Background The red macroalgae Asparagopsis is an effective methanogenesis inhibitor due to the presence of halogenated methane(CH_(4))analogues,primarily bromoform(CHBr_(3)).This study aimed to investigate the degradation process of CHBr3 from A taxiformis in the rumen and whether this process is diet-dependent.An in vitro batch culture system was used according to a 2×2 factorial design,assessing two A taxiformis inclusion rates[0(CTL)and 2%DM diet(AT)]and two diets[high-concentrate(HC)and high-forage diet(HF)].Incubations lasted for 72 h and samples of headspace and fermentation liquid were taken at 0,0.5,1,3,6,8,12,16,24,48 and 72 h to assess the pattern of degradation of CHBr_(3) into dibromomethane(CH_(2)Br_(2))and fermentation parameters.Additionally,an in vitro experiment with pure cultures of seven methanogens strains(Methanobrevibacter smithii,Methanobrevibacter ruminantium,Methanosphaera stadtmanae,Methanosarcina barkeri,Methanobrevibacter millerae,Methanorhermobacter wolfei and Methanobacterium mobile)was conducted to test the effects of increasing concentrations of CHBr3(0.4,2,10and 50μmol/L).Results The addition of AT significantly decreased CH_(4) production(P=0.002)and the acetate:propionate ratio(P=0.003)during a 72-h incubation.The concentrations of CHBr_(3) showed a rapid decrease with nearly 90%degraded within the first 3 h of incubation.On the contrary,CH_(2)Br_(2) concentration quickly increased during the first 6 h and then gradually decreased towards the end of the incubation.Neither CHBr_(3) degradation nor CH_(2)Br_(2) synthesis were affected by the type of diet used as substrate,suggesting that the fermentation rate is not a driving factor involved in CHBr_(3)degradation.The in vitro culture of methanogens showed a dose-response effect of CHBr3 by inhibiting the growth of M.smithii,M.ruminantium,M.stadtmanae,M.barkeri,M.millerae,M.wolfei,and M.mobile.Conclusions The present work demonstrated that CHBr_(3) from A.taxiformis is quickly degraded to CH_(2)Br_(2)in the rumen and that the ferm

There is no effective treatment to face Alzheimer’s disease complexity.Multitarget molecules are a good approach against the multiple physiopathological events associated with its development and progression.In this context,N-((5-(3-(1-benzylpiperidin-4-yl)propoxy)-1-methyl-1H-indol-2-yl)methyl)-N-methylprop-2-yn-1-amine(ASS234)has been tested achieving promising results.ASS234 has demonstrated to cross the blood-brain barrier in vivo,and a good in silico safety profile being less toxic than donepezil.Besides,ASS234 reversibly inhibits human acetyl-and butyryl-cholinesterase,and irreversibly inhibits human monoamine oxidase A and B.Moreover,this multitarget molecule has antioxidant and neuroprotective properties,and inhibitsΑβ1–42 andΑβ1–40 self-aggregation.Inquiring about the mechanism of action,several signaling pathways related to Alzheimer’s disease had been explored showing that ASS234 induces the wingless-type MMTV integration site(Wnt)family and several members of the heat shock proteins family and moreover counteracts neuroinflammatory and oxidative stress-related genes promoting the induction of several key antioxidant genes.Finally,in vivo experiments with ASS234 in C57BL/6J mice displayed its ability to reduce amyloid plaque burden and gliosis in the cortex and hippocampus,ameliorating scopolamine-induced learning deficits.Here we gather the information regarding ASS234 evaluated so far,showing its ability to face different targets,necessary to counteract a neurodegenerative disease as complex as the Alzheimer’s disease.

Throughout the shores of the world, a confluence of different sources of pollution is common, e.g., through outlets releasing brine, industrial, and domestic sewage. In this study, we assessed whether a combined disposal of domestic sewage and brine altered the patterns of abundance and assemblage structure of sub- tidal macrofauna inhabiting sandy seabeds on the south coast of Gran Canaria （Canary Islands, NE Atlantic Ocean）. Samples were collected in two surveys （May 2008 and January 2009） at three distances （0, 15, and 30 m） away from the discharge point. Macrofaunal abundances showed different patterns with varying proximity from the disposal point in the two surveys. In May 2008, lower abundances were observed at 0 m （53.89±46.82 ind.） than at 15 m （120±91.7 ind.） and 30 m （283.89±189.33 ind.） away from the dis- posal point. In January 2009, however, higher abundances were observed at 0 m （131.33±58.69 ind.） than at 30 m （100±24.44 ind.） and 15 m （84.78±58.39 ind.） away from the disposal point. Significant differ- ences in macrofaunal assemblage structure were found with varying proximity from the discharge point in both surveys. The effect of proximity from the disposal point was confounded by changes in sedimen- tary composition between surveys, which contributed to explain differences in macrofaunal abundances and assemblage structure with varying proximity from the discharge point. As a result, confounding driver- s of macrofaunal assemblage structure, here, changes in sediment composition, accounted for changes in macrofaunal abundances and assemblage structure with varying proximity from a combined disposal point of brine and sewage.

