Spatiotemporal microRNA profile in peripheral nerve regeneration:miR-138 targets vimentin and inhibits Schwann cell migration and proliferation

while the peripheral nervous system has regenerative ability,restoration of sufficient function remains a *** has been shown to be localized in axonal growth fronts and associated with nerve regeneration,including myelination,neuroplasticity,kinase signaling in nerve axoplasm,and cell migration;however,the mechanisms regulating its expression within Schwann cell（SC） remain *** aim of this study was to profile the spatial and temporal expression profile of micro RNA（mi RNA） in a regenerating rat sciatic nerve after transection,and explore the potential role of mi R-138-5 p targeting vimentin in SC proliferation and migration.A rat sciatic nerve transection model,utilizing a polyethylene nerve guide,was used to investigate mi RNA expression at 7,14,30,60,and 90 days during nerve *** levels of mi RNA expression were determined using microarray analysis and subsequently validated with quantitative real-time polymerase chain *** vitro assays were conducted with cultured Schwann cells transfected with mi RNA mimics and assessed for migratory and proliferative *** top seven dysregulated mi RNAs reported in this study have been implicated in cell migration elsewhere,and GO and KEGG analyses predicted activities essential to wound *** of one of these,mi RNA-138-5 p,into SCs reduced cell migration and *** R-138-5 p has been shown to directly target vimentin in cancer cells,and the luciferase assay performed here in rat Schwann cells confirmed *** results detail a role of mi R-138-5 p in rat peripheral nerve regeneration and expand on reports of it as an important regulator in the peripheral nervous system.

Neuromodulation for diabetic peripheral neuropathy represents a significant area of interest in the management of chronic pain associated with this *** peripheral neuropathy,a common complication of diabetes,is characterized by nerve damage due to high blood sugar levels that lead to symptoms,such as pain,tingling,and numbness,primarily in the hands and *** aim of this systematic review was to evaluate the efficacy of neuromodulatory techniques as potential therapeutic interventions for patients with diabetic peripheral neuropathy,while also examining recent developments in this *** investigation encompassed an array of neuromodulation methods,including frequency rhythmic electrical modulated systems,dorsal root ganglion stimulation,and spinal cord *** systematic review suggests that neuromodulatory techniques may be useful in the treatment of diabetic peripheral *** the advantages of these treatments will enable physicians and other healthcare providers to offer additional options for patients with symptoms refractory to standard pharmacologic *** these efforts,we may improve quality of life and increase functional capacity in patients suffering from complications related to diabetic neuropathy.

Prostate cancer cells metastasize to the hematopoietic stem cell niche in bone

The majority of men with advanced prostate cancer develop bone metastases as opposed to metastases at other sites. It has been unclear why prostate cancer selectively metastasizes to and proliferates in bone. Recently, Shiozawa et al. delineated a mechanism that may account for the establishment of prostate cancer in bone. Specifically, they identified that prostate cancer cells compete with hematopoietic stem cells （HSC） for the osteoblast in the HSC niche of the bone. Defining the mechanisms through which prostate cancer cells establish themselves in bone is critical towards developing effective therapeutic strategies to prevent or target bone metastases.

Genetic variation and potential coinfection of wolbachia among widespread Asian citrus psyllid (Diaphorina citri Kuwayama) populations

wolbachia can profoundly influence the survival, reproduction, and defenses of insect hosts. These interactions could potentially be harnessed for managing pests or insecttransmitted diseases. Diaphorina citri Kuwayama is a phloem-feeding pest capable of transmitting the putative causal agent of citrus greening, Candidatus Liberibacter asiaticus (CLas). Like many insects, D. citri is also infected with wolbachia (wDi). Recent studies indicate that the relative abundance of wDi could be associated with the abundance of CLas, and that wDi may contribute to regulating expression of phage lytic cycle genes in CLas, suggesting the need for better understanding of wDi biology in general. This study investigated the genetic diversity of wDi among D. citri in populations spanning eleven countries and two U.S. territories. Six wolbachia genes, wsp, ***, gatB, and hep A, were sequenced and compared across samples. Two prevalent wDi strains were identified across the samples, and screening of clone libraries revealed possible coinfection of wDi strains in specific populations. D. citri mitochondrial cytochrome oxidase subunit I gene (mtCOI) were more divergent between D. citri populations that were infected with different wDi strains or had different infection statuses (single infection vs. coinfection). while we could not eliminate the possibility that maternal transmission may contribute to such patterns, it is also possible that wDi may induce cytoplasmic incompatibility in their host. These fin dings should contribute to the understanding of wDi population ecology, which may facilitate manipulation of this endosymbiont for management of citrus greening disease worldwide.

