Upwelling coastal systems can be used to understand how dissolved oxygen and biological productivity control the accumulation of redox-sensitive metals in marine *** aluminium(Al),cadmium(Cd),iron(Fe),nickel(Ni),molybdenum(Mo),vanadium(V),total organic carbon(TOC),total nitrogen(TN)and total sulfur(TS)contents in surficial sediment collected from different water depths(30,70,and 120 m)in three northern Chilean bays influenced by coastal upwelling and oxygen minimum zones(OMZs)were *** component analysis(PCA),cluster analysis,and Spearman’s rank correlation were used to identify the mechanisms responsible for the redox-sensitive metal *** content of redox-sensitive metals and organic components in sediment increased with increasing water column depth,whereas lithogenic metals *** the Mejillones del Sur and Caldera bays,the enrichment factors of the redox-sensitive metals showed enrichment for all metals with *** Cd and V enrichments are mainly the product of biogenic flow to the seabed,and the Mo and Ni enrichments are due to preservation under low subsurface oxygen *** reduction is not an important mechanism in the accumulation of redox-sensitive metals in the sediment of the three *** PCA showed that the behaviors of the redox-sensitive metals and organic components reflect differences in the effects of the OMZ in sediment along the coast of northern Chile,with a more intense OMZ in Mejillones del Sur bay and weaker OMZs in Caldera and Inglesa ***,the high degree of enrichment in redox-sensitive metals in Caldera Bay can be attributed to the intense activity of the mining industry near the bay,a situation that produces geochemical behavior similar to that observed in Mejillones del Sur Bay.

Intrinsic subtypes and bladder cancer metastasis

Recent studies demonstrated that bladder cancers can be grouped into basal and luminal molecular subtypes that possess distinct biological and clinical *** bladder cancers express biomarkers characteristic of cancer stem cells and epithelial-tomesenchymal transition(EMT).Patients with basal cancers tend have more advanced stage and metastatic disease at *** preclinical models basal human orthotopic xenografts are also more metastatic than luminal xenografts are,and they metastasize via an EMT-dependent ***,preclinical and clinical data suggest that basal cancers are also more sensitive to neoadjuvant chemotherapy(NAC),such that most patients with basal cancers who are aggressively managed with NAC have excellent ***,luminal bladder cancers can also progress to become invasive and metastatic,but they appear to do so via mechanisms that are much less dependent on EMT and may involve help from stromal cells,particularly cancer-associated fibroblasts(CAFs).Although patients with luminal cancers do not appear to derive much clinical benefit from NAC,the luminal tumors that are infiltrated with stromal cells appear to be sensitive to anti-PDL1 antibodies and possibly other immune checkpoint ***,neoadjuvant and/or adjuvant immunotherapy may be the most effective approach in treating patients with advanced or metastatic infiltrated luminal bladder cancers.

Indirect calorimetry to estimate energy requirements for growing and finishing Nellore bulls

Determination of nutritional requirements is the basis for diet formulation. The objectives of this study were to determine the net energy requirements for maintenance （NEro） and weight gain （NEg） in Nellore bulls during the growing and finishing phases, and to estimate efficiency of metabolizable energy （ME） utilization for maintenance and gain （km, kg）. Five Nellore bulls were housed in individual pens at the Universidade Federal de Minas Gerais （Belo Horizonte, Brazil） and evaluated over four experimental periods at 210, 315,378 and 454 kg shrunk body weight （SBW）, approximately. During each period, heat production （HP） was quantified by open circuit indirect calorimetry for three feeding levels： ad libitum, restricted and fasting. The NEm requirement was determined by linear regression between the Log of HP andthe ME intake （MEI） for the ad libitum and restricted levels. This requirement was also determined by quantifying fasting heat production （FHP）. The NEQ requirement was calculated by the difference between MEI and HP during ad libitum feeding. The k and kg were calculated by the relationship between net energy （NE） and ME requirements for maintenance and weight gain （MEm, MEp）, respectively. The NEm requirements per kg of metabolic empty body weight （EBW0.75） fluctuated between 348 and 517 kJ d-1, showing a decreasing trend with age, and were higher than the values reported in the literature. The NEg requirements ranged between 48.3 and 164 kJ kg-1 EBW0.75 d-1, and varied according to age and weight gain. The k values varied between 58.6 and 69.7%, while kg varied between 23.4 and 40.2%. We concluded that NEm and NEg requirements were influenced by age and possibly by the level of stress, nervousness and activity of animals into the respirometry chamber. Further studies should quantify HP with records of positional changes （time spent standing vs. lying down）. Novel pharmacological therapy in type 2 diabetes mellitus with established cardiovascular disease: Current evidence

