In vitro toxicity of particulate matter emissions from residential pellet combustion

Particulate matter emissions(PM10)from the combustion,in a residential stove,of two com-mercial brands of certified(ENplus A1)pellets,a non-certified brand and laboratory made pellets of acacia were tested for their ability to induce ecotoxic,cytotoxic,and mutagenic re-sponses in unicellular organisms and a human cell *** was evaluated through the Vibrio fischeri bioluminescence inhibition ***,cytotoxicity was assessed at two time points(24-and 48-hr)through two complementary techniques in order to evaluate the cellular metabolic activity and membrane integrity of human lung epithelial cells *** Ames test using two Salmonella typhimurium strains(TA100 and TA98)was employed to assess the mutagenic potential of the polycyclic aromatic hydrocarbon fraction extracted from the PM10 *** obtained with the bioluminescent bacteria indicated that only particles from the combustion of acacia pellets were *** samples induced im-pairment on the A549 cells metabolic activity,while no significant release of lactate dehy-drogenase was ***10 emissions from acacia pellets were the most cytotoxic,while samples from both certified pellets evoked significant cytotoxicity at lower ***-icity time-dependency was only observed for PM10 from the combustion of acacia pellets and one of the brands of certified *** activity was not detected in both *** murium *** study emphasises the role of the raw material for pellet manu-facturing on the toxicological profile of PM *** raw materials should be deeply investigated before their use in pelletisation and combustion in residential appli-ances.

Particulate matter(PM_(2.5))samples were collected in the vicinity of an industrial chemical pole and analysed for organic and elemental carbon(OC and EC),47 trace elements and around 150 organic *** average,OC and EC accounted for 25.2% and 11.4% of the PM_(2.5) mass,*** compounds comprised polycyclic aromatic hydrocarbons(PAHs),alkylated PAHs,anhydrosugars,phenolics,aromatic ketones,glycerol derivatives,aliphatic alcohols,sterols,and carboxyl groups,including aromatic,carboxylic and dicarboxylic *** factors>100 were obtained for Pb,Cd,Zn,Cu,Sn,B,Se,Bi,Sb and Mo,showing the contribution of industrial emissions and nearby major *** component analysis revealed that vehicle,industrial and biomass burning emissions accounted for 66%,11% and 9%,respectively,of the total PM_(2.5)-bound *** of the detected organic constituents are likely associated with plasticiser ingredients and thermal stabilisers used in the manufacture of PVC and other plastics in the industrial *** products of both anthropogenic(e.g.,toluene)and biogenic(e.g.,isoprene and pinenes)precursors were also *** was estimated that biomass burning accounted for 13.8%of the PM_(2.5) concentrations and that secondary OC represented 37.6% of the total *** lifetime cancer risk from inhalation exposure to PM_(2.5)-bound PAHs was found to be negligible,but it exceeded the threshold of 10−6 for metal(loi)s,mainly due to Cr and As.

Lacustrine sediments accumulate pollutants that input from the lake watershed and can be released to the water column by sediment resuspension; thus, pollutants can change their bioavailability and exert adverse effects to aquatic biota. Shallow-urban lakes are particularly susceptible to receive pollutants from urban discharges and sediment resuspension. Lake Xochimilco, in Mexico City, an urban-shallow lake, faces multiple problems： urban sprawl, overexploitation of aquifers, drying of springs, discharge of wastewater from treatment plants, and sediment resuspension. The aquatic biota living in this ecosystem is continuously exposed to the release of pollutants from the sediments. We assessed the risk that pollutants released from sediments from Lake Xochimilco, Touristic（TZ） and Agriculture zone（AZ）, can exert on a native amphibian species of the lake（Ambystoma mexicanum） through exposure bioassays to sediment elutriates. We evaluate alterations in the amphibian by three approaches： biochemical（level of lipid peroxidation,LPO）, cellular（ultrastructure） and the liver histology of A. mexicanum and we compare them with a batch control. Additionally, we assessed heavy metals（Pb, Cd and Hg） in *** from TZ showed the highest concentrations of the metals assessed. Organisms exposed to sediment elutriates from either study sites showed higher LPO values than control organisms（p 〈 0.05）. Organisms exposed to elutriates from the TZ showed the most conspicuous damages： hepatic vasodilation of sinusoids, capillaries with erythrocytes,leukocyte infiltration and cytoplasmic vacuolation in hepatocytes. The biological responses assessed reflected the risk that faces A. mexicanum when is exposed for prolonged periods to sediment resuspension in Lake Xochimilco.

BACKGROUNd：Liver ischemia reperfusion（IR）injury triggers a systemic inflammatory response and is the main cause of organ dysfunction and adverse postoperative outcomes after liver ***（PTX）and hypertonic saline solution（HTS）have been identified to have beneficial effects against IR *** study aimed to investigate if the addition of PTX to HTS is superior to HTS alone for the prevention of liver IR ***： Male Wistar rats were allocated into three groups. Control rats underwent 60 minutes of partial liver ischemia, HTS rats were treated with 0.4 mL/kg of intravenous 7.5% NaCl 15 minutes before reperfusion, and HPTX group were treated with 7.5% NaC1 plus 25 mg/kg of PTX 15 minutes before reperfusion. Samples were collected after reperfusion for determination of ALT, AST, TNF-α, IL-6, IL-10, mitochondrial respiration, lipid peroxidation, pulmonary permeability and myeloperoxidase. RESULTS： HPTX significantly decreased TNF-α 30 minutes after reperfusion. HPTX and HTS significantly decreased ALT,AST, IL-6, mitochondrial dysfunction and pulmonary myelo- peroxidase 4 hours after reperfusion. Compared with HTS only, HPTX significantly decreased hepatic oxidative stress 4 hours after reperfusion and pulmonary permeability 4 and 12 hours after reperfusion. CONCLUSION： This study showed that PTX added the beneficial effects of HTS on liver IR injury through decreases of hepatic oxidative stress and pulmonary permeability.

