Is human hepatocellular carcinoma a hormone-responsive tumor?

Before the positive results recently obtained with multitarget tyrosine kinase inhibitor sorafenib,there was no standard systemic treatment for patients with advanced hepatocellular carcinoma(HCC).Sex hormones receptors are expressed in a significant proportion of HCC *** preclinical and epidemiological studies supporting a relationship between sex hormones and HCC tumorigenesis,several randomized controlled trials (RCTs)tested the efficacy of the anti-estrogen tamoxifen as systemic *** among these trials showed no survival advantage from the administration of tamoxifen,and the recent Cochrane systematic review produced a completely negative *** questions the relevance of estrogen receptor-mediated pathways in ***,a possible explanation for these disappointing results is the lack of proper patients selection according to sex hormones receptors expression,but unfortunately the interaction between this expression and efficacy of tamoxifen has not been studied *** has been also proposed that negative results might be explained if tamoxifen acts in HCC via an estrogen receptor-independent pathway,that requires higher doses than those usually administered, but an Asian RCT conducted to assess dose-response effect was completely ***,preliminaryresults have been obtained when hormonal treatment (tamoxifen or megestrol)has been selected according to the presence of wild-type or variant estrogen receptors respectively,but no large RCTs are available to support this *** results have been obtained also with anti-androgen *** conclusion,there is no robust evidence to consider HCC a hormone-responsive *** treatments should not be part of the current management of HCC.

Occurrence and patterns of nutritional traits and polycyclic aromatic hydrocarbons(PAHs)in sea cucumber(Holothuria poli)tissues:benefits and risk for human health

Objectives:The paper evaluates the benefit and risk for human health associated with consumption of sea cucumber Holothuria poli(*** from Italian coasts(Central Mediterranean Sea).Materials and Methods:Body wall(BW),internal tunic(ITu),muscle bands(MBs),alimentary canal(AC),gonad(Gd),and respiratory tree(RT)of *** were analyzed for proximate ***,amino acids(AAs),fatty acids(FAs)and polycyclic aromatic hydrocar bons(PAHs)were determined with high-performance liquid chromatography coupled with ultraviolet-visible spectroscopy(UPLC UV/Vis),gas chromatography-flame ionization detector(GC-FID)and gas chromatography mass spectrometry(GC-MS),***:Differences in the contents of total amino acids(TAAs)occurred based on tissue and sex,with AC and MB of female and Gd of male showing higher contents(range 47.8-60.2 g/kg we weight(ww)).Glycine and glutamic acid were the most *** fatty acid(PUFA)was the major class of FAs and arachidonic acid and eicosapentenoic acid(EPA)were the predominant PUFA.n-3 PUFA showed higher content in Gd,AC,and RT,indicating higher quality.A favorable n-3/n-6 in the range of 1.04-1.67 was *** showed values ranging from 23 to 207μg/kg ww with the highest levels in Gd and AC tissues and the lower in ***[alpyrene,the most toxic compound,was detected in all tissues,of both sexes,at levels of 1.5-18μg/kg ***:All tissues of ***,although with differences among them,are valuable food and can contribute for a healthy *** cancer risk(CR)values for Gd and AC tissues were above the considerable CR threshold of one in 10000 established by the United States Environmental Protection Agency(USEPA)for high ingestion rate of this seafood.

Iron homeostasis and H63D mutations in alcoholics with and without liver disease

MM： To evaluate the prevalence of HFE gene mutation and indices of disturbed iron homeostasis in alcoholics with and without liver disease. METHODS： One hundred and fifty-three heavy drinkers （defined as alcohol consumption 〉 80 g/d for at least 5 years） were included in the study. These comprised 78 patients with liver disease [liver disease alcoholics （LDA）] in whom the presence of liver disease was confirmed by liver biopsy or clinical evidence of hepatic decompensation, and 75 subjects with no evidence of liver disease, determined by normal liver tests on two occasions [non-liver disease alcoholics （NLDA）], were consecutively enrolled. Serum markers of iron status and HFE C282Y and H63D mutations were determined. HFE genotyping was compared with data obtained in healthy blood donors from the same geographical area. RESULTS： Gender ratio was similar in both study groups. LDA patients were older than NLDA patients （52 ± 10 years vs 48 ± 11 years, P = 0.03）. One third and one fifth of the study population had serum transferrin saturation （TS） greater than 45% and 60% respectively. Serum iron levels were similar in both groups. However, LDA patients had higher TS （51 ± 27 vs 36 ± 13, P 〈 0.001） and ferritin levels （559 ± 607 ng/mL vs 159 ± 122 ng/mL, P 〈 0.001）, and lower total iron binding capacity （TIBC）（241 ± 88 μg/dL vs 279 ± 40 μg/dL, P = 0.001）. The odds ratio for having liver disease with TS greater than 45% was 2.20 （95% confidence interval （CI）： 1.37-3.54）. There was no difference in C282Y allelic frequency between the two groups. However, H63D was more frequent in LDA patients （0.25 vs 0.16, P = 0.03）. LDA patients had a greater probability of carrying at least one HFE mutation than NLDA patients （49.5% vs 31.6%, P = 0.02）. The odds ratio for LDA in patients with H63D mutation was 1.57 （95% CI： 1.02-2.40）. CONCLUSION： The present study confirms the presence of iron overload in alcoholics, which was more severe in the subset of subjects with liver disease,

Essential Oils from the Leaves and Flowers of Callistemon viminalis:Chemical Characterization and Evaluation of the Insecticide and Antifungal Activities

The chemical compositions of the essential oils from the leaves and flowers of Callistemon viminalis and their insecticide and fungitoxic activities were determined. The essential oil was extracted by the hydrodistillation method using a modified Clevenger apparatus. The chemical characterization was performed by GC-MS and GC-FID. The evaluation of the insecticidal activity was performed with the Myzus persicae aphid, and the antifungal potential was determined via the inhibition of the mycelial growth of Alternaria alternata, Fusarium oxysorum and Botrytis cinérea phytopathogenic fungi. The principal components of the essential oils from the leaves and flowers were eucalyptol (84.60% and 61.47%), α-pinene (10.28% and 21.48%) and α-terpineol (2.59% and 2.79%), respectively. The use of a 0.5% concentration of the essential oil from the flowers influenced the preference of aphids and their reproduction. The number of adult aphids decreased within a period of 48 hours in the presence of the essential oil from the leaves. In the test with no chance of choice, the mean number of adults decreased with both oils within 48 hours. No inhibition of the mycelial growth of the A. alternate microorganism was observed in any of the treatments with the two essential oils. However, the growth of F. oxysporum and B. cinerea fungi was inhibited. The mycelial growth of F. oxysporum was inhibited with the concentration of 100 μL·Lˉ1 of the oil from the flowers and 250 μL·Lˉ1 of the oil from the leaves. The inhibition of the growth of B. cinerea was observed at concentrations of 500 and 100 μL·Lˉ1 for the oils from the flowers and leaves, respectively. The essential oils presented insecticidal and antifungal potentials. However, further studies are needed for these oils to be used in integrated pest management.