Metasurfaces are ultrathin optical elements that are highly promising for constructing lightweight and compact optical *** their practical implementation,it is imperative to maximize the metasurface *** optimization provides a pathway for pushing the limits of metasurface efficiency;however,topology optimization methods have been limited to the design of microscale devices due to the extensive computational resources that are *** introduce a new strategy for optimizing large-area metasurfaces in a computationally efficient *** stitching together individually optimized sections of the metasurface,we can reduce the computational complexity of the optimization from high-polynomial to *** a proof of concept,we design and experimentally demonstrate large-area,high-numerical-aperture silicon metasurface lenses with focusing efficiencies exceeding 90%.These concepts can be generalized to the design of multifunctional,broadband diffractive optical devices and will enable the implementation of large-area,high-performance metasurfaces in practical optical systems.

Acute liver failure due to natural killer-like T-cell leukemia/ lymphoma: A case report and review of the Literature

Acute liver failure （ALF） is a medical emergency requiring immediate evaluation for liver transplantation. we describe an unusual case of a patient who presented with ascites, jaundice, and encephalopathy and was found to have ALF due to natural killer （NK）-Iike T cell leukemia/lymphoma. The key immunophenotype was CD2＋, CD3＋, CD7＋, CD56＋. This diagnosis, which was based on findings in the peripheral blood and ascitic fluid, was confirmed with liver biopsy, and was a contraindication to liver transplantation. A review of the literature shows that hematologic malignancies are an uncommon cause of fulminant hepatic failure, and that NK-like T-cell leukemia/lymphoma is a relatively recently recognized entity which is characteristically CD3＋ and CD56＋. This case demonstrates that liver biopsy is essential in diagnosing unusual causes of acute liver failure, and that infiltration of the liver with NK-like T-cell lymphoma/leukemia can cause acute liver failure.

Predicting the synthesizability of crystalline inorganic materials from the data of known material compositions

Reliably identifying synthesizable inorganic crystalline materials is an unsolved challenge required for realizing autonomous materials *** this work,we develop a deep learning synthesizability model(SynthNN)that leverages the entire space of synthesized inorganic chemical *** reformulating material discovery as a synthesizability classification task,SynthNN identifies synthesizable materials with 7×higher precision than with DFT-calculated formation *** a head-to-head material discovery comparison against 20 expert material scientists,SynthNN outperforms all experts,achieves 1.5×higher precision and completes the task five orders of magnitude faster than the best human ***,without any prior chemical knowledge,our experiments indicate that SynthNN learns the chemical principles of charge-balancing,chemical family relationships and ionicity,and utilizes these principles to generate synthesizability *** development of SynthNN will allow for synthesizability constraints to be seamlessly integrated into computational material screening workflows to increase their reliability for identifying synthetically accessible materials.

Clinical and experimental research in antituberculosis drug-induced hepatotoxicity:a review

Drug-induced liver injury is the common adverse effect seen in patients receiving antituberculosis drugs （ATDs）. There are several risk factors associated with the development of hepatotoxicity in such patients. Though there have been appreciable efforts taken by carrying out studies investigating the efficacy of several natural and synthetic compounds in minimising this effect, the only choice available for clinicians is withdrawal of drugs. This review would give a precise idea of ATD-induced hepatotoxicity its underlying mechanisms and alternative therapies for the same.
