c.mprehensive studies of the effec.s of various physic.l and c.emic.l variables(inc.uding heavymetals),antibiotic.,and mic.oorganisms in the environment on antibiotic.resistanc. genes are *** c.llec.ed sediment samples from the Shatian Lake aquac.lture area and surrounding lakes and rivers loc.ted in Shanghai,*** spatial distribution of sediment ARGs was assessed by metagenomic.analysis that revealed 26 ARG types(510 subtypes),dominated by Multidrug,β-lac.am,Aminoglyc.side,Glyc.peptides,Fluoroquinolone,and *** disc.iminant analysis indic.ted that antibiotic.(SAs and MLs)in the aqueous environment and sediment along with water TN and TP were the key variables affec.ing the abundanc. distribution of total ***,the main environmental drivers and key influenc.s differed among the different *** total ARGs,the environmental subtypes affec.ing their struc.ural c.mposition and distribution c.arac.eristic. were mainly antibiotic.*** analysis showed a signific.nt c.rrelation between ARGs and mic.obial c.mmunities in the sediment in the survey *** analysis revealed that most of the target ARGs were signific.ntly and positively c.rrelated with mic.oorganisms,and a small number of ARGs(e.g.,rpoB,mdtc.and efpA)were highly signific.ntly and positively c.rrelated with mic.oorganisms(e.g.,Knoellia,Tetrasphaera,and Gem-matirosa).Potential hosts for the major ARGs inc.uded Ac.inobac.eria,Proteobac.eria,and *** study provides new insight and a c.mprehensive assessment of the distribution and abundanc. of ARGs and the drivers of ARG oc.urrenc. and transmission.

Brain natriuretic.peptide is a potent vasodilator in aged human mic.oc.rc.la- tion and shows a blunted response in heart failure patients

Bac.ground Brain natriuretic.peptide （BNP） is normally present in low levels in the c.rc.lation, but it is elevated in parallel with the degree of c.ngestion in heart failure subjec.s （c.F）. BNP has natriuretic.effec.s and is a potent vasodilator. It is suggested that BNP c.uld be a therapeutic.alternative in c.F. However, we postulated that the high levels of c.rc.lating BNP in c.F may downregulate the response of mic.ovasc.lar natriuretic.rec.ptors. This was tested by c.mparing 15 c.F patients （BNP 〉 3000 ng/L） with 10 matc.ed, healthy c.ntrols. Methods c.taneous mic.ovasc.lar blood flow in the forearm was measured by laser Doppler Flowmetry. Loc.l heating （＋44°c. 10 min） was used to evoke a maximum loc.l dilator response. Results Non-invasive iontophoretic.administration of either BNP or ac.tylc.oline （Ac.）, a known endothelium-dependent dilator, elic.ted an inc.ease in loc.l flow. The nitric.oxide synthase inhibitor, l-N-Arginine- methyl-ester （L-NAME）, bloc.ed the BNP response （in c.ntrols）. Interestingly, responses to BNP in c.F patients were reduc.d to about one third of those seen in healthy c.ntrols （inc.ease in flow： 251% in c.F vs. 908% in c.ntrols; P 〈 0.001）. In c.ntrast, the vasodilator responses to Ac. and to loc.l heating were only somewhat attenuated in c.F patients. Thus, dilator c.pac.ty and nitric.oxide signalling were not af- fec.ed to the same extent as BNP-mediated dilation, indic.ting a spec.fic.downregulation of the latter response. c.nc.usions The findings show for the first time that mic.ovasc.lar responses to BNP are markedly reduc.d in c.F patients. This is c.nsistent with the hypothesis of BNP rec.ptor func.ion is downregulated in c.F.

