Immunoprofiling of Drosophila Hemocytes by Single-cell Mass Cytometry

Single-cell mass cytometry(SCMC)combines features of traditional flow cytometry(i.e.,fluorescence-activated cell sorting)with mass spectrometry,making it possible to measure several parameters at the single-cell level for a complex analysis of biological regulatory *** this study,we optimized SCMC to analyze hemocytes of the Drosophila innate immune *** used metal-conjugated antibodies(against cell surface antigens H2,H3,H18,L1,L4,and P1,and intracellular antigens 3A5 and L2)and anti-IgM(against cell surface antigen L6)to detect the levels of antigens,while anti-GFP was used to detect crystal cells in the immune-induced *** investigated the antigen expression profile of single cells and hemocyte populations in naive states,in immune-induced states,in tumorous mutants bearing a driver mutation in the Drosophila homologue of Janus kinase(hopTum)and carrying a deficiency of the tumor suppressor gene lethal(3)malignant blood neoplasm-1[l(3)mbn1],as well as in stem cell maintenance-defective hdcD84 mutant *** analysis enabled the discrimination of the functionally different major hemocyte subsets for lamellocytes,plasmatocytes,and crystal cells,and delineated the unique immunophenotype of Drosophila *** have identified subpopulations of L2^(+)/P1^(+)and L2^(+)/L4^(+)/P1^(+)transitional phenotype cells in the tumorous strains l(3)mbn1 and hopTum,respectively,and a subpopulation of L4^(+)/P1^(+)cells upon immune *** results demonstrated for the first time that SCMC,combined with multidimensional bioinformatic analysis,represents a versatile and powerful tool to deeply analyze the regulation of cell-mediated immunity of Drosophila.

Characterisation of the granular dynamics at the interface between a pipe and a granular flow in a rotating drum

A new mobile bed heat exchanger is presented in this work which is composed of a flowing granular material in a rotating drum and a cylindrical pipe with potential interest in different energy applications as cooling,heating or heat recovery *** optimal design of the device requires a characterisation of the phenomena involved at the interface between the granular flow and the *** process is modelled by the discrete element method and a global classification of the flow patterns around the pipe is presented with respect to the three main control parameters of the problem:the Froude number,the diameter ratio and the relative filling height of the *** second part is devoted to the characterisation of the structure of the flow at the interface(velocity field,density field)in particular in a so-called Biflow regime where granular motion occurs above as well as below the pipe which is favourable to transfer by convection.A typical behavior at the interface with the pipe consists of a zone I with high velocities of particles at the top of the pipe,a second zone with quasistatic particles or low velocity particles at the front and at the bottom of the pipe and a last zone ll of depletion of particles at the back of the *** Froude number has a limited effect on the features of this structure on the first layer in the range of Froude numbers considered whereas the relative height is a more determinant parameter to control the relative magnitude of velocities in zone I and zone Il as well as the extent of the depletion *** first hydrodynamical characterisation can shed light on the dynamical regimes with improved transfer between the particles and the pipe boundary.

Animals constantly need to evaluate available external and internal information to make appropriate ***,assessing,and acting on relevant cues in contexts such as mate choice,intra-sexual competition,and parental care is particularly important for optimizing individual reproductive *** factors can infuence decision-making,such as external environmental cues and the animal’s own internal state,yet,we have limited knowledge on how animals integrate available ***,we used an entire island population(57 males,53 females,and 1,109 tadpoles)of the neotropical brilliant-thighed poison frog Allobates femoralis to investigate how 2 factors(olfactory cues and personality traits)infuence the ability of males to fnd and use new resources for tadpole *** experimentally manipulated the location of tadpole deposition sites and their associated olfactory cues,and repeatedly measured exploration and boldness in adult *** further reconstructed tadpole deposition choices via inferred parent–offspring relationships of adult frogs and tadpoles deposited in our experimental pools using molecular parentage *** found that the discovery and use of new rearing sites were heavily infuenced by olfactory cues;however,we did not fnd an effect of the measured behavioral traits on resource discovery and *** conclude that in highly dynamic environments such as tropical rainforests,reliable external cues likely take priority over personality traits,helping individuals to discover and make use of reproductive resources.

