patients. The analysis of association between categorical variables was performed using logistic regression, survival by Kaplan-Meier method and multivariate analysis by the Cox regression method. RESULTS In the univariate analysis there was a significant association(OR = 0.32; P = 0.048) between genotype CC of the VEGf-A-1498C>T polymorphism and the presence of CRC liver metastasis. There was no association between VEGf-A-1498C>T polymorphism and VEGf-A-634G>C polymorphism with further clinical or anatomopathologic variables. The genotype CC of the VEGf-A-1498C>T polymorphism was significantly correlated with the 5-year survival(P = 0.032), but not significant difference(P = 0.27) was obtained with the VEGf-A-634G>C polymorphism with the 5-year survival in the univariate analysis. The genotype CT(HR = 2.79) and CC(HR = 4.67) of the polymorphism VEGf-A-1498C>T and the genotype CC(HR = 3.76) of the polymorphism VEGf-A-634C>G acted as an independent prognostic factor for the risk of death in CRC patients. CONCLUSION The CT and CC genotypes of the VEGf-A-1498C>T and the CC genotype of the VEGf-A-634C>G polymorphisms are prognostic factors of survival in Brazilians patients with spora

A contactor vessel was built to operate as either a mechanically or a conventional spouted bed for the purpose of analyzing the drying of pasty and granular materials under comparable conditions in both configurations. A classical conical–cylindrical spouted bed with a 60° conical base and an air inlet central orifice was modified to enable the switch by introducing in the vessel’s center an open helicoidal conveyor screw placed above the air inlet orifice. This screw is removable and conveniently returns the bed to its conventional spouted configuration. Experiments on drying solutions (calcium carbonate suspensions and skimmed milk) and granular materials (porous alumina particles) were performed for various bed parameter settings. The spouting pressure drop, outlet air temperature, and relative humidity were measured over time under different conditions. Mechanical agitation is proved effective in drying diluted carbonate calcium liquid suspensions and coarse porous alumina particles at an air velocity approximately 50% lower in comparison to drying in a conventional spouted bed. The conventional spouted bed performed better at drying liquid skimmed milk as the mechanical agitation combined with the axial inlet air flow is not effective in mitigating sticking and powder agglomeration in the bed when handling pastes of complex composition. Introducing mechanical agitation in the designed setup broadens the operating range of a conventional spouted bed with axial air flow in the inlet.

Objective: To evaluate the influence of fruiting phenological stage on total flavonoid content, antioxidant activity, and antiproliferative effects of Cereus jamacaru(C. jamacaru)(mandacaru) cladodes and fruit. Methods: fruit and cladodes at vegetative and fruiting stage of C. jamacaru were collected. The fruit was dissected and bark, pulp, and seeds were separated. Vegetative and fruiting cladodes, together with bark, pulp, and seeds were used to obtain five hydroalcoholic extracts. The extracts were investigated for total flavonoid content, using AlCl3 colorimetric method, antioxidant activity by 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) radical scavenging capacity and fe^(2+) ion chelating activity, and in vitro antiproliferative effects(sarcoma 180 cells) by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2 H-tetrazolium bromide assay. Results: The extract of C. jamacaru cladodes at the fruiting stage showed higher flavonoid content compared to the other extracts. Seed extracts showed the highest antioxidant activity in 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) assays, and the extract of cladodes at vegetative stage showed better antioxidant activity in fe^(2+) ion chelating activity. The extract of fruiting cladodes promoted higher antiproliferative effects compared to the other extracts. Conclusions: These findings suggest that fruiting increases the content of flavonoids and antiproliferative effects of C. jamacaru cladodes. Data reinforce the potential use of C. jamacaru cladodes and fruits as natural antioxidants and potent anticancer agent.

Technological innovation, driven towards the educational context, has favored the concept of interactive technological environments that may significantly contribute towards the teaching-learning processes. In that sense, mapping interactivity indicators that consider technical and operational aspects, supported by the available technical literature and based on the perspective of undergraduate engineers and teachers, becomes a fundamental activity in order to build interactive environments that may in fact adequately contribute for the professional education of our students, mainly the ones in engineering courses. Specifically, this paper shows preliminary studies within this perspective, showing how technological innovation may be understood and applied in the educational context. The study also shows the first interactivity indicator for a collaborative learning perspective, obtained from data collected through a qualitative content analysis methodology.

