Background:Replication stress response is crucial for the maintenance of a stable ***3(DNA polymerase delta interacting protein 3)was initially identified as one of the DNA polymeraseδ(Polδ)interacting proteins almost 20 years *** a variety of in vitro biochemical assays,we previously established that POLDIP3 is a key regulator of the enzymatic activity of Polδ.However,the in vivo function of POLDIP3 in DNA replication and DNA damage response has been ***:We first generated POLDIP3 knockout(KO)cells using the cRISPR/cas9 *** then investigated its biological functions in vivo using a variety of biochemical and cell biology ***:We showed that although the POLDIP3-KO cells manifest no pronounced defect in global DNA synthesis under nonstress conditions,they are sensitive to a va-riety of replication fork ***,we found that POLDIP3 plays a crucial role in the activation and maintenance of the DNA damage checkpoint in response to exogenous as well as endogenous replication ***:Our results indicate that when the DNA replication fork is blocked,POLDIP3 can be recruited to the stalled replication fork and functions to bridge the early DNA damage checkpoint response and the later replication fork repair/restart.

Recent evidence shows that certain microRNAs （miRNAs） play a role in both obesity and prostate cancer recurrence, but the association between the expression of these miRNAs and obesity in prostate cancer recurrence is unknown. In this study, we examined the effect of the interaction between obesity and miR-21, miR-221 or miR-222 expression on prostate cancer recurrence among 28 recurrent and 37 non-recurrent prostate cancer cases, miRNA expression was determined using quantitative real-time polymerase chain reaction. cox proportional hazard models adjusting for age at diagnosis, clinical stage and Gleason score were used to estimate hazard ratios （HR） and 95% confidence intervals （95% cI） for recurrence free survival. A significantly （P=0.014） higher proportion of recurrent cases （78,6%） than non-recurrent cases （48.6%） had a low expression of miR-21 and the difference was more prominent in obese than non-obese patients. Multivariate analysis showed that the expression of miR-21 was an independent risk factor for recurrence in obese （HR=6.15, 95% cI= 1.04-36.48, P=-0.045）, but not in non-obese （HR= 1.28, 95% c1=0.30-5.49, P=0.74） cases. A significant association with recurrence was not observed for the expression of miR-221 and miR-222. In summary, our findings show that miR-21 is associated with prostate cancer recurrence after radical prostatectomy and suggest that the differential expression of miR-21 is more prominent in obese than in non-obese cases. Future larger studies are warranted to confirm these initial findings and to elucidate the mechanisms involved.

Lidar Measurements of Aerosols in the Tropical Atmosphere

Measurements of atmospheric aerosols and trace gases using the laser radar (lidar) techniques, have been in progress since 1985 at the Indian Institute of Tropical Meteorology, Pune (18°32'N, 73°51'E, 559 m AMSL), India. These observations carried out during nighttime in the lower atmosphere (up to 5.5 km AGL), employing an Argon ion / Helium-Neon lidar provided information on the nature, size, concentration and other characteristics of the constituents present in the tropical atmosphere. The time-height variations in aerosol concentration and associated layer structure exhibit marked differences between the post-sunset and pre-sunrise periods besides their seasonal variation with maximum concentration during pre-monsoon / winter and minimum concentration during monsoon months. These observations also revealed the influence of the terrain of the experimental site and some selected meteorological parameters on the aerosol vertical distributions. The special observations of aerosol vertical profiles obtained in the nighttime atmospheric boundary layer during October 1986 through September 1989 showed that the most probable occurrence of mixing depth lies between 450 and 550 m, and the multiple stably stratified aerosol layers present above the mixing depth with maximum frequency of occurrence at around 750 m. This information on nighttime mixing depth / stable layer derived from lidar aerosol observations showed good agreement with the height of the ground-based shear layer / elevated layer observed by the simultaneously operated sodar at the lidar site.

