Ocular penetration of topically applied 1% tigecycline in a rabbit model

AIM：To evaluate ocular penetration of topically applied 1% ***：Forty-two New Zealand White rabbits were divided into 3 groups. A 50 μL drop of 1% tigecycline was administered in group 1. In groups 2 and 3, the drop was administered every 15 min for 60min（keratitis protocol）. Aqueous humor samples in groups 1 and 2 were collected under general anesthesia at 15, 30, 45, 60, 120, and 180 min after the last drop. All animals in group 3 were euthanatized. Cornea, vitreous and blood samples were collected 60 and 120 min after the last drop. Tigecycline concentrations were measured using high performance liquid chromatography-mass spectrometry（LC-MS/MS）.RESULTS：The peak aqueous humor tigecycline concentration [mean 0.73±0.14 mg/L（SD） and 2.41±0.14 mg/L, respectively] occurred 45 min after topical drug application in groups 1 and 2. Group 3 mean values in the cornea, and vitreous, were 3.27±0.50 μg/g, and 0.17±0.10 mg/L at 60 min and 3.17±0.77 μg/g and 0.20±0.07 mg/L at 120 min, respectively. Tigecycline serum concentrations were ***：Tigecycline levels in the aqueous humor in groups 1 and 2, and in the cornea in group 3 exceeded the minimum inhibitory concentrations of most grampositive organisms that cause bacterial keratitis and endophthalmitis.

目的：比较1%布林唑胺和0.5%噻吗洛尔联合制剂（FCBT）与0.5%阿拉可乐定（APRA）滴眼液预防接受Nd：YAG激光晶状体囊切开术患者术后眼压升高的疗效。方法：前瞻随机对照临床研究。研究包括90例（90眼）接受Nd：YAG激光晶状体囊切开术治疗后囊膜混浊（PCO）患者。患者术前1h随机给予APRA（n=45）或FCBT（n=45）治疗。一名设盲检查者使用Goldmann压平眼压计检查术前及术后1,2,3,24h和7d的眼压。眼压检查结果分为以下两类：术后眼压升高5~〈10 mm Hg和眼压升高≥10 mm Hg。眼压升高〈5 mm Hg认为没有临床上的显著改变。结果：APRA组和FCBT组术前当天的平均眼压分别为14.1±2.1 mm Hg和13.2±2.1 mm Hg,差异无统计学意义（P=0.066）。随访期间,FCBT组的平均眼压较低但差异无统计学意义。APRA组中的6名患者（13.3%）和FCBT组中的4名患者（8.9%）术后至少有一次眼压升高5~〈10 mm Hg,差异无统计学意义（P=0.243）。眼压升高≥10 mm Hg在APRA组与FCBT组中分别出现3眼（6.7%）和1眼（2.2%）,差异无统计学意义（P=0.542）。结论：APRA和FCBT均可有效预防眼压升高且APRA足以应对常规病例。对于需要加强降低眼压的患者-如预先存在青光眼的患者（其术后眼压升高的风险更大）可选择FCBT。

Measurement of urinary β2M （beta 2-microglobulin） is a sensitive and reliable assay for detecting tubular injury, renal toxicity, lymphomas, leukemia, or myeloma. Some chemical substrates may increase the level of β2M in-vivo. Elevated β2M level in urine is unusual because it rapidly degrades when pH is below 6. The level of β2M in the bladder can also be used as a marker to assess renal tubular maturation in neonates. β2M in the bladder could be a result of fetal megacystis, which is an abnormally enlarged bladder appearing after 10 weeks of gestation, when the fetus begins to produce urine. Identification of the pregnant women instead of the fetus is a common pre-analytical error with samples sent from the gynecology clinic to the laboratory. Here we present the case of a 24-year-old pregnant woman whose urine analysis results indicated excessively high β2M level in the urine. The present study could improve the understanding of urinary β2M analysis, laboratory errors, and the interpretation of test results.

Understanding how plant cells adapt dynamically to changes in the environment is a fundamental problem of plant biology. Under many conditions, plant cells respond to environmental changes by modifying their intracellular organization. A critical example of intracellular reorganization is chloroplast photo-relocation, which is required for optimal energy harvesting and avoiding photodamage. A key system responsible for the spatial organization of intracellular components is the microtubule cytoskeleton and its associated motor proteins, kinesins. Here we tested the hypothesis that members of the kinesin 4II subfamily are important for chloroplast photo-relocation in the moss Physcomitrella patens. Most land plants, including P. patens, use an actin cytoskeleton-dependent mechanism to transport chloroplasts in response to light. In addition to the actin-based system, P. patens can also transport chloroplasts via a microtubule-dependent mechanism, which is absent in flowering plants. Here, we used a P. patens line that contains an inducible RNAi system to silence all three kinesin 4-II genes present in this moss and evaluated their participation in the microtubule-dependent chloroplast light avoidance response. Because we found a significant effect on cell growth when kinesin 4IIs are silenced, we took advantage of the inducible system to establish a reproducible and quantitative assay to evaluate chloroplast photo-relocation in full-grown cells. Using a laser scanning confocal-based chloroplast light avoidance response assay, we found a reduction in chloroplast motility when kinesin 4IIs were silenced. Hence, in addition to identifying a role for kinesin 4II proteins in protonemal cell growth, our results strongly support the hypothesis that these kinesins play an important role in the chloroplast light avoidance response.