BACKGROUNd: Ischemic preconditioning(IPC) has been shown to decrease liver injury and to increase hepatic microvascular perfusion after liver ischemia reperfusion. This study aimed to evaluate the effects of IPC on hemodynamics of the portal venous system. METHOdS: Thirty-two rats were randomized into two groups: IPC group and control group. The rats of the IPC group underwent IPC by 10 minutes of liver ischemia followed by 10 minutes of reperfusion before liver ischemia, and the rats of the control group were subjected to 60 minutes of partial liver ischemia. Non-ischemic lobes were resected immediately after reperfusion. The animals were studied at 4 hours and 12 hours after reperfusion. Mean arterial pressure, heart rate, portal vein flow and pressure were analyzed. Blood was collected for the determination of the levels of aspartate aminotransferase, alanine aminotransferase, calcium, lactate, pH, bicarbonate, and base excess. RESULTS: IPC increased the mean portal vein flow at 4 hours and 12 hours after reperfusion. IPC recovered 78% of the meanportal vein flow at 12 hours after reperfusion. IPC decreased the levels of aspartate aminotransferase, alanine aminotransferase and lactate, and increased the levels of ionized calcium, bicarbonate and base excess at 12 hours after reperfusion. CONCLUSIONS: This study demonstrated that IPC increases portal vein flow and enhances hepatoprotective effects in liver ischemia reperfusion. The better recovery of portal vein flow after IPC may be correlated with the lower levels of transaminases and with the better metabolic profile.

Celecoxib enhances the detoxification of diethylnitrosamine in rat liver cancer

AIM： To study the effect of celecoxib （CXB） on diethylnitrosamine activation through the regulation of cytochrome P450 in a hepatocarcinogenesis ***： Six-week-old male Sprague-dawley rats were randomly divided into five groups, a non-treated group （NT）, a diethylnitrosamine-treated group （dEN）, a dEN＋CXB-treated group （dEN＋CXB）,and CXB 8 d-treated and CXB 32 d-treated groups. The effects of celecoxib on the enzymatic activities of CYP1A1, 2A, 2B1/2, and 2E1 were assessed in hepatic microsomes 24 h after dEN *** in CYPIA1 and CYP2B1/2 protein expression were also evaluated. The rate of dEN metabolism was measured by the production of the deethylation metabolite acetaldehyde, and the denitrosation metabolite ***： dEN＋CXB administration produced a significant increase in the enzymatic activities ofCYP2B1/2 and 1A1, whereas it did not change the activities of CYP2A and 2E1, compared to that of the dEN group. CXB treatment for eight days did not produce a significant effect on enzymatic activity when compared to the NT group; however, when it was administered for prolonged times （CXB 32 d group）,the enzymatic activities were increased in a similar pattern to those in the dEN＋CXB group. The observed increase in the enzymatic activities in the dEN＋CXB group was accompanied by an increase in the CYP2B1/2 protein levels; no changes were observed in the levels of CYPIA1. In vitro, CXB increased the denitrosation of dEN, a pathway of metabolic detoxification. The addition of SKF-525A, a preferential inhibitor of CYP2B, abrogated the denitrosation of ***： These results suggest that the mechanism of action of CXB involves enhancement of the detoxification of dEN by an increasing denitrosation via CYP2B1/2.

Post-fire recovery of Puya raimondii, vegetation and birds in the puna of Huascarán National Park, Perú

A large fire of 233 ha in Huascarán National Park in Peru provided an opportunity to compare plant and bird responses in burned and nearby unburned zones of the puna. Heights and live diameters of flagship Puya raimondii rosettes(assigned to four broad developmental phases), plant communities(66 species in 24 families and nine growth forms) and bird communities(77 species in six trophic guilds) were monitored after the fire. Although no mortality was observed, Puya raimondii plants were affected by the fire, losing approximately 60% of their photosynthetic area across all developmental phases, but recovered quickly during the first two years after fire. The comparison of Puya rosette recovery after fire was complicated by the changes in live rosette diameter for unburned plants, which showed plasticity of photosynthetic area linked to seasonal and annual fluctuations in precipitation in this relatively dry environment(decreased by 26% for mature adult plants over the study period). Fire caused an immediate change in the density, biomass and composition of vegetation. Although the species present remained similar, their abundances changed significantly immediately after the fire, with notable reductions in dominant tussock grasses. This provided opportunities for other plants, resulting in higher postfire diversity of plant species, genera, families and growth forms. In turn, the changes in vegetation after fire affected the composition of birds according to their trophic guild. Granivores largely disappeared,generalists were mostly unaffected, and other guilds showed a more complex response. As the vegetation recovered, most displaced birds returned within approximately one year.

On the distribution of the Minimum or Maximum of a Random Number of i.i.d. Lifetime Random Variables

Statisticians are usually concerned with the proposition of new distributions. In this paper we point out that a unified and concise derivation procedure of the distribution of the minimum or maximum of a random number N of indepen-dent and identically distributed continuous random variables Yi,{i = 1,2,…,N} is obtained if one compounds the probability generating function of N with the survival or the distribution func-tion of Yi. Expressions are then derived in closed form for the density, hazard and quantile func-tions of the minimum or maximum. The methodology is illustrated with examples of the distributions proposed by Adamidis and Loukas (1998), Kus (2007), Tahmasbi and Rezaei (2008), Barreto-Souza and Cribari-Neto (2009), Cancho, Louzada, and Barriga (2011) and Louzada, Roman and Cancho (2011).
