Acute lymphoblastic leukemia (ALL) is the most common malignancy in children, with the majority of cases being of precursor B-cell phenoltype. Conventional cytogenetic analysis plays an important role in the diagnosis of B-cell ALL, identifying characteristic chromosomal abnormalities associated with a given prognosis therein facilitating optimized treatment. The more recent introduction of microarray technology to the analysis of B-cell ALL has afforded both higher resolution for the detection of known abnormalities and an ability to identify novel copy number abnormalities (CNAs) with potential clinical relevance. In the current study, microarray analysis was performed on 20 cytogenetically abnormal B-cell ALL cases (10 pediatric and 10 adult), while a novel microarray-based balanced-translocation detection methodology (translocation CgH or tCgH) was applied to that subset of cases with a known or suspected recurrent balanced translocation. Standard microarray analysis identified that CNAs was not detected by previous conventional cytogenetics in 75% (15/20) cases. tCgH identified 9/9 (100%) balanced translocations defining BCR/ABL1 (x4), ETV6/RUNX1 (x3), and MLL/AFF1 (x2) breakpoints with high resolution. The results illustrate the improved molecular detail afforded by these technologies and a comparison of translocation breakpoints, CNAs and patient age offers new insights into tumor biology with potential prognostic significance.

国际糖尿病足工作组(IWgDF)从1999年开始发布糖尿病足病防治指南。本指南是有关糖尿病足感染的诊断和治疗的指南,是在2015年糖尿病足感染指南基础上更新的。糖尿病足感染指南编辑委员会基于患者-干预-比较-结局(PICO)原则,整合内部与外部专家顾问的建议,并对2016年以后有关糖尿病足感染和处治的文献进行系统评价后,提出了27条推荐要点。这些推荐要点涵盖了软组织与骨组织感染的诊断,包括感染的严重性分级。这是IWgDF15年以来第一次对这个分级表进行更新。此外,指南还评估了糖尿病足感染的微生物学,包括如何收集标本及如何识别致病菌。最后,指南讨论了治疗糖尿病足感染的方法,包括经验性抗生素的选择、软组织和骨组织感染的抗微生物治疗、何时和如何进行外科处理以及各种辅助治疗对糖尿病足感染的有效性。本版指南在原指南的基础上更新了4个表格和1个图。我们认为该指南可以为糖尿病足感染的临床诊断和治疗提供更好的帮助。

Thermodynamics of the Second Stage Dissociation Step (pK<sub>2</sub>) of Buffer Monosodium 1,4-Piperazinediethanesulfonate from (278.15 to 328.15) K

Values of the second thermodynamic dissociation constant pK2 of the protonated form of monosodium 1,4-piperazinediethanesulfonate (PIPES) have been determined at twelve different temperatures in the temperature range from (278.15 to 328.15) K including the body temperature 310.15 K by measurement of the electromotive-force for cells without liquid junction of the type: Pt (s), H2 (g), 101.325 kPa|Na-PIPES (m1) + Na 2-PIPES (m2) + NaCl (m3)|AgCl (s), Ag (s), where m1, m2 and m3 indicate the molalities of the corresponding species at 1 atm = 101.325 kPa in SI units. The pK2 values for the dissociation of Na-PIPES are represented by the equation: pK2 = -1303.76/T + 48.369 - 6.46889 lnT with an uncertainty of ± 0.001. The values of pK2 for Na-PIPES were found to be 7.1399 ± 0.0004 at 298.15 K and 7.0512 ± 0.0004 at 310.15 K, respectively, and indicate that this buffer would be useful as pH standard in the range of physiological application. Standard thermodynamic quantities for the acidic dissociation process of Na-PIPES have been derived from the temperature coefficients of the pK2. These values are compared with those of structurally related N-substituted PIPERAZINE and TAURINE at 298.15 K.

