斑石鲷肾脏细胞系的建立、特征分析及其免疫相关基因的表达分析

斑石鲷(Ople.nathuspunctatus)是我国重要的海水养殖鱼类,本研究以斑石鲷肾脏组织为材料,建立了斑石鲷肾脏组织细胞系(O. punctatus kidne. ce.l line. OPK)。斑石鲷肾脏细胞系生长旺盛,目前已成功传至34代。本研究检测了不同FBS浓度对斑石鲷肾脏细胞生长的影响,结果表明最适生长FBS浓度为20%。将OPK细胞液氮冷冻复苏后,细胞具有活性,可正常生长和传代。核型分析结果显示,第24代斑石鲷肾脏细胞系核型为正常二倍体核型(2n=2sm+46t)。将Cy3-siRNA转染到OPK细胞后,可以成功表达荧光。对斑石鲷OPK细胞提取DNA,用斑石鲷线粒体色素细胞C氧化酶I (CO I)基因进行检测,结果表明该细胞系来源于斑石鲷。斑石鲷肾脏细胞系细胞在受到脂多糖(LPS)和聚肌胞苷酸(poly I∶C)刺激后能产生免疫反应,免疫相关基因IκB、IL-1β、IL-8和IRF3的表达水平发生显著变化。本研究成功建立了斑石鲷肾脏细胞系,可运用于斑石鲷基因功能分析、细胞遗传学、致病性细菌和病毒感染机制等研究,可为斑石鲷的基础研究和细胞工程育种等提供重要的基础材料。

为了探讨小鼠卵巢颗粒细胞体外生长的特点及增殖的调控因素,分别添加2μg/mL胰岛素、50ng/mL FSH和20 ng/mL e.F的培养液培养小鼠卵巢颗粒细胞,每隔24 h对细胞进行计数,研究其对小鼠卵巢颗粒细胞增殖的影响。此外,还对培养过程中出现的卵巢颗粒细胞集落进行了AKP染色和C-kit免疫组化染色鉴定。结果显示,在培养的第4天添加e.F组和FSH组与对照组相比均具有显著的促增殖作用(P0.05);卵巢颗粒细胞集落经AKP染色和C-kit免疫组化染色均呈阳性。提示e.F和FSH对小鼠卵巢颗粒细胞具有促增殖效应,小鼠卵巢颗粒细胞具有干细胞的部分特性。

采用常规压片法对甘蓝类作物的7个变种进行了核型分析和比较。结果表明:青花菜2n=2x=18=8m+10sm(2SAT),随体位于第6对染色体上;结球甘蓝2n=2x=18=8m+10sm(2SAT),随体位于第6对染色体上;花椰菜2n=2x=18=12m+6sm(2SAT),随体位于第7对染色体上;芥蓝2n=2x=18=8m+10sm(2SAT),随体位于第7对染色体上;抱子甘蓝2n=2x=18=8m+10sm(2SAT),随体位于第7对染色体上(首次报道);羽衣甘蓝2n=2x=18=10m+8sm;球茎甘蓝2n=2x=18=8m+10sm,羽衣甘蓝和球茎甘蓝未观察到随体染色体。7个变种核型分类均属于2A型,为基本对称核型,但不同变种间的进化程度存在不同程度的差异。此外,对甘蓝类作物的亲缘关系及遗传多样性也进行了探讨。

目的:探讨超声引导下真空辅助活检(vacuum-assiste. biopsy,VAB)切除乳腺癌肿块后保乳手术的可行性与安全性。方法:选择2008年1月至2014年12月VAB肿块切除术后行保乳手术治疗的45例浸润性导管癌(invasive.ductal carcinoma,IDC)患者(VAB组)。选取同一时期单发、直径小于2cm、肿块切除活检后行保乳手术的147例IDC患者为对照组。分析并比较两组患者的临床病理特征与预后。结果:VAB组肿块均单发、直径e.se.fre. survival,DFS)和5年总生存期(ove.all survival,OS)差异均无统计学意义。结论:VAB切除不可触及的较小乳腺癌肿块后续行保乳手术安全可行,值得进一步大样本研究。
