Modular protein engineering is suited to recruit complex and multiple functionalities in single-chain polypeptides. Although still unexplored in a systematic way, it is anticipated that the positioning of functional domains would impact and refine these activities, including the ability to organize as supramolecular entities and to generate multifunctional protein materials. To explore this concept, we have repositioned functional segments in the modular protein T22-GFP-H6 and characterized the resulting alternative fusions. In T22-GFP-H6, the combination of T22 and H6 promotes selfassembling as regular nanoparticles and selective binding and internalization of this material in CXCR4-overexpressing tumor cells, making them appealing as vehicles for selective drug delivery. The results show that the pleiotropic activities are dramatically affected in module-swapped constructs, proving the need of a carboxy terminal positioning of H6 for protein self-assembling, and the accommodation of T22 at the amino terminus as a requisite for CXCR4^+ cell binding and internalization. Furthermore, the failure of self-assembling as regular oligomers reduces cellular penetrability of the fusions while keeping the specificity of the T22-CXCR4 *** these data instruct how multifunctional nanoscale protein carriers can be designed for smart, protein-driven drug delivery, not only for the treatment of CXCR4^+ human neoplasias, but also for the development of anti-HIV drugs and other pathologies in which CXCR4 is a relevant homing marker.

The maternal behavior in mammals is one of the most stable and highly conserved motivated species-specific behaviors and crucial for reproductive functions. During maternal behavior nursing and feeding are associated factors with survival of mammalian offspring, mammals nurse their newborn in a wide diversity in the behavioral. In the mother relationship the suckling is the behavioral patterns that identified and unified mammals and showed several selected evolutionary conserved behavior. The milk is an important reward for the pups, it ensure the surviving. The quality of maternal behavior is transmitted between generations;the inadequate maternal care has harmful effects in the adulthood individual. The specific maternal pattern modalities differ greatly between species;the human bonding has underlying biological mechanisms similar in mammals. Maternal brain is modified in order to prepare the mother-pup relationship, being crucial for the adequate behavioral display in the mother-pup relationship. A bulk of surrounding factor such as sensorial stimulus, temperature, ultrasonic communication, and reinforcing stimulus determines the quality of the nursing and care of the progeny. Mother care and nursing quality have long lasting effects in adulthood, determining cognitive and behavioral development and some specific psychiatric and psychological condition in human populations.

Designing technologies is a process that relies on multiple interactions between design and use contexts. These interactions are essential to the development and establishment of technologies. This article seeks to understand the attempts of healthcare organisations to integrate use contexts into the design of healthcare technologies following insights of the theoretical approaches of social learning and user representations. We present a multiple case study of three healthcare technologies involved in improving elderly care practice. These cases were part of a Dutch quality improvement collaborative program, which urged that development of these technologies was not “just” development, but should occur in close collaboration with other parts of the collaborative program, which were more focused on implementation. These cases illustrate different ways to develop technologies in interaction with use contexts and users. Despite the infrastructure of the collaborative program, interactions were not without problems. We conclude by arguing that interactions between design and use are not naturally occurring phenomena, but must be actively organised in order to create effects.

