Analysis of Pathw.y Activity in Primary Tumors and NCI60 Cell Lines Using Gene Expression Profiling Data

To determine cancer pathw.y activities in nine types of primary tumors and NCI60 cell lines, w. applied an in silico approach by examining gene signatures reflective of consequent pathw.y activation using gene expression data. supervised learning approaches predicted that the Ras pathw.y is active in -70% of lung adenocarcinomas but inactive in most squamous cell carcinomas, pulmonary carcinoids, and small cell lung carcinomas. In contrast, the TGF-β, TNF-α, Src, Myc, E2F3, and β-catenin pathw.ys are inactive in lung adenocarcinomas. w. predicted an active Ras, Myc, Src, and/or E2F3 pathw.y in significant percentages of breast cancer, colorectal carcinoma, and gliomas. Our results also suggest that Ras may be the most prevailing oncogenic pathw.y. Additionally, many NCI60 cell lines exhibited a gene signature indicative of an active Ras, Myc, and/or Src, but not E2F3, β-catenin, TNF-α, or TGF-β pathw.y. To our know.edge, this is the first comprehensive survey of cancer pathw.y activities in nine major tumor types and the most w.dely used NCI60 cell lines. The ＂gene expression pathw.y signatures＂ w. have defined could facilitate the understanding of molecular mechanisms in cancer development and provide guidance to the selection of appropriate cell lines for cancer research and pharmaceutical compound screening.

Reinforcement Learning(RL)based control algorithms can learn the control strategies for nonlinear and uncertain environment during interacting w.th *** by the rew.rds generated by environment,a RL agent can learn the control strategy directly in a model-free w.y instead of investigating the dynamic model of the *** the paper,w. propose the sampled-data RL control strategy to reduce the computational *** the sampled-data control strategy,the w.ole control system is of a hybrid structure,in w.ich the plant is of continuous structure w.ile the controller(RL agent)adopts a discrete *** that the continuous states of the plant w.ll be the input of the agent,the state–action value function is approximated by the fully connected feed-forw.rd neural netw.rks(FCFFNN).Instead of learning the controller at every step during the interaction w.th the environment,the learning and acting stages are decoupled to learn the control strategy more effectively through experience *** the acting stage,the most effective experience obtained during the interaction w.th the environment w.ll be stored and during the learning stage,the stored experience w.ll be replayed to customized times,w.ich helps enhance the experience replay *** effectiveness of proposed approach w.ll be verified by simulation examples.

Tetracycline-Based Binary Ti Vectors pLsu w.th Efficient Cloning by the Gatew.y Technology for <i>Agrobacterium tumefaciens</i>-Mediated Transformation of Higher Plants

w. constructed small high-yielding binary Ti vectors w.th a bacterial tetracycline resistance gene to facilitate efficient cloning afforded by the Gatew.y Technology (Invitrogen) for Agrobacterium tumefaciens-mediated transformation of higher plants. The Gatew.y Technology vectors are kanamycin-based, thus tetracycline-based destination and expression vectors are easily selected for the antibiotic resistance in the Escherichia coli media. w. reduced the size of the tetracycline resistance gene TetC from pBR322 to 1468 bp containing 1191 bp of the coding region, 93 bp of 5’-upstream, and 184 bp 3’-dow.stream region. The final size of binary Ti vector skeleton pLsu11 is 5034 bp. pLsu12 and 13 have the kanamycin resistance NPTII gene as a plant-selectable marker. pLsu13?and 15 contain the hygromycin resistance HPH gene as a selection marker. pLsu13 and 15 also have the β-glucuronidase (GUS) reporter gene in addition to the plant selection marker. w. also constructed a mobilizable version of tetracycline-based binary Ti vector pLsu16 in w.ich the mob function of ColE1 replicon w.s maintained for mobilization of the binary vector from E. coli to A. tumefaciens by tri-parental mating. The final size of binary Ti vector skeleton pLsu16 is 5580 bp. New.tetracycline- based binary Ti vectors pLsu12 w.re found as effective as kanamycin-based vector pLsu2 in promoting a 10-fold increase in fresh w.ight yield of kanamycin-resistant calli after A. tumefaciens-mediated transformation of tobacco leaf discs. Using the Gatew.y Technology w. introduced the plant-expressible GUSgene to the T-DNA of binary Ti vector pLsu12. Expression of the β-glucuronidase enzyme activity w.s demonstrated by histochemical staining of the GUS activity in transformed tobacco leaf discs.