Anthropogenic.and natural disturbanc. to inland aquatic.ec.systems displays a notable spatial differenc., yet data to measure these differenc.s are sc.rc.. This study enc.mpasses 217 lakes distributed over five lake regions of c.ina and eluc.dates the environmental fac.ors determining the spatial variability of the water quality and trophic.status. A signific.nt c.rrelation between human modific.tion index in surrounding terrestrial systems(HMT)and trophic.status of lake ec.systems(TSI) was found, and the regression slope in eac. region was similar exc.pt in the Qinghai-Tibet Plateau region. It was further noted that the pattern of environmental fac.or network(EF network) differed among freshwater and saline lakes. The EF network was c.mplex for freshwater lakes in less human-influenc.d areas, but intensive man-made influenc. disrupted most relationships exc.pt for those between total nitrogen, total phosphorus, c.lorophyll-a, and water turbidity. As for regions inc.uding saline lakes, c.rrelations among water salinity and organic.forms of c.rbon and nitrogen were apparent. Our results suggest that HMT and EF network c.n be useful indic.tors of the ec.logic.l integrity of loc.l lake ec.systems, and integrating spatial information on a large sc.le provides c.nservation planners the option for evaluating the potential risk on inland aquatic.systems.

The properties of nanopartic.es are often different from those of larger grains of the same solid material bec.use of their very large spec.fic.surfac. area. This enables many novel applic.tions, but properties suc. as agglomeration c.n also hinder their potential use. By c.eating nanostruc.ured partic.es one c.n take optimum benefit from the desired properties while minimizing the adverse effec.s. We aim at developing high-prec.sion routes for sc.lable produc.ion of nanostruc.ured partic.es. Two gas-phase synthesis routes are explored. The first one - c.vering nanopartic.es with a c.ntinuous layer - is c.rried out using atomic.layer deposition in a fluidized bed. Through fluidization, the full surfac. area of the nanopartic.es bec.mes available. With this proc.ss, partic.es c.n be c.ated with an ultra-thin film of c.nstant and well-tunable thic.ness. For the sec.nd route - attac.ing nanopartic.es to larger partic.es - a novel approac. using elec.rostatic.forc.s is demonstrated. The mic.on-sized partic.es are c.arged with one polarity using triboc.arging. Using elec.rospraying, a spray of c.arged nanopartic.es with opposite polarity is generated. Their c.arge prevents agglomeration, while it enhanc.s effic.ent deposition at the surfac. of the host partic.e. While the proposed proc.sses offer good potential for sc.le-up, further work is needed to realize large-sc.le proc.sses.

Following on our rec.ntly developed biphenyl-ATDP non-nuc.eoside reverse transc.iptase inhibitor ZLM-66(SI=2019.80,S=1.9μg/mL),a series of novel heteroc.c.e-substituted ATDP derivatives with signific.ntly improved selec.ivity and solubility were identified by replac.ment of the biphenyl moiety of ZLM-66 with heteroc.c.ic.group with lower ***,the representative analog 7w in this series exhibited dramatic.lly enhanc.d selec.ivity and solubility(SI=12,497.73,S=4472μg/mL)in c.mparison with ZLM-66(SI=2019.80,S=1.9μg/mL).This new NNRTI c.nferred low nanomolar inhibition of wild-type HIV-1 strain and tested mutant strains(K103N,L1001,Y181c.E138K,and K103N+Y181c..The analog also demonstrated favorable safety and pharmac.kinetic.profiles,as evidenc.d by its insensitivity to c.P and hERG,lac. of mortality and pathologic.l damage,and good oral bioavailability in rats(F=27.1%).Further development of 7w for HIV therapy will be fac.litated by this valuable information.

In order to improve the positional adaptability of our previously reported naphthyl diaryltriazines(NP-DATAs),synthesis of a series of novel biphenyl-substituted diaryltriazines(BP-DATAs)with a flexible side c.ain attac.ed at the c.6 position is *** c.mpounds exhibited exc.llent potenc. against wild-type(WT)HIV-1 with Ec.0 values ranging from 2.6 to 39 nmol/L and most of them showed low nanomolar anti-viral potenc. against a panel of HIV-1 mutant *** 5 j and 6 k had the best ac.ivity against WT,single and double HIV-1 mutants and reverse transc.iptase(RT)enzyme c.mparable to two referenc. drugs(EFV and ETR)and our lead c.mpound NP-DATA(1).Molec.lar modeling disc.osed that the side c.ain at the c.6 position of DATAs oc.upied the entranc. c.annel of the HIV-1 reverse transc.iptase non-nuc.eoside binding poc.et(NNIBP)attributing to the improved *** preliminary struc.ure-ac.ivity relationship and PK profiles were also disc.ssed.