海参加工废弃物中多糖的理化性质及减肥活性研究

海参含有丰富的生物活性物质,具有低糖、低脂、高蛋白质、高膳食纤维的营养特性,并具有多种生物活性,近年来备受研究者关注。海参加工会产生大量废弃物,其中含有大量活性成分,但由于缺乏研究,没有得到充分利用,造成了宝贵资源的浪费和环境污染。基于此,本论文以海参加工废弃物为原料,提取分离海参多糖,并对其结构、理化性质和减肥活性进行了研究。采用单因素实验和响应面实验研究了海参多糖的最优提取条件,之后利用纤维素DEAE-52色谱柱和Sepharose 4B色谱柱对海参多糖进行了分离纯化。结果表明,最优提取条件为:81.7℃提取2次,每次提取1.16 h,提取得率为4.26%±0.11%;色谱柱分离纯化后得到三种多糖组分,分别为海参多糖I(SCIP-I)、海参多糖(SCIP-II)和海参多糖III(SCIP-III),其中SCIP-III占比最高。采用化学分析方法,结合HPLC、GC、FT-IR、NMR、SEM、AFM、TGA、DSC和微流变仪等仪器分析研究了SCIP-III的组成、结构和理化性质。结果显示,SCIP-III的总糖和蛋白质含量分别为82.20%和6.34%,由甘露糖,阿拉伯糖,半乳糖,葡萄糖和岩藻糖组成(摩尔百分比为22.03:19.31:11.78:3.22:42.57),Mw为4.91×10Da,多分散指数(Mw/Mn)为1.02;单糖残基之间以β型糖苷键连接;SCIP-III在SEM图像中为椭圆形颗粒,并与它由不同大小、球形结构和聚集的分子组成,AFM检测结果一致;DSC检测结果显示SCIP-III有两个峰,第一个峰是失水峰,Tp为89.12℃,总焓(ΔH)为-68.18,第二个峰是由于多糖的分子结构破坏导致,Tp为193.87°C,总焓(ΔH)为-70.84 J/g;热重分析结果中,30-130℃存在结合水和自由水损失的失重峰,200-400℃存在多糖糖苷键、糖环结构破坏的失重峰;微流变结果表明SCIP-III溶液的流动性、弹性和黏度随多糖浓度的增加而减小。采用高脂饲料建立肥胖小鼠模型,研究了海参多糖(SCIPs)的减肥作用。结果显示,SCIPs组能明显抑制肥胖小鼠体重增加,显著降低肥胖小鼠的脂肪指数、脂肪细胞大小和血脂水平(p<0.05),显著降低血清LPS含量,改善实验小鼠的糖耐受量(p<0.05),并可显著减少肝脏脂肪积累(p<0.05);此外,SCIPs可显著提高小鼠肠道短链脂肪酸含量(p<0.05),并可显著提高甘油三酯脂肪酶(ATGL)、激素敏感脂肪酶(HSL)、乙酰辅酶乙酰化酶(ACC1)、白细胞介素-6(Il-6)的m RNA含量(p<0.05)。肠道菌群分析结果显示,补充SCIPs可以改变厚壁菌门、变形菌门和拟杆菌门的相对丰度。厚壁菌门和拟杆菌门的比例从NC组的1.2增加到HFD组(肥胖小鼠)的1.45。海参多糖干预后,LSCIP、MSCIP和HSCIP组的厚壁菌门/拟杆菌门比例分别为0.87、1.3和0.95。SCIPs干预可以降低脱硫弧菌、双歧杆菌、细小杆菌和肠杆菌等有害厚壁细菌的比例,增加拟杆菌属、乳杆菌属、粪杆菌属和毛螺菌科等有益肠道菌群的多样性,有助于膳食未消化的生物活性脂质的发酵。上述结果证明SCIPs可通过调节与脂质代谢相关的酶、细胞因子的表达和肠道菌群组成实现降脂减肥的作用。

Background The positive influence of most types of exercise has been reported repeatedly,but what the most effective exercise approaches are for improving health-related quality of life(HRQoL)in people with cancer remains *** aim of this systematic review and network meta-analysis was to synthesize the evidence from intervention studies to assess the effects of different types of exercise on HRQoL during and after cancer *** MEDLINE,SPORTDiscus,the Cochrane Library,Web of Science,and Scopus were searched for randomized controlled trials aimed at testing the effects of exercise interventions meant to improve HRQoL in people with *** analyses were conducted for HRQoL as measured by general and cancer-specific *** also evaluated whether the effects of exercise were different during and after cancer treatment in both the physical and mental HRQoL *** In total,93 studies involving 7435 people with cancer were *** effect size estimates comparing exercise intervention *** care were significant for combined exercise(0.35,95%confidence interval(95%CI):0.14–0.56)for HRQoL as measured by general questionnaires,and for combined(0.31,95%CI:0.13–0.48),mind–body exercise(0.54,95%CI:0.18–0.89),and walking(0.39,95%CI:0.04–0.74)for HRQoL as measured by cancer-specific *** Exercise programs combining aerobic and resistance training can be recommended to improve HRQoL during and after cancer *** scarcity and heterogeneity of these studies prevents us from making recommendations about other exercise modalities due to insufficient evidence.

The development of a new generation of nonantibody protein drug delivery systems requires site-directed conjugation strategies to produce homogeneous,reproducible and scalable *** that,the genetic addition of cysteine residues into solvent-exposed positions allows the thiol-mediated cysteine coupling of therapeutic drugs into protein-based ***,the high reactivity of unpaired cysteine residues usually reduces protein stability,consequently imposing the use of more methodologically demanding purification *** is especially relevant for disulfide-containing nanocarriers,as previously observed in ***,although many protein scaffolds and targeting ligands are also rich in disulfide bridges,the use of these methodologies over emerging non-antibody carrier proteins has been completely ***,we report the development of a simple and straightforward procedure for a onestep production and site-directed cysteine conjugation of disulfide-containing non-antibody thiolated carrier proteins(THIOCAPs).This method is validated in a fluorescent C-X-C chemokine receptor 4(CXCR4)-targeted multivalent nanocarrier containing two intramolecular disulfide bridges and one reactive cysteine residue strategically placed into a solvent-exposed position(THIO-T22-GFP-H6)for drug conjugation and in a humanized alternative intended for clinical applications(T22-HSNBT-H6).Thus,we produce very stable,homogeneous and fully functional antitumoral nanoconjugates(THIO-T22-GFP-H6-MMAE and T22-HSNBT-H6-MMAE)that selectively eliminate target cancer cells via ***,the developed methodology appears as a powerful tool for the rational engineering of emerging non-antibody,cell-targeted protein nanocarriers that contain disulfide bridges together with a solvent-exposed reactive cysteine(THIOCAP).This should pave the way for the development of a new generation of stable,homogeneous and efficient nanomedicines.