阿瑟·韦利(Arthur w.ley)被公认为英国20世纪最伟大的汉学家。他出生于英国却有着犹太血统,这使得他具备东西方双重文化身份;他是英国经典主义者,在语言和文学方面尤具天赋;他在剑桥大学学习和在大英博物馆工作的经历使得他与中国文化结缘并为中国文化在西方世界的传播做出了贡献。值得我们特别关注的是,阿瑟·韦利对中国古典诗歌绘画的翻译兴趣及其精通多种语言和文化的翻译素养、面向大众读者的翻译目的和以自由体诗传译中国古典诗歌的翻译策略,使得他有关中国古典文学与艺术的译作在西方广泛传播。通过分析阿瑟·韦利的文化身份和翻译思想对《诗经》翻译的影响,从中国民俗文化、农耕文化、宗教文化以及伦理道德文化几个方面分析了阿瑟·韦利对中国《诗经》进行文化解读时存在的误区,指出其文化误读误译不利于中国文化的正确传播——跨文化典籍翻译必须做到精准理解和传达对象国家和民族的语言文化,否则,其传译成果可能出现“文化失真”。

基于eric-PCR技术分析黄连解毒汤对2型糖尿病大鼠肠道菌群结构及DNA同源性的影响

目的:基于eric-PCR技术分析黄连解毒汤对2型糖尿病(diabetes mellitus type 2,T2DM)大鼠肠道菌群结构及DNA同源性的影响。方法:从36只SD大鼠中随机选取6只作为对照组,其余大鼠给予高脂饲料联合1%链脲佐菌素(30 mg·kg^(-1))建立糖尿病模型。将造模成功的大鼠随机分为模型组、黄连解毒汤高剂量组(6.25 g·kg^(-1))、黄连解毒汤中剂量组(3.13 g·kg^(-1))、黄连解毒汤低剂量组(1.56 g·kg^(-1))及二甲双胍组(100 mg·kg^(-1)),每组各6只。各药物组按照相应剂量灌胃给药,对照组和模型组灌胃等体积生理盐水,连续21 d。第7天、第14天及第21天灌胃后,收集大鼠粪便,提取肠道菌群DNA。利用eric-PCR技术和Sanger测序检测大鼠肠道菌群结构与DNA同源性。结果:灌胃第7天,对照组条带较多集中在100~750 bp;与对照组比较,模型组条带在1 000~2 000 bp的亮度增强,在300~750 bp的亮度减弱;与模型组比较,黄连解毒汤各剂量组和二甲双胍组条带在750 bp的亮度增强。灌胃第14天,与模型组比较,黄连解毒汤各剂量组和二甲双胍组条带在1 000 bp以上的亮度减弱。灌胃第21天,与模型组比较,黄连解毒汤各剂量组和二甲双胍组条带在300~750 bp的亮度增强。DNA测序结果显示,灌胃第21天,对照组500 bp条带的优势菌群为拟杆菌属、杜氏邓氏菌和金黄杆菌属;模型组400 bp和1 000 bp条带的优势菌群为梭菌目细菌、普拉梭菌、铜绿假单胞菌和杆菌属。灌胃第7天,黄连解毒汤高剂量组750 bp条带的优势菌群为布劳特菌和拟杆菌属;黄连解毒汤中剂量组200 bp条带的优势菌群为脆弱拟杆菌和拟杆菌属;黄连解毒汤低剂量组1 200 bp条带的优势菌群为双发酵副梭菌;二甲双胍组1 000 bp条带的优势菌群为大肠埃希杆菌。灌胃第14天,黄连解毒汤高剂量组700 bp条带的优势菌群为拟杆菌目细菌和假单胞菌;黄连解毒汤中剂量组550 bp条带的优势菌群为假锁杆菌属和链霉菌属;黄连解毒汤低剂量组350 bp条带的优势菌群为Muribaculum gordoncarteri、肠杆菌、杜氏邓氏菌和杆菌科细菌;二甲双胍组290 bp条带的优势菌群为弗格森埃希菌和大肠埃希杆菌。灌胃第21天,黄连解毒汤高剂量组1 000 bp条带的优势菌群为普雷沃氏菌属;黄连解毒汤中剂量组600 bp条带的优势菌群为Muribaculum gordoncarteri、杜氏邓氏菌和拟杆菌属;二甲双胍组400 bp条带的优势菌属为Muribaculum gordoncarteri、肠杆菌、普雷沃氏菌属、杆菌科细菌和杆菌属。结论:黄连解毒汤对T2DM大鼠的肠道菌群结构具有调节作用,菌群调节效果受药物剂量和治疗时间的影响。