Cardiovascular diseases(CVDs) remain the leading cause of death in the world and in most developed countries. Patients with type 2 diabetes mellitus(T2DM)suffer from both microvascular and macrovascular diseases and therefore have higher rates of morbidity and mortality compared to those without T2DM. If current trends continue, the Center for Disease Control and Prevention estimates that 1 in 3 Americans will have T2DM by year 2050. As a consequence of the controversy surrounding rosiglitazone and the increasing prevalence of diabetes and CVDs, in 2008 the Food and Drug Administration(FDA) established new expectations for the evaluation of new antidiabetic agents, advising for pre and,in some cases, post-marketing data on major cardiovascular events. As a direct consequence, there has been a paradigm shift in new antidiabetic agents that has given birth to the recently published American Diabetes Association/European Association for the Study of Diabetes consensus statement recommending sodium-glucose cotransporter-2 inhibitors(SGLT2i) and glucagon like peptide-1 receptor agonists(GLP-1RA) in patients with T2DM and established CVD. As a result of over a decade of randomized placebo controlled cardiovascular outcome trials, the aforementioned drugs have received FDA approval for risk reduction of cardiovascular(CV) events in patients with T2DM and established CV ***2i have been shown to have a stronger benefit in patients with congestiveheart failure and diabetic kidney disease when compared to their GLP-1RA counterparts. These benefits are not withstanding additional considerations such as cost and the multiple FDA Black Box warnings. This topic is currently an emerging research area and this mini-review paper examines the role of these two novel classes of drugs in patients with T2DM with both confirmed, and at risk for, CVD.

Land cover changes and fragmentation in mountain neotropical ecosystems of Oaxaca, Mexico under community forest management

Changes in land cover have a direct impact on forest ecosystem goods and services. In this study, changes in land cover in Sierra de Juarez–Oaxaca ecosystems were estimated using a consistent processing of Landsat images and OBIA methodology. Additionally, landscape analyses using FRAGSTAT were conducted. In 2014, Sierra de Juarez–Oaxaca was covered by approximately 84% of forests, mainly pine-oak and cloud forests. After extensive deforestation until 2001, this trend was reversed and the forest cover surface area in 2014 was slightly higher than in 1979. The comparison of the landscape structure of the forested and agricultural lands suggests an increase in habitat heterogeneity. However, interspersion and juxtaposition indices, showing the patch shape by patch area and perimeter, were similar throughout the study period(1979–2014). Social and economic drivers can explain this situation: namely, community organization, forest enterprises, payment for ecosystem services programs, and changes of agricultural activity. Communities in the Sierra of Oaxaca have reforested degraded lands, created community forest enterprises, and preserved the forest under conservation schemes like those proposed by the Mexican payment for ecosystem services programs. However, their sustainable management faces internal challenges and has become highly dependent on political and institutional decisions beyond their control.

Edible coatings (EC) applied to fresh-cut fruits are used to increase their shelf-life and to deliver antioxidant bioactives such as phenolic compounds (PC) that reduce their oxidative damage while enhance their functional value. However, the combination of different PC may have synergetic, additive or antagonic effects on the final antioxidant capacity (AOXC). The aim of this study was to examine the AOXC of binary combinations of selected PC from mango peel and their bioaccessibility from 6% alginate-based EC applied to fresh-cut papaya, under simulated gastrointestinal conditions. Among equimolar (0.1 mM) combinations, gallic + protocatechuic acids (AB) were synergic in radical scavenging activity (RSA) as assayed by DPPH (90% RSA) and FRAP (0.39 mg TE/mL) methods;when assayed in 6% alginate-based EC, their RSA increased (117.85% RSA, 0.88 mg TE/mL). The application of EC + AB to papaya cubes and further in vitro digestion decreased their AOXC probably due to interactions between EC and papaya’s matrix. Therefore, further studies are needed in order to evaluate the effect of combination of phenolic and EC applied in other fruits matrix on antioxidants bioaccessibility.

Recognition of a multiple antigen peptide containing sequence from mimotope of the dengue type 3 virus NS4B protein by human antibodies

Objective: To evaluate the recognition of NS4 B mimotope, as multiple antigen peptide(MAP), by dengue antibodies presents in serum samples from patients with different serotype infections. Methods: A MAP containing mimotope sequence was synthesized and used to evaluate the recognition of NS4 B mimotope as MAP by a panel of 66 human sera from dengue cases by an indirect ELISA assay. Results: The MAP differentiated between sera from dengue viruses infected patients and sera from healthy individuals and the best reactivity was shown by serum from dengue type 3 virus patients. The recognition was more intense with serum from patients with secondary infection. Conclusions: The findings suggest the potential use of NS4 B mimotope on the development of a multi-epitope diagnostic tool. These results are important for further immunogenicity studies.

Anaplerosis in cancer: Another step beyond the warburg effect

Biosynthesis is up-regulated in tumors and thus the demand for anabolic intermediates is increased. The metabolic routes providing the building blocks for macromolecules are thus a very attractive target as they are not normally up-regulated in a normal quiescent cell. Some routes for glycolysis-derived intermediates production have been identified, but these do not constitute the whole pool of biosynthetic molecules in the cell, as many of these derive from mitochondria in the Krebs cycle. Indeed, this metabolic pathway is considered a “biosynthetic hub” from which anabolism is fed. If a metabolite efflux is indeed occurring, anaplerotic reactions must keep a steady supply of substrates. In spite of this obvious relevance of anaplerosis, it has been poorly characterized in the malignant cell context. Glutaminolysis and and pyruvate carboxylation are two pathways that function in an anaplerotic fashion. In spite of the increasing evidence implicating these two processes in cancer metabolism their role as intermediate providers is overlooked. In this review we analyze the implications of an active anaplerosis in cancer and we discuss experimental evidence showing the relevance of these metabolic routes in tumor physiology.
