Upscaling human mesenchymal stromal cell production in a novel vertical-wheel bioreactor enhances extracellular vesicle secretion and cargo profile

Human mesenchymal stromal cells(hMSCs)are mechanically sensitive undergoing phenotypic alterations when subjected to shear stress,cell aggregation,and substrate changes encountered in 3d dynamic bioreactor ***,little is known about how bioreactor microenvironment affects the secretion and cargo profiles of hMSC-derived extracellular vesicles(evs)including the subset,“exosomes”,which contain therapeutic proteins,nucleic acids,and lipids from the parent *** this study,bone marrow-derived hMSCs were expanded on 3d Synthemax II microcarriers in the PBS mini 0.1L Vertical-Wheel bioreactor system under variable shear stress levels at 25,40,and 64 RPM(0.1-0.3 dyn/cm^(2)).The bioreactor system promotes ev secretion from hMSCs by 2.5-fold and upregulates the expression of ev biogenesis markers and glycolysis genes compared to the static 2d *** microRNA cargo was also altered in the evs from bioreactor culture including the upregulation of miR-10,19a,19b,21,132,and *** protein cargo was characterized by proteomics analysis,showing upregulation of metabolic,autophagy and ROS-related proteins comparing with 2d cultured *** addition,the scalability of the Vertical-Wheel bioreactor system was demonstrated in a 0.5L bioreactor,showing similar or better hMSC-ev secretion and cargo content compared to the 0.1L *** study advances our understanding of bio-manufacturing of stem cell-derived evs for applications in cell-free therapy towards treating neurological disorders such as ischemic stroke,Alzheimer’s disease,and multiple sclerosis.

Targeting γ-secretase triggers the selective enrichment of oligomeric APP-CTFs in brain extracellular vesicles from Alzheimer cell and mouse models

This review compiles information on PCdd/F-and PCB-contaminated eggs from 20 years of global egg monitoring around emission sources in four continents conducted by the International Pollutants Elimination Network(IPEN)and Arnika as well as a compilation of data from scientific *** monitored 127 pooled egg samples including samples from 113 chicken flocks at potential PCdd/F-and PCB-contaminated sites around priority sources listed in the Stockholm Convention(*** incinerators,metal industries,cement plants,and open burning).99(88%)of pooled egg samples were above the EU maximum limits for PCdd/Fs(2.5 pg PCdd/F-TEQ/g fat)or the sum of PCdd/Fs and dioxinlike PCBs(5 pg PCdd/F-PCB-TEQ/g fat).Children consuming such eggs exceed the tolerable weekly intake(TWI).This demonstrates that close to 90%of these areas were not safe for the production of free-range *** out of the 113 egg samples(14%)were contaminated above 50 pg TEQ/g fat and exceeded the EU maximum limit more than 10 *** the 26 pooled egg samples around incinerators 24(92%)exceeded the limit with a mean of 43.1 pg TEQ/g fat(2.6e234 pg TEQ/g).All 21 egg samples around metal industries(4.4e112.6 pg TEQ/g fat)were above limits with mean concentration of 26.0 pg TEQ/g *** all 7 egg samples measured at e-waste recycling sites were above limits(mean 308 pg TEQ/g fat).In 58(51%)pooled egg samples the PCB-TEQ was above 5 pg TEQ/g fat exceeding the EU maximum limit with dioxin-like PCBs *** highlights the role of commercial PCBs for global contamination with dioxin-like *** was discovered that around metal industries,shredder plants,open burning sites of e-waste and dump sites,a high share of contamination was caused by *** clearly shows severe PCB release from the end-of-life management of PCB-containing equipment in developing *** highly contaminated eggs were found at many sites where plastic was *** highest contaminated egg sample ev

Problems faced by evidence-based medicine in evaluating lymphadenectomy for gastric cancer

Gastric cancer surgical management differs between Eastern Asia and Western countries. Extended lymphadenectomy(d2) is the standard of care in Japan and South korea since decades, while the majority of United States patients receive at most a limited lymphadenectomy(d1). United States and Northern Europe are considered the scientific leaders in medicine and evidencebased procedures are the cornerstone of their clinical practice. However, surgeons in Eastern Asia are more experienced, as there are more new cases of gastric cancer in Japan(107898 in 2012) than in the entire European Union(81592), or in South korea(31269) than in the entire United States(21155). For quite a long time evidence-based medicine(EBM) did not solve the question whether d2 improves long-term prognosis with respect to d1. Indeed, eastern surgeons were reluctant to perform d1 even in the frame of a clinical trial, as their patients had a very good prognosis after d2. evidence-based surgical indications provided by Western trials were questioned, as surgical procedures could not be properly standardized. In the present study we analyzed indications about the optimal extension of lymphadenectomy in gastric cancer according to current scientific literature(2008-2012) and surgical guidelines. We searched PubMed for papers using the key words "lymphadenectomy or d1 or d2" ANd "gastric cancer" from 2008 to 2012. Moreover, we reviewed national guidelines for gastric cancer management. The support to d2 lymphadenectomy increased progressively from 2008 to 2012: since 2010 papers supporting d2 have achieved a higher overall impact factor than the other papers. Till 2011, d2 was the procedure of choice according to experts' opinion, while three meta-analyses found no survival advantage after d2 with respect to d1. In 2012-2013, however, two meta-analyses reported that d2 improves prognosis with respect to d1. d2 lymphadenectomy was proposed as the standard of care for advanced gastric cancer by Japanese

Implantable(Bio)sensors as new tools for wireless monitoring of brain neurochemistry in real time

Implantable electrochemical microsensors are characterized by high sensitivity, while amperometric biosensors are very selective in virtue of the biological detecting element. Each sensor, specific for every neurochemical species, is a miniaturized hightechnology device resulting from the combination of several factors: electrode material, shielding polymers, applied electrochemical technique, and in the case of biosensors, biological sensing material, stabilizers, and entrapping chemical nets. In this paper, we summarizethe available technology for the in vivo electrochemical monitoring of neurotransmitters(dopamine, norepinephrine, serotonin, acetylcholine, and glutamate), bioenergetic substrates(glucose, lactate, and oxygen), neuromodulators(ascorbic acid and nitric oxide), and exogenous molecules such as ethanol. We also describe the most represented biotelemetric technologies in order to wirelessly transmit the signals of the abovelisted neurochemicals. Implantable(Bio)sensors, integrated into miniaturized telemetry systems, represent a new generation of analytical tools that could be used for studying the brain's physiology and pathophysiology and the effects of different drugs(or toxic chemicals such as ethanol) on neurochemical systems.

为了满足不同的技术和经济目标,从轻度混合动力、插电式混合动力到全电池动力的电动汽车,都将依赖于新型的、先进的(如基于锂的)蓄电池。这些电池在各种应用条件下的性能预测和寿命表征费工、费时,目前尚未得到充分的发展。一些国家已投入资金和人力进行相关的研究,其实通过国际合作,这些努力和花费也许能发挥更大的作用,例如目前正在国际能源机构(The International Energy Agency,IEA)框架内开展的准备工作。正在致力于开发一套标准化的、加速的测试程序,将允许各个测试机构合作分析电池的测量数据。该文评述了欧洲、日本和美国在加速寿命测试程序上的最新进展。以国际合作为目标,搜集、对比和分析现有的测试程序。