This study contributes with the first data on physical and taxonomical structure of macrofaunal assemblages of maerl beds from the Canary Islands. Maerl beds and Cymodocea nodosa meadows of the Canary Islands are considered biodiversity hot-spots in terms of taxonomic and functional biodiversity with a broad geographical and depth ranges. The authors have studied the structure of the macrofaunal assemblages on different habitat types （Cymodocea, Caulerpa, sabellid field, garden eel and maerl beds）. Samples were taken at a range of depths between 14 and 46 m. Correlations were performed among abiotic variables （granulometry, organic matter, nitrogen and phosphates） and the most abundant taxa. Similarity analysis was performed to explore the patchiness of seabeds at a local scale. Significant differences were found in macrofaunal assemblages among seabed types, with highest abundances and lowest biodiversity in sabellid fields, where the sabellid Bispira viola dominated. The polychaetes Aponuphis bilineata and Chone filicauda and the mollusc Turritella brochii were the most abundant taxa on maerl beds. The mosaic of granulometric conditions would explain the associated macroinfaunal community structure and contribute to the creation of diversity on these relatively well preserved seabeds at a local scale.

Abundances of the fireworm Hermodice carunculata were counted through a monitoring assessment study of fish cages in Barranco Hondo（NE Tenerife）. Seven campaigns were conducted from November 2007 to June 2010 and temporal variations were found, as well as differences among sampling stations. The polychaete H. carunculata obtained its highest abundance in sediments beneath fish cages throughout the study period. Thus, the assemblages of this omnivorous species were favoured by the presence of fish cages.

Background The pandemic caused by severe acute respiratory coronavirus 2(SARS-CoV-2)has had great effects on health systems worldwide,not only in relation to coronavirus disease 2019(COVID-19)cases but also affecting patients with other pathologies.Methods ECIEN-2020 is an observational study conducted in a tertiary referral hospital in Navarra,Spain.It describes the effects of COVID-19 pandemic and the preventive measures adopted,in pediatric admissions for non-COVID-19 diseases.Admissions during March-June 2020(first wave of the COVID-19 pandemic in Spain)are described and compared with the same quarter in 2019.A sub-analysis was performed delving into epidemiology.Patient characteristics(age,sex,past medical history),disease characteristics(symptoms,duration of symptoms,previous consultation in Primary Care Health Center),and admission characteristics(place and average stay)were analyzed.Results A 33% reduction in the number of pediatric hospital admissions was observed,decreasing from 529 hospitaliza-tions in 2019 to 353 in 2020(P<0.001).This highlights a 48% reduction in patients admitted for pulmonary diseases.There were no significant changes in average hospital-stay,percentage of intensive care unit admissions,or in admissions for other reasons.Percentage of patients admitted among those seen in the emergency department rose from 5.1%in 2019 to 10.9% in 2020,whereas the total number of consultations in the emergency department decreased by 68%.Conclusion The pandemic and the measures adopted due to SARS-CoV-2 have significantly decreased pediatric admissions for non-COVID-19 diseases,especially due to a reduction in the hospitalization for respiratory diseases.

Alzheimer’s disease (AD) is a neurodegenerative disease characterized by senile plaques and neurofibrillary tangles. Senile plaques are deposits of amyloid ?-peptide (A?) produced by the cleavage of a transmembrane protein termed Amyloid Precursor Protein (APP). The amyloidogenic cleavage of APP is performed by γ-secretase complex and ?-site APP cleaving enzyme 1 (BACE1), a key enzyme in AD that can be activated by different noxious stimuli. Interestingly, some viruses could activate double-stranded RNA-activated protein kinase (PKR), which phosphorylates Eukaryotic Initiation Factor 2 alpha (eIF2α). This phosphorylation stops global translation to avoid any synthesis of viral infective proteins, but paradoxically up-regulates BACE1 translation. One of the viral mechanisms to circumvent eIF2α phosphorylation is the recruitment of protein phosphatase 1 (PP1), to fully dephosphorylate eIF2α and allow viral protein synthesis. Due to the functional relationship between BACE1, PKR, PP1 and AD we have performed a large (1122 cases and 1191 control individuals) case-control genetic analysis using two biallelic polymorphisms rs2254958 and rs7480390, located within the genes coding for PKR and the catalytic unit A of PP1, respectively. Although a trend to association of the rs2254958 TT genotype with AD risk was found, our results show that neither rs7480390 nor rs2254958 are associated with AD susceptibility.
