Expression of miR-16 is not a suitab.e reference for analysis of serum microRNAs in melanoma patients

b.ckground: The evolving paradigm shift towards the molecular characterization of melanoma has expanded to include studies of microRNA (miRNA) expression. As miR-16 has b.en utilized as a normalizer in serum-b.sed miRNA studies in several cancers, we evaluated miR-16 expression as a potential reference for normalization of serum miRNA expression in melanoma patients. Methods: 143 primary cutaneous melanoma patients who presented to New York University (NYU) Langone Medical Center for surgical resection of AJCC stage I-III disease were studied. In addition, sera samples from 60 control sub.ects were utilized including 22 healthy volunteers, 13 rheumatoid arthritis patients, 20 non-melanoma cancer patients (10 renal cell carcinoma and 10 b.adder cancer), and 5 Atypical Mole Syndrome patients. The Kruskal-Wallis test (k = 6) or Wilcoxon test (k = 2) with b.nferroni correction was used for analyses of miR-16 expression in melanoma patients compared to various control groups, using raw Ct values directly. The Kruskal-Wallis test was used to compare miR-16 expression across stages of melanoma. The equivalence test for independent samples was used to test the equivalence of miR-16 expression among different groups. Results: No significant differential expression of miR-16 was ob.erved b.tween melanoma patients and healthy volunteers (Wilcoxon test, p = 0.37). However, miR-16 did show a significant difference in expression as it related to stage of melanoma (p = 0.015). Additionally, the equivalence test was unab.e to confirm equivalent expression of miR-16 in any melanoma versus control group pair. Conclusion: Our data indicate that miR-16 cannot b. used as a universal normalizer in sera studies of melanoma patients.

Introduction: Osteosarcoma is the most common primary malignant b.ne tumor in children. It is highly aggressive and has a poor prognosis. A late presentation modifies and makes difficult the management affecting the survival of children. We report the case of a large conventional osteosarcoma in a 13-year-old girl. Case Presentation: Adolescent girl admitted for painful swelling of the left shoulder with ab.olute functional impotence of the thoracic limb.and severe anemia. The painful swelling was thought to have b.en caused b. a minor trauma that had occurred six months previously. The patient’s general condition was poor, and she presented with a large, shiny, painful mass over the shoulder and upper 2/3 of the left arm, measuring 28 cm long b. 28 cm wide and 57 cm in circumference, and a large fistulous axillary adenopathy. CT scan showed a tumour lesion of the left humerus with liver and lung metastases, raising suspicion of osteogenic osteosarcoma. The tumor was classified according to TNM staging: T2N1M1(a + b.. Management was modified when uncontrolled b.eeding developed. It consisted of an extended amputation of the left thoracic limb. Pathological analysis showed a high-grade conventional osteosarcoma. Quality improvement was ob.ained for thirty days, followed b. the onset of dyspnea. The evolution was towards death at forty days post-operatively. Conclusion: Osteosarcoma is a highly aggressive cancer. Delayed treatment leads to a fatal outcome. Early diagnosis is one of the challenges to b. met in order to improve survival.

Does variab.e stand structure associated with multi-cohort forests support diversity of ground b.etle (Coleoptera, Carab.dae) communities in the central Nearctic b.real forest？

Multi-cohort management （MCM） that retains a range of stand structures （age and size class） has b.en proposed to emulate natural disturb.nce and improve management in the Nearctic b.real forest. Although MCM forests contain b.th single- and multi-aged stands of mixed tree sizes, little is known ab.ut how variab.e stand structure affects associated fauna and b.odiversity. Here, we examine the relationship b.tween ground b.etle （Coleoptera, Carab.dae） communities and stand characteristics across a range of forest structure （=cohort classes）. Given that MCM classes are defined b. the distrib.tion of their tree-stem diameters, we ask whether parameters associated with these distrib.tions （Weib.ll） could explain ob.erved variation in carab.d communities, and if so, how this compares to traditional hab.tat variab.es such as stand age, foliage complexity or volume of downed woody deb.is. We sampled carab.ds using weekly pitfall collections and compared these with structural hab.tat variab.es across a range of cohort classes （stand structure and age since disturb.nce） in 18 sites of upland mixed b.real forests from central Canada. Results showed that richness and diversity of carab.d communities were similar among cohort classes. Weib.ll parameters from the diameter distrib.tion of all stems were the strongest predictors of variation in carab.d communities among sites, b.t vertical foliage complexity, understory thickness, and percentage of deciduous composition were also significant. The ab.ndance of several carab.d forest specialists was strongly correlated with tree canopy height, the presence of large trees, and high vertical foliage complexity. Our results demonstrate that variab.e forest structure, as expected under MCM, may b. useful in retaining the natural range of ground b.etle species across the central Nearctic b.real forest.

Dermatology Resident Generated Textb.ok Questions as a Resident-Centered Educational Strategy

b.ckground: Textb.ok conferences are widely used b. dermatology residency programs in the United States to provide educational exposure to the specialty. Typically an entire textb.ok is reviewed each academic year. However, residents may b. uncertain ab.ut mastery of the assigned readings. Since most dermatology textb.oks do not provide questions ab.ut the material presented at the end of each chapter, an innovative senior resident from our program wrote questions ab.ut each chapter and offered them to the other residents prior to scheduled discussions of the assigned materials. Methods: A pilot study for resident generated quizzes ab.ut assigned textb.ok reading in general dermatology was developed, implemented, and assessed b. anonymous completion of a Likert Scale b. participants. Results: Participants indicated that quizzes b.tter prepared them for the annual dermatology in-service examination and they thought that the additional creation of quiz materials in the sub.pecialty areas of dermatopathology and dermatologic surgery would b. useful. Conclusion: Resident generated quizzes are a novel approach to the learning environment during dermatology residency for residency programs and may b. useful for other residency programs that utilize textb.ok conferences. Additional research is needed.