机械制浆是一个耗电量较大的制浆过程,而且成纸强度较差。生物机械制浆就是在机械制浆前,用菌种对木素纤维素物质进行预处理。实验证明菌种预处理后再进行机械制浆至少可节约30％左右的电能,而且成纸强度也大为改善。为了把生物机械制浆工艺应用到工业生产中,我们采用最优质的生物制浆菌种连续接种50 吨云杉木片,然后把它们置于室外,堆放两个星期。木片堆需要进行强制通风,以保持菌种最优的生长温度和木片堆的水分。随后用对照木片和菌种预处理木片进行热磨机械法制TMP浆,生产低定量涂布纸。与对照木片相比,用菌种预处理的木片进行机械制浆可节约33％的电能,而且成纸的强度特性也大大提高。与传统TMP浆相比,生物机械浆的纤维强度较好。因此,在成纸中至少可减少5％的漂白针叶木硫酸盐浆。但是,用菌种预处理木片却降低了成浆白度,成浆白度的降低可通过漂白时增加过氧化氢用量来得到弥补。如果达到与对照漂白浆相同的白度,那么需要多加60％的过氧化氢。生物机械制浆的经济性很有吸引力。

Magnetotactic bacteria(MTB) are known to produce single-domain magnetite or greigite crystals within intracellular membrane organelles and to navigate along the Earth’s magnetic field lines. MTB have been suggested as being one of the most ancient biomineralizing metabolisms on the Earth and they represent a fundamental model of intracellular biomineralization. Moreover, the determination of their specific crystallographic signature(e.g. structure and morphology) is essential for palaeoenvironmental and ancient-life studies. Yet, the mechanisms of MTB biomineralization remain poorly understood, although this process has been extensively studied in several cultured MTB strains in the Proteobacteria phylum. Here, we show a comprehensive transmission electron microscopy(TEM) study of magnetic and structural properties down to atomic scales on bullet-shaped magnetites produced by the uncultured strain MYR-1 belonging to the Nitrospirae phylum, a deeply branching phylogenetic MTB group. We observed a multiple-step crystal growth of MYR-1 magnetite: initial isotropic growth forming cubo-octahedral particles(less than approx. 40 nm), subsequent anisotropic growth and a systematic final elongation along [001] *** the crystal growth, one major f111 g face is well developed and preserved at the larger basal end of the crystal. The basal f111 g face appears to be terminated by a tetrahedral – octahedral-mixed iron surface, suggesting dimensional advantages for binding protein(s), which may template the crystallization of magnetite. This study offers new insights for understanding magnetite biomineralization within the Nitrospirae phylum.

Abdominal aortic aneurysm(AAA)is a progressive aortic dilatation,causing~80%mortality upon ***,there is no approved drug therapy for *** repairs are invasive and risky and thus not recommended to patients with small AAAs which,however,account for~90%of the newly diagnosed *** is therefore a compelling unmet clinical need to discover effective non-invasive strategies to prevent or slow down AAA *** contend that the first AAA drug therapy will only arise through discoveries of both effective drug targets and innovative delivery *** is substantial evidence that degenerative smooth muscle cells(SMCs)orchestrate AAA pathogenesis and *** this study,we made an exciting finding that PERK,the endoplasmic reticulum(ER)stress Protein Kinase R-like ER Kinase,is a potent driver of SMC degeneration and hence a potential therapeutic ***,local knockdown of PERK in elastase-challenged aorta significantly attenuated AAA lesions in *** parallel,we also conceived a biomimetic nanocluster(NC)design uniquely tailored to AAA-targeting drug *** NC demonstrated excellent AAA homing via a platelet-derived biomembrane coating;and when loaded with a selective PERK inhibitor(PERKi,gSK2656157),the NC therapy conferred remarkable benefits in both preventing aneurysm development and halting the progression of pre-existing aneurysmal lesions in two distinct rodent models of *** summary,our current study not only establishes a new intervention target for mitigating SMC degeneration and aneurysmal pathogenesis,but also provides a powerful tool to facilitate the development of effective drug therapy of AAA.

Searching for the neutrinoless double beta decay(NLDBD)is now regarded as the topmost promising technique to explore the nature of neutrinos after the discovery of neutrino masses in oscillation *** X-III(particle and astrophysical xenon experiment III)will search for the NLDBD of136Xe at the China jin Ping Underground Laboratory(CjPL).In the first phase of the experiment,a high pressure gas Time Projection Chamber(TPC)will contain 200 kg,90%136Xe enriched gas operated at10 *** pitch micro-pattern gas detector(Microbulk Micromegas)will be used at both ends of the TPC for the charge readout with a cathode in the *** signals can be used to reconstruct the electron tracks of the NLDBD events and provide good energy and spatial *** detector will be immersed in a large water tank to ensure～5 m of water shielding in all *** second phase,a ton-scale experiment,will consist of five TPCs in the same water tank,with improved energy resolution and better control over backgrounds.