AIM: To investigate the effects of bariatric surgery on metabolic parameters, incretin hormone secretion, and duodenal and ileal mucosal gene ***: Nine patients with type 2 diabetes mellitus(T2DM), chronic serum hyperglycemia for more than2 years, and a body mass index(BMI) of 30-35 kg/m2 underwent metabolic surgery sleeve gastrectomy with transit bipartition between May 2011 and December2011. Blood samples were collected pre and 3, 6 and12 mo postsurgery. Duodenal and ileal mucosa samples were collected pre- and 3 mo postsurgery. Pre- and postoperative blood samples were collected in the fasting state before ingestion of a standard meal(520kcal) and again 30, 60, 90, and 120 min after the mealto determine hemoglobin A1c(HbA1c) levels and the lipid profile, which consisted of triglyceride and total cholesterol levels. Intestinal gene expression of p53 and transforming growth factor(TGf)-β was analyzed using quantitative reverse-transcription PCR. Gastric inhibitory polypeptide(GIP) and glucagon-like peptide-1(GLP-1)were quantified using the enzyme-linked immunoassay method and analyzed pre- and ***'s t test or repeated measurements analysis of variance with Bonferroni corrections were performed as ***: BMI values decreased by 15.7% within the initial 3 mo after surgery(31.29 ± 0.73 vs 26.398± 0.68, P f the patients experienced improved T2 DM, with 7 patients(78%)achieving complete remission(HbA1c f oral hypoglycemic agents). Background: Schizophrenia is a severe psychiatric disorder with a complex genetic factor determining its disease onset. Nevertheless, it is not clear in this mental disorder. Objective: To conduct a systematic review of articles regarding the genetic markers and mutations in schizophrenia. Methods: A systematic review of articles on genetic markers and mutations in schizophrenia, published from January 1, 2011, to September 7, 2015, on SCOPUS database was carried out. Search terms were “Genetic markers”, “Mutation”, and “Schizophrenia”. Results: Of the 527 retrieved studies, 31 met the eligibility criteria. Genetic polymorphism, Immune-associated genes, TCf4 and ZNf804A association with microRNA, Neuregulin gene, Chromosome 13q32 and 11p15.4, genes involved in glutamatergic via schizophrenia and brain structure, appeared to be associated with the origin of schizophrenia. Conclusion: Some studies show genes involved in several pathways leading to the disease pathogenesis such as that one related with the dopaminergic and immune system, or rare alleles. Some genes present no involvement in the etiology of this mental disorder. These findings clarify the genetic complexity of schizophrenia and affirm that together, the genes have an overall effect greater than the sum of the individual effect of each gene.

Repeat blocks, microsatellites or simple sequence repeats (SSRs) can produce good co-dominant molecular markers for genetic diversity analysis and the determination of self-pollination rates in progenies originating from open pollination of selected genotypes. The enrichment of guarana genomic libraries was underway when it was confirmed that we are working with a complex polyploid species with 210 chromosomes. The probes (CA)12, (CT)12 and (TC)14 were used to finish the enrichment of four libraries for repeat blocks and the screening of a databank of expressed sequence tags (ESTs) from guarana seeded-fruits was accomplished as well. fifteen clonal cultivars were genotyped with three replicas at 10 out of 27 identified loci using the 59 alleles that passed the reproducibility criterion. A large number of short repeat blocks were identified and this was considered to be a consequence of the recent polyploidization event. However, blocks with eight or more repeats ideal for genotyping were scarce. Annealing of most probes to short blocks by partial complementarity could explain the scarcity of longer blocks in genomic libraries but cannot explain why they were rare in the ESTs. Due to the complexity of the genotypes, alleles were treated as dominant traits. ESTs harboring repeat blocks had the functional annotation renewed. Locus GRN07 is inserted in a homologue of the MOTHER Of fLOWERING LOCUS T AND TfL1 (MfT), in which 3’-UTR displays clear post-transcriptional regulatory features. MfT and its variants are probably involved in the determination of seed germination and embryo growth characteristics. Other accessed loci can be involved in plant architecture and defense reactions. It was concluded that the alleles described in the present work can be used to distinguish guarana cultivars and possibly to analyze segregation using the progenies of controlled pollinations between divergent genitors.
