Local anes.hetics.inhibit cell p.oliferation and induce ap.p.os.s.in various.cell typ.s. Rop.vacaine, a unique, novel tertiary amine-typ. anes.hetic, was.s.own to inhibit the p.oliferation of s.veral cell typ.s.including keratinocytes. We found that Rop.vacaine could inhibit the p.oliferation and induce ap.p.os.s.in an immortalized human keratinocyte line, HaCaT, in a dos.- and time-dep.ndent manner and with the dep.ivation of s.rum. The dos.-dep.ndent induction of ap.p.os.s.by rop.vacaine was.demons.rated by DNA fragmentation analys.s.and the p.oteolytic cleavage of a cas.0}s.-3 s.bs.rate — p.ly (ADp.ribos.) p.lymeras. (p.Rp.. In addition, rop.vacaine downregulated the exp.es.ion of clus.erin/ ap.lip.rotein J, a p.otein with anti-ap.p.otic p.op.rties. in a dos.-dep.ndent manner, which well correlated with the induction of ap.p.os.s.of HaCaT cells. To inves.igate the role of clus.erin/ap.lip.rotein J in rop.vacaine-induced ap.p.os.s. HaCaT cells.overexp.es.ing clus.erin/ap.lip.rotein J were generated and comp.red to cells.exp.es.ing the well es.ab- lis.ed anti-ap.p.otic Bcl-2 p.otein. Ectop.c overexp.es.ion of the s.creted form of clus.erin/ap.lip.rotein J or Bcl-2 decreas.d the s.ns.tivity of HaCaT cells.to toxic effects.of rop.vacaine as.demons.rated by DNA fragmentation, the p.oteolytic cleavage of p.Rp.and by a reduction in p.ocas.0}s.-3 exp.es.ion. Furthermore, the downregulation of endogenous.clus.erin/ap.lip.p.otein J levels.by rop.vacaine s.gges.ed that this.might be one mechanis. by which rop.vacaine induced cell death in HaCaT cells. In conclus.on, the ability of rop.vacaine to induce antip.oliferative res.0}ns.s.and to s.p.res. the exp.es.ion of the anti-ap.p.otic p.otein clus.erin/ap.lip.p.otein J, combined with p.evi- ous.y rep.rted anti-inflammatory activity and analges.c p.op.rty of the drug, s.gges.s.that rop.vacaine may have p.ten- tial utility in the local treatment of tumors.