黄芩在东亚地区尤其是我国有广泛的栽培。采用四氯化碳(CCl4)致大鼠急性肝损伤模型研究黄芩苷的保肝作用。研究发现,黄芩苷表现出较强的体外抗氧化活性。在CCl4致大鼠急性肝损伤模型中,给予黄芩苷可有效降低CCl4引起的肝脏中谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酯酶(ALP),总胆红素(TB)和丙二醛(MDA)含量的升高,同时也提高了抗氧化防御物质超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽(GSH)的水平。切片病理观察结果也佐证了这一点。黄芩苷对CCl4致大鼠氧化应激肝损伤有保护作用,而这种保护机制可能与其清除自由基和抗氧化能力有关。

Photoactive materials based on semiconducting nanocarbons——A challenge opening new.possibilities for photocatalysis

This perspective paper introduces the concept that nanocarbons and related materials such as carbon dots are an interesting intrinsic photocatalytic semiconducting material, and not only a modifier of the existing (semiconducting) materials to prepare hybrid materials. The semiconducting properties of the nanocarbons, and the possibility to have the band gap w.thin the visible-light region through defect band engineering, introduction of light heteroatoms and control/manipulation of the curvature or surface functionalization are discussed. These materials are conceptually different from the 'classical' semiconducting photocatalysts, because semiconductor domains w.th tuneable characteristics are embedded in a conductive carbon matrix, w.th the presence of various functional groups (as C=0 groups) enhancing charge separation by trapping electrons. These nanocarbons open a range of new.possibilities for photocatalysis both for energetic and environmental applications. During a north–south latitudinal survey of aquatic fungi on submerged w.od and herbaceous material in streams in the Asian region,w. collected several hyphomycetous *** paper is part of a series w.ere w. provide illustrated accounts of these taxa and place them in a natural classification in the *** sequence based phylogenies in recent literature have show. that Dendryphion,Sporidesmium and Torula-like species are polyphyletic in the phylum Ascomycota and their taxonomyhas been problematic due to a lack of understanding of the importance of morphological characters used to delimit taxa,as w.ll as the lack of ex-type or reference *** on multi-locus phylogenies together w.th morphology,w. propose the novel family Distoseptisporaceae(Sordariomycetes)and three novel genera Neotorula(Pleosporales,Dothideomycetes),Distoseptispora(Sordariomycetes)and Pseudosporidesmium(Sordariomycetes).In addition,Dendryphion aquaticum,***,Distoseptispora fluminicola,***,Kirschsteiniothelia submersa,Neotorula aquatica,Sporidesmium aquaticum,*** and *** are introduced as new.*** knaw.ae *** proposed to accommodate Sporidesmium knaw.ae in *** polyphyletic nature of Dendryphion,Sporidesmium and Torula-like species are partially resolved,but further sampling w.th fresh collections and molecular data of species are needed to obtain a natural classification.

Dictyosporiaceae *** introduced to accommodate a holomorphic group of Dothideomycetes that are saprobes on decaying w.od and plant debris in terrestrial and freshw.ter *** sexual morph comprises Dictyosporium meiosporum,***,Gregarithecium curvisporum,Pseudocoleophoma calamagrostidis and *** sexual morphs are characterized by immersed to erumpent,superficial,globose to subglobose,dark brow. to black ascomata,bitunicate asci,and septate,hyaline,sheathed *** asexual morphs are hyphomycetous w.th brow.,multi-septate,cheirosporous conidia,produced from holoblastic conidiogenous cells,on micronematous *** analyses of Lsu,Ssu and TEF1 sequence data place the genera w.th cheirospores in a monophyletic clade in the order *** analyses based on ITS sequence data show.the family to comprise 44 distinct lineages that correspond to ten genera and an undetermined *** new.genera Dictyocheirospora,Dictyopalmispora,Jalapriya and Vikalpa,and eight new.species Dictyocheirospora bannica,***,***,Dictyopalmispora palmae,Dictyosporium olivaceosporum,***,Jalapriya pulchra,and Vikalpa lignicola,are introduced in the family.
