Immunoexpression of Cathepsin D and S100A4 Protein and Their Molecular Subtyptes in Canine Mammary Carcinomas

Cathepsin D (CD), a lysosomal protease, and S100A4 protein, a calcium binding motif, are considered to be involved in metastasis in various human cancers. No data regarding such proteins are available for canine mammary carcinomas (CMCs). Accordingly, their expression in association with known factors of prognosis was investigated in this study. For that, 66 surgically resected CMCs were submitted to an immunohistochemical evaluation using anti CD, S100A4 protein, HER2, estrogen receptor α, cytokeratin 5, and p63 antibodies, further characterizing the tumors' molecular subtype. An increase in S100A4 immunoexpression by neoplastic luminal mammary cells was associated with an infiltrative tumor mode of growth, consequently leading us to conclude that S100A4 protein could be related to progression in CMCs. Additionally, the occurrence of the luminal A molecular subtype was associated with the complex histotype in CMCs. Although we have demonstrated that changes in S100A4 protein immunoexpression occurs in CMCs, further studies are needed to determine whether this represents important independent biomarkers for CMCs.

Reaction of Oxidized CuZnSOD with Polyphenols

Several polyphenolic compounds, including twelve flavonols and a variety of lower molecular weight compounds were found to diminish the effects of CuZnSOD on NMR relaxation of fluoride ion in a pH and concentration dependent manner. While we originally thought the effect arose from binding of the compounds to the enzyme active site, several lines of evidence indicated that active polyphenols reduced the paramagnetic copper(II) form of the enzyme to the diamagnetic copper(I) form, thereby giving false positive indications of enzyme inhibition in the NMR assay. First, docking experiments failed to provide a satisfactory explanation of the SAR. Second, effects on the enzyme’s EPR spectrum indicated that catechols could bind the active site copper leading to enzyme reduction. Third, while these reactions did not proceed to completion in aerobic solution, they did so under inert atmosphere. Fourth, experiments employing superoxide producing compounds demonstrated that loss of NMR activity did not prevent the enzyme from redox cycling. Thus, while the polyphenols appeared to inhibit the enzyme in the NMR assay, the compounds did not inhibit the enzyme’s reactions with superoxide.

Objective: To identify risk factors associated with transitions from, cognitively normal to various forms of mild cognitive impairment (MCI) and then from MCI into early dementia with death as a competing state. Methods: Cognitive assessments from 554 subjects participating in a longitudinal study at the University of Kentucky AD Center were used to classify individuals into one of three transient states at any visit: cognitively normal, amnestic MCI, or mixed MCI. Between visits subjects could die or become demented. A series of polytomous logistic models were used to model transitions among these states over time and to determine how the log odds of these transitions vary with age, education, sex, family history of dementia, and APOE status. Results: Age affects all transitions among transient states as well as those to dementia or death. Presence of at least one apolipoprotein 4 allele affects transitions from cognitively normal into amnestic MCI or into dementia. At most 12 years of education affects transitions into mixed MCI. Transitions do not vary with sex or family history. Conclusion: Aside from age, the usual risk factors associated with conversion from cognitively normal into dementia are likely risk factors for transitions into mild cognitive impairment.

Comparison of Different Approaches to Force Controlled Precision Honing of Bores

Honing is a machining process which can economically produce exact bores regarding form, geometry and surface quality. During the honing process, the tool, equipped with one or several abrasive honing stones, combines three movement components: rotation, oscillation in axial direction and radial feed movement of the honing stone. The feed movement is a decisive factor for the results of the honing process and can be controlled either by a gradual lining in fixed time intervals or by the regulation of the occurring forces. The presented studies show different approaches for the regulation of force controlled honing and their effects on the measured forces, the torque and the quality parameters of the process.

Background: Data from neuropathologic studies of the frequency of Alzheimer disease (AD) among African American persons conflict as to whether the neuropathologic phenotype of AD is identical in African American and white persons. Objectives: To examine clinical and neuropathologic phenotypes of AD in African American individuals and to compare AD and vascular burdens between African American and white persons. Design, Setting, and Patients: Ten African American decedents who underwent brain autopsy at the Washington University Alzheimer's Disease Research Center were matched for age, sex, and Clinical Dementia Rating with 10 white decedents between January 1, 1990, and January 1, 2000. The presence and degree of neurofibrillary tangles, senile plaques, Lewy bodies, cerebral infarcts, and cerebral amyloid angiopathy were determined. Results: All 20 individuals had a neuropathologic diagnosis of AD. There were no group differences in the presence or number of infarcts, plaques, tangles, Lewy bodies, or amyloid angiopathy. Conclusion: In this small sample, we found no substantive differences in the neuropathology of AD among African American and white individuals.