Geological hazards caused by high-temperature rocks cooling down after encountering water are closely related to underground mining and tunneling *** fully understand the impact of temperature changes on the mechanical properties of rocks,yellow rust granite samples were subjected to heating-natural cooling and heating-water cooling cycles to experimentally study the effects of these processes on the mechanical properties of the *** mechanism of the heating-cooling process on the macromechanical properties of the rock was *** on the Drucker-Prager criterion and Weibull distribution function,a damage variable correction factor was introduced to reflect the post-peak strain softening characteristics,and a thermo-mechanical coupled damage constitutive model of the granite was *** results showed that in the natural cooling mode,the mechanical properties deteriorate significantly when the temperature exceeded 600C,and the failure mode changed from brittle failure to ductile *** the water cooling mode,the peak strength and deformation modulus increased at temperatures below 400C with an increase in the cycle number,while at 600C,the peak strength and elastic modulus notably *** peak strain increased with the increase of the cycle number and temperature at all temperatures,and the failure mode of the granite tended to change from tensile failure mode to shear failure *** experimental results were used to validate the damage constitutive *** shape parameter r and scale parameter S in the Weibull distribution function of the model were used as indicators to reflect the brittleness degree and peak *** study helps to understand the behavior of rocks in hightemperature environments,in order to prevent and mitigate potential geological hazards.

To the editor： We read with great interest the article by Guo and co-authors published in the current issue of Chinese Medical Journal. These authors 1 assessed the effect of aliskiren on arterial stiffness, compared with ramipril in mild to moderate essential hypertensive patients. The study revealed that aliskiren （150 mg/d） could improve arterial stiffness and its effect was similar to ramipril （5 mg/d） in mild to moderate hypertensive patients. This study gives detailed and valuable information and highlights the effects of aliskiren on arterial stiffness compared with ramipril. Some comments may be of interest.

In Turkey, studies devoted to conservation of animal genetic resources are carried out by general directorate of agricultural research （TAGEM） under the ministry of food, agriculture and livestock. There are 9 sheep and 3 goat breeds and 3 silkworm lines which produce animal fiber have been put under conservation by TAGEM since 1996. A considerable amount of these genotypcs are preserved as in situ. The success of this conservation method is directly associated with economically making the best of these genotypes which are preserved. Hence, making the most of fiber production alongside meat and milk production would make a great contribution to their conservation. For this purpose, determining production biology of mohair and textile features of Ankara goats at present, determining all features of traditional items （especially handicrafts） made of fibers obtained from these breeds and lines which have been put under protection and creating new usage areas have been issues worked upon. Along with this, in recent years, just as the world, especially EU countries, interests and studies for issues such as organic animal fiber production, rural development and geographical indications have been increasing in Turkey, as well. In the near future, this situation is supposed to have a great effect on increasing of animal fiber production in a positive way, too. Hence, to benefit more effectively from fibers obtained from these breeds which have been put under protection will make a great contribution to be protected by breeders. For this purpose, there will be studies in the near future devoted to determining production biology of fiber and determining features of textile in breeds and lines and increasing the consumption of products to be obtained from these fibers as well as Ankara goats. Therefore, in this article, it was emphasized on the more efficient of ways to be benefited from fiber production in these breeds and lines under protected.

Chemotherapy-Induced Tumour Lysis Syndrome in Gastric Adenocarcinoma with Diffuse Liver Metastases:A Case Report

Tumour Lysis Syndrome (TLS) is an important oncological emergency case which is often found together with haematological malignities and, much less often, with solid tumours. While TLS seen in solid tumour cases usually develops following a cytotoxic chemotherapy and its prognosis is poor. We present the case of a 60-year-old man with gastric adenocarcinoma with diffuse liver metastases (image shows diffuse liver metastatic lesions) and high serum LDH levels, who developed TLS after systemic chemotherapy. With urgent and proper supportive treatment (intravenous intensive hydration, sodium bicarbonate, diuretic, calcium gluconate, allopurinol and haemodialysis), an impressive recovery from TLS was achieved in the patient with an advanced stage gastric cancer. The purpose of this report is to emphasize that although the present case was a rare, high physician attention is required because significant morbidity or mortality may occur when the syndrome is not duly considered during the pre-cytotoxic evaluation of the patient, when preventive measures are not taken, or if the appropriate treatment is not applied immediately once the syndrome appears, especially in patients who have high tumour burden solid cancer.

Statistical Analysis of Bursa Nilufer Creek’s Water Quality Parameters for Period of 2002-2010

Increasing contamination of water resources in the world and our country and decreasing water quality over time, not having met the objectives of utilization of water resources;it has increased the importance of water management. The monitoring of the water resources and evaluation of these monitoring results have given direction to the studies’ outcome in order to control factors that pollute water resources and reduce water quality. Nilüfer Creek is very important for both being a source of drinking and potable water and a discharge area for wastewaters for the city of Bursa. In this study, the results of the analysis belonging to the period between 2002-2010 which are taken from 15 points by General Directorate of Bursa Water and Sewerage Administration (BUWSA) were evaluated in relation to water quality of the Nilüfer Creek. Non-parametric methods were used in the evaluation of the water quality data due to the lack of normally distributed data. The identification of the best represented parameters of the water quality was provided by applying Principal Component Analysis. According to results of the analysis, the best representative 9 parameters from the 19 water quality parameters were defined as parameters of BOD5, COD, TSS, ***, Zn, conductivity, NO2-N, Ni and NO3-N that taking part of the first two components.