Dysfunction in circadian rhythms is a common occurrence in patients with Alzheimer’s disease.A predominant function of the retina is circadian synchronization,carrying information to the brain through the retinohypothalamic tract,which projects to the suprachiasmatic ***,Alzheimer’s disease hallmarks,including amyloid-β,are present in the retinas of Alzheimer’s disease patients,followed/associated by structural and functional ***,the mechanistic link between circadian dysfunction and the pathological changes affecting the retina in Alzheimer’s disease is not fully understood,although some studies point to the possibility that retinal dysfunction could be considered an early pathological process that directly modulates the circadian rhythm.

Dieback of managed spruce stands in western Germany promotes beetle diversity

Biodiversity loss is a significant problem at a global scale and may be amplified by climate *** recent years,coniferous forests have had substantial die-back across Europe due to drought and subsequent bark-beetle *** many studies on the consequences of disturbance and subsequent management have focused on natural stands,management implications for managed spruce stands are not well understood,even though such stands are widespread throughout *** this study,beetle taxonomy,conservation value,and community com-position are compared among spruce plantations and four post-disturbance management approaches:standing dead-wood,lying deadwood,clear cuts,and long-term *** and community composition differed significantly among management categories,while different beetle fami-lies responded *** spruce stands harbored the lowest beetle diversity while the highest taxonomic diver-sity and conservation value was on clear cuts and stands with lying or standing *** proportion of forest specialists was highest in successional *** summary,different forest management categories harbored distinct beetle communities at the family-,species-,and ecological guild ***,post-disturbance management should consider the landscape scale and include different management *** enhances landscape heterogeneity and thus overall biodiversity but could also mitigate negative impacts of natural disturbances on ecosystem services.

The intricacies of Alzheimer’s disease pathogenesis are being increasingly illuminated by the exploration of epigenetic mechanisms,particularly DNA *** review comprehensively surveys recent human-centered studies that investigate whole genome DNA methylation in Alzheimer’s disease *** examination of various brain regions reveals distinctive DNA methylation patterns that associate with the Braak stage and Alzheimer’s disease *** entorhinal cortex emerges as a focal point due to its early histological alterations and subsequent impact on downstream regions like the ***,ANK1 hypermethylation,a protein implicated in neurofibrillary tangle formation,was recurrently identified in the entorhinal ***,the middle temporal gyrus and prefrontal cortex were shown to exhibit significant hypermethylation of genes like HOXA3,RHBDF2,and MCF2L,potentially influencing neuroinflammatory *** complex role of BIN1 in late-onset Alzheimer’s disease is underscored by its association with altered methylation *** the disparities across studies,these findings highlight the intricate interplay between epigenetic modifications and Alzheimer’s disease *** research efforts should address methodological variations,incorporate diverse cohorts,and consider environmental factors to unravel the nuanced epigenetic landscape underlying Alzheimer’s disease progression.

A short climatology of black and brown carbon and their sources at a suburban site impacted by smoke in Brazil

