Genomic characterization of multidrug-resistant extraintestinal pathogenic Escherichia coli isolated from grain culture soils

Multidrug-resistant(MDR)Escherichia coli,mainly extraintestinal pathogenic E.coli(ExPEC),has been widely reported in infections worldwide.In agricultural soils,manure is a hotspot for the dissemination of antimicrobial resistance genes(ARGs)and pathogenic bacteria;however,MDR bacteria have also been reported in soils with no history of manure use.In addition,cross-resistance and co-resistance have been described as responsible for the metal-driven selection of bacteria resistant to antimicrobials.Therefore,the aim of this study was to analyze three MDR E.coli isolates obtained from Brazilian grain culture soil samples with no history of manure use by whole-genome sequencing.The MDR E.coli isolates were recovered from soils from corn and coffee fields,and presented resistance toβ-lactams,quinolones,aminoglycosides,tetracyclines,sulphonamides,and dihydrofolate reductase inhibitor.Resistome analysis showed ARGs to several antimicrobials(i.e.,β-lactams,tetracyclines,aminoglycosides,sulphonamides,trimethoprim,phenicols,fosfomycin,and macrolides)as well as several metal resistance genes and antibacterial biocide resistance genes.In addition,known mutations in quinolone-resistance-determining regions of GyrA(Ser83Leu and Asp87Asn),ParE(Ser458Thr),and ParC(Ser80Ile)were also detected.Virulome analysis showed the presence of virulence genes(lpfA,mcmA,gad,mchF,iroN,cma,and iss)associated with ExPEC.Multidrug-resistant ExPEC isolates were assigned to phylogenetic group B1.The presence of MDR B1-ExPEC in soil samples shows the ability of these isolates to survive in soils.This study reports for the first time some sequence types(i.e.,ST345,ST448,and ST1146)of MDR E.coli in Brazilian soils.Therefore,these findings contribute to the monitoring of antimicrobial resistance and surveillance studies based on whole-genome sequencing worldwide.

The suicide phenomenon involves complex interactions between psychological,biological,cultural and socio-environmental factors.This study aimed to assess the epidemiological and toxicological profiles of officially confirmed suicide victims.Aretrospective study was performed using autopsy reports,forensic anthropology examinations and police summaries of all suicide cases that underwent toxicological analysis at an Official Forensic Laboratory(Minas Gerais,Brazil)in 1 year.The sample set was composed of 351 suicide victims,predominantly men(70.32%),most of them were adults between 31 and 64 years old(62.11%),with mixed skin colour(48.89%)and low educational level(66.44%).The most common suicide method was hanging(57.79%),followed by intoxication(30.45%).Most victims presented positive toxicological results(56.41%),especially for the presence of medicines(37.6%),illicit drugs(36.3%)and pesticides(26.1%).Our study corroborated previous data that most suicide victims have low educational levels.The most common toxicological findings were medicines,especially prescription drugs,followed by illicit drugs and pesticides.We hope this study contributes to reflections and planning of preventive suicide programmes,considering the described profiles of victims.

The collapse of the B1 Dam of VALESAmining company in Brumadinho,Minas Gerais,Brazil was the largest humanitarian disaster and occupational accident in the country’s history,and it posed challenges regarding the management and identification of multiple victims.We evaluated the impact of the iron ore tailings on the victims’bodies.We examined the scientific identification of the victims and the dynamics of the disaster over the 1st year after it occurred.We also determined the socio-demographic profiles of the victims.In this retrospective,cross-sectional study,we investigated the expert reports of the victims’biological remains from 25 January 2019 to 25 January 2020.We analysed the socio-demographic data,identification methods,identification status,identification time,and necroscopic information.During the study period,259 of 270 victims were identified,and 603 biological materials were analysed;among them,86.2%were body parts and 13.8%were whole bodies.Of the total cases registered that year,476(78.9%)were submitted during the first 10 weeks after the disaster.Friction ridge analysis accounted for 67.9%of primary identifications and DNAanalysis did so for 91.6%of re-identification cases.Body dismemberment was 3.4 times greater among mine workers than among community victims.Adult males accounted for the greatest number of victims(P<0.001).Polytraumatic injury was the prevalent single cause of death.Necropsy examination revealed the occurrence of asphyxia in 7%of cases.The higher number of fatalities and greater dismemberment among employees than with community residents underlines the occupational dangers in the mining industry and clarifies the dynamics of the disaster.In the initial weeks after the dam collapsed,friction ridge analysis was the most appropriate method for identification.Subsequently,DNAanalysis became the most-used technique for identification and re-identification owing to the great volume of body parts and decomposed biological tissue.Autopsy allowed diagnosis of the causes of death to be c

Laboratory measurements of airborne particles from sliding contacts are often performed using a tri- bometer located in a ventilation chamber. Although knowledge of particle transport behavior inside the chamber is required because it can influence the analysis of measurements, this knowledge is lacking. A numerical model was built based on the same geometry as a pin-on-disc measurement system to explain particle transport behavior inside the chamber and to determine the deviation between real amounts of generated and measured particles at the outlet. The effect of controlled flow conditions on the airflow pattern and particle transport inside the chamber was studied for different experimental conditions. Cal- culations show that a complex airflow pattern is formed by the spinning disc, and that it differs for each rotational speed. Simulation results reveal that particle transport in the chamber is governed mainly by the airflow pattern. The deposition velocity in the chamber was estimated and the possibility that part of the generated particles would remain in the chamber was studied. This led to an approximate estima- tion of particle loss rate. A comparison between experimental and simulated results with respect to the particle mass flow rate close to the outlet yields a reference factor of 0.7, which provides an indication of the difference between measured and real values.