Using the Differential Absorption Lidar (DIAL) technique, two types of approaches, namely, reflection from retroreflector / topographic target and backscatter from atmosphere, are available for studying remotely the atmospheric NO, concentration. The Argon ion lidar system at the Indian Institute of Tropical Meteorology (IITM), Pune, India has been used for the measurements by following both the path-averaged and range-resolved approaches. For the former, a topographic target (hill) is used for determining path-averaged surface concentration. In the latter, spectral properties of atmospheric attenuation is used for making range-resolved measurements in the surface layer. The results of the observations collected by following both approaches are presented. The average surface NO2 concentration was found to vary between 0.01 and 0.105 ppm and the range-resolved measurements exhibited higher values suggesting treatment of the lidar data for scattering and extinction effects due to atmospheric aerosols and air molecules, and atmospheric turbulence. certain modifications that are suggested to the experimental set-up, data acquisition and analysis to improve the measurements are briefly described.

Optimal timing and route of nutritional support after esophagectomy: A review of the literature

Some controversy surrounds the postoperative feeding regimen utilized in patients who undergo *** in practices during the perioperative period exists including the type of nutrition started,the delivery route,and its *** nutrition is essential for this patient population as these patients often present with weight loss and have altered eating patterns after surgery,which can affect their ability to regain or maintain *** of feeding after an esophagectomy include total parenteral nutrition,nasoduodenal/nasojejunal tube feeding,jejunostomy tube feeding,and oral *** evidence suggests that early oral feeding is associated with shorter LOS,faster return of bowel function,and improved quality of *** recovery pathways after surgery pathways after esophagectomy with a component of early oral feeding also seem to be safe,feasible,and cost-effective,albeit with limited ***,data on anastomotic leaks is mixed,and some studies suggest that the incidence of leaks may be higher with early oral *** risk of anastomotic leak with early feeding may be heavily modulated by surgical *** definitive data is currently available to definitively answer this question,and further studies should look at how these early feeding regimens vary by surgical *** review aims to discuss the existing literature on the optimal route and timing of feeding after esophagectomy.

Spatio-temporal variations of water vapor optical depth in the lower troposphere (450-3850 m) over Pune (18°32′N, 73°51′E, 559 m Above Mean Sea Level), India have been studied over a period of five years. The mean vertical structure showed that the moisture content is greatest at the lowest level and decreases with increasing altitude, except in the south-west monsoon season (June to September) where an increase upto 950 m has been found. Optical depths are maximum in the monsoon season. The increase from pre-monsoon (March-May) to monsoon season in moisture content on an average is by about 58% in the above altitude range. The temporal variations in surface Relative Humidity and optical depth at 450 m show positive correlation. The amplitude of seasonal oscillation is the largest at 1465 m altitude. The time-height cross-sections of water vapor optical depths in the lower troposphere showed a contrast between years of good and bad monsoon.

Assessment of aerobic biodegradation of lower-chlorinated benzenes in contaminated groundwater using field-derived microcosms and compound-specific carbon isotope fractionation

Biodegradation of lower chlorinated benzenes(tri-, di-and monochlorobenzene) was assessed at a coastal aquifer contaminated with multiple chlorinated aromatic hydrocarbons. Field-derived microcosms, established with groundwater from the source zone and amended with a mixture of lower chlorinated benzenes, evidenced biodegradation of monochlorobenzene(McB) and 1,4-dichlorobenzene(1,4-DcB) in aerobic microcosms,whereas the addition of lactate in anaerobic microcosms did not enhance anaerobic reductive dechlorination. Aerobic microcosms established with groundwater from the plume consumed several doses of McB and concomitantly degraded the three isomers of dichlorobenzene with no observable inhibitory effect. In the light of these results, we assessed the applicability of compound stable isotope analysis to monitor a potential aerobic remediation treatment of McB and 1,4-DcB in this site. The carbon isotopic fractionation factors(ε) obtained from field-derived microcosms were-0.7‰ ± 0.1 ‰ and-1.0‰ ± 0.2 ‰ for McB and1,4-DcB, respectively. For 1,4-DcB, the carbon isotope fractionation during aerobic biodegradation was reported for the first time. The weak carbon isotope fractionation values for the aerobic pathway would only allow tracing of in situ degradation in aquifer parts with high extent of biodegradation. However, based on the carbon isotope effects measured in this and previous studies, relatively high carbon isotope shifts(i.e., Δδ13c > 4.0 ‰) of McB or 1,4-DcB in contaminated groundwater would suggest that their biodegradation is controlled by anaerobic reductive dechlorination.