Emissions from biomass burning challenge efforts to curb air pollution in cities downwind of fire-prone regions,as they contribute large amounts of brown carbon(Br C)and black carbon(BC)*** investigated the patterns of Br C and BC concentrations using Aethalometer data(atλ=370 and 880 nm,respectively)spanning four years at a site impacted by the outflow of *** data required to be post processed for the shadowing effect since,without correction,concentrations would be between 29%and 35%*** Br C concentrations were consistently higher than the BC concentrations,indicating the prevalence of aerosols from biomass *** results were supported by the Angstrom coefficient(A_(370/880)),with values predominantly larger than 1(mean±standard deviation:1.25±0.31).A_(370/880)values below 1 were more prevalent during the wet season,which suggests a contribution from fossil fuel *** observed sharp Br C and BC seasonal signals,with mean minimum concentrations of 0.40μg/m^(3)and 0.36μg/m^(3),respectively,in the wet season,and mean maximum concentrations of 2.05μg/m^(3)and 1.53μg/m^(3)in the dry *** largest concentrations were observed when northerly air masses moved over regions with a high density of fire *** burning of residential solid waste and industrial combustion caused extreme Br C and BC concentrations under favourable wind *** neither pollutant is included in any ambient air quality standards,our results suggest that transboundary smoke may hamper efforts to meet the World Health Organization guidelines for fine particles.

Since the discovery of hydride superconductors,a significant challenge has been to reduce the pressure required for their *** this context,we propose that alloying could be an effective strategy to achieve *** focus on a series of alloyed hydrides with the AMH_(6)composition,which can be made via alloying A15 AH_(3)(A=Al or Ga)with M(M=a group IIIB or IVB metal),and study their behavior under *** of them are predicted to maintain the A15-type structure,similar to AH_(3)under pressure,providing a platform for studying the effects of alloying on the stability and superconductivity of AH_(3).Among these,the A15-type phases of AlZrH_(6)and AlHfH_(6)are found to be thermodynamically stable in the pressure ranges of 40–150 and 30–181 GPa,***,they remain dynamically stable at even lower pressures,as low as 13 GPa for AlZrH_(6)and 6 GPa for AlHfH_(6).These pressures are significantly lower than that required for stabilizing A15 ***,the introduction of Zr or Hf increases the electronic density of states at the Fermi level compared with *** enhancement leads to higher critical temperatures(Tc)of 75 and 76 K for AlZrH_(6)and AlHfH_(6)at 20 and 10 GPa,*** the case of GaMH_(6)alloys,where M represents Sc,Ti,Zr,or Hf,these metals reinforce the stability of the A15-type structure and reduce the lowest thermodynamically stable pressure for GaH_(3) from 160 GPa to 116,95,80,and 85 GPa,*** noteworthy are the A15-type GaMH_(6)alloys,which remain dynamically stable at low pressures of 97,28,5,and 6 GPa,simultaneously exhibiting high Tc of 88,39,70,and 49 K at 100,35,10,and 10 GPa,***,these findings enrich the family of A15-type superconductors and provide insights for the future exploration of high-temperature hydride superconductors that can be stabilized at lower pressures.

The effect of slime accumulated in a long-term operating UASB using crude glycerol to treat S-rich wastewater

An up-flow anaerobic sludge blanket(UASB)reactor targeting sulfate reduction was operated under a constant TOC/S-SO2-4ratio of 1.5±0.3 g C/g S for 639 days using crude glycerol as carbon source.A filamentous and fluffy flocculant material,namely slime-like substances(SLS),was gradually accumulated in the bioreactor after the cease of methanogenic *** accumulation of SLS was followed by a decrease in the removal efficiencies and a deterioration in the *** characteristics of SLS were investigated to explore the causes of its formation and the effect of SLS on the UASB *** showed that glycerol fermentation and sulfate reduction processes taking place in the reactor were mainly accomplished in the bottom part of the UASB reactor,as the sludge concentration in the bottom was *** accumulation of SLS in the UASB reactor caused sludge flotation that further led to biomass washout,which decreased the sulfate and glycerol removal *** activity tests performed with granular sludge(GS),slime-covered granular sludge(SCGS)and SLS showed that there was no difference between GS and SLS in the mechanism of glycerol fermentation and sulfate ***,the specific sulfate reduction rate of GS was higher than that of SLS,while SLS showed a higher glycerol fermentation rate than that of *** different rates in GS and SLS were attributed to the higher relative abundances of fermentative microorganisms found in SLS and higher relative abundances of sulfate reducing bacteria(SRB)found in GS.