Comparison between Different ESI Methods on Refractory Epilepsy Patients Shows a High Sensitivity for Bayesian Model Averaging

Electrical Source Imaging (ESI) is a non-invasive technique of reconstructing brain activities using EEG data. This technique has been applied to evaluate epilepsy patients being evaluated for epilepsy surgery, showing encouraging results for mapping interictal epileptiform discharges (IED). However, ESI is underused in planning epilepsy surgery. This is basically due to the wide availability of methods for solving the electromagnetism inverse problem (e-IP) associated to few studies using EEG setups similar to those most commonly used in clinical setting. In this study, we applied six different methods of solving the e-IP based on IEDs of 20 focal epilepsy patients that presented abnormalities in their MRI. We compared the ESI maps obtained by each method with the location of the abnormality, calculating the Euclidian distances from the center of the lesion to the closest border of the method solution (CL-BM) and also to the solution’s maxima (CL-MM). We also applied a score system in order to allow us to evaluate the sensitivity of each method for temporal and extra temporal patients. In our patients, the Bayesian Model Averaging method had a sensitivity of 86% and the shortest CL-MM. This method also had more restricted solutions that were more representative of epileptogenic activities than those obtained by the other methods.

AIM: To determine the clinical reasons for conversion to everolimus(EVL) and long-term outcomes in heart transplant(HT) recipients.METHODS: A retrospective 12-mo study has been carried out in 14 Spanish centres to assess the efficacy and safety of conversion to EVL in maintenance HT recipients.RESULTS: Two hundred and twenty-two patients were included(mean age: 53 ± 10.5 years; mean time from HT: 8.1 ± 4.5 years). The most common reasons for conversion were nephrotoxicity(30%), chronic allograft vasculopathy(20%) and neoplasms(17%). The doses and mean levels of EVL at baseline(conversion to EVL) and after one year were 1.3 ± 0.3 and 1.2 ± 0.6 mg/d and 6.4 ± 3.4 and 5.6 ± 2.5 ng/mL, respectively. The percentage of patients receiving calcineurin inhibitors(CNIs) at baseline and on the final visit was 95% and 65%, respectively. The doses and mean levels of CNIs decreased between baseline and month 12 from 142.2 ± 51.6 to 98.0 ± 39.4 mg/d(P < 0.001) and from 126.1 ± 50.9 to 89.2 ± 47.7 ng/mL(P < 0.001), respectively, for cyclosporine, and from 2.9 ± 1.8 to 2.6 ± 1.9 mg/d and from 8.3 ± 4.0 to 6.5 ± 2.7 ng/mL(P = 0.011) for tacrolimus. In the subgroup of patients converted because of nephrotoxicity, creatinine clearance increased from 34.9 ± 10.1 to 40.4 ± 14.4 mL/min(P < 0.001). There were 37 episodes of acute rejection in 24 patients(11%). The most frequent adverse events were oedemas(12%), infections(9%) and gastrointestinal problems(6%). EVL was suspended in 44 patients(20%). Since the database was closed at the end of the study, no further followup data is available.CONCLUSION: Conversion to EVL in maintenance HT recipients allowed minimisation or suspension of the CNIs, with improved kidney function in the patients with nephrotoxicity, after 12 mo.

The 2004 discovery of EGFR mutations,followed by ALK rearrangements,ushered in a targeted therapy era for advanced non-small cell lung cancer(NSCLC).Tyrosine kinase inhibitors targeting gene alterations have substantially improved survival and quality of life for patients with NSCLC.In the last decade,rearrangements of the ROS1 oncogene have been incorporated into healthcare practice that are applicable to another small subgroup of patients who benefit from similar targeted strategies.Recent genome studies of lung adenocarcinoma have identified other possible therapeutic targets,including RET,NTRK fusions,c-MET alterations,and activating mutations in KRAS,BRAF,and HER2,all with frequencies greater than 1%.Lung cancers harbouring these genome changes can potentially be treated with agents approved for other indications or under clinical development.This review updates the therapeutic arsenal that especially targets those genes.

The kinetics of the Fischer-Tropsch reaction over a Co/Nb2O5 catalyst in a fixed bed reactor was investigated experimentally. Experiments were carried out under isothermal and isobaric conditions （T=543 K, P=2.1 MPa） and under different conditions of several H2/CO feed molar ratio （0.49-4.79）, space velocities （0.2-3.8 h^-1）, mass of catalyst （0.3-1.5 g）, and CO conversion （10%-29%）. Synthesis gas conversion was measured and data were reduced to estimate the kinetic parameters for different Langmuir-Hinshelwood rate expressions. Differential and integral reactor models were used for the nonlinear regression of kinetics parameters. One of the rate equations could well explain the data. The hydrocarbon product distributions that were experimentally determined exhibited an unusual behavior, and a possible explanation was discussed.
