Cardiovascular disease remains the top threat to human *** the prevalence and mortality of cardiovascular diseases have been persistently increasing in China,the cardiovascular disease becomes the leading cause of death,accounting for nearly half of all deaths in China[1].In order to improve the situation,many basic cardiovascular studies have been conducted and have obtained great achievement,which is fundamental to the future advance in clinical practice[2].In the current issue,several excellent cardiovascular scientists summarize the advance of basic cardiovascular research from different perspectives,from particular critical molecules to cellular function and animal models,as well as the advanced *** reviews collaboratively bring us an overall vision of the frontiers and trends of cardiovascular research.

The biased ligands in G protein-coupled receptors(GPCRs)have opened new avenues for developing safer and more effective ***,the identification of such biased ligands as drug candidates is highly ***,we report that Higenamine,a compound isolated from a Chinese herb,functions as a novel β-arrestin-biased ligand of the β_(2)-adrenergic receptor(β_(2)-AR).The radioligand binding assays demonstrated that Higenamine was the ligand of β_(2)-*** induced phosphorylation of extracellular signal-regulated kinase 1/2(ERK1/2),which can be blocked by propranolol,an inhibitor of β_(2)-*** Gi protein inhibitor,pertussis toxin,had no effect on the phosphorylation of ERK1/2 induced by ***,Higenamine induced ERK1/2 phosphorylation through transactivation of Epithelial growth factor receptor(EGFR).We also found that Higenamine-induced-ERK1/2 phosphorylation is dependent on β-arrestin1/2,and HG inhibits Doxorubicin-induced cardiomyocyte *** results identify Higenamine as a novel biased ligand via the β-arrestin-dependent *** findings give us a better understanding of Higenamine’s potential role in designing diagnostic and therapeutic strategies.

Population aging has emerged as one of the most pressing global demographic *** population aging is being caused by a sharp drop in fertility rates over time,as well as a significant increase in life expectancy in the past *** World Health Organization(WHO)estimates that the proportion of those aged 60 years and older in the world’s population will almost double from approximately 12%in 2015 to 22%in 2050,with an absolute increase of 900 million to a total of 2 billion older adults(1).At the same time,the burden of global disease has changed greatly and the main diseases affecting human health have switched from acute and chronic infectious diseases to chronic non-communicable diseases(NCDs)(2).Such changing demographics and disease spectrum will profoundly affect all aspects of human society.

Purpose:Given the information overload of scientific literature,there is an increasing need for computable biomedical knowledge buried in free *** study aimed to develop a novel approach to extracting and measuring uncertain biomedical knowledge from scientific ***/methodology/approach:Taking cardiovascular research publications in China as a sample,we extracted subject-predicate-object triples(SPO triples)as knowledge units and unknown/hedging/conflicting uncertainties as the knowledge *** introduced information entropy(IE)as potential metric to quantify the uncertainty of epistemic status of scientific knowledge represented at subject-object pairs(SO pairs)***:The results indicated an extraordinary growth of cardiovascular publications in China while only a modest growth of the novel SPO *** evaluating the uncertainty of biomedical knowledge with IE,we identified the Top 10 SO pairs with highest IE,which implied the epistemic status *** presentation of the SO pairs overlaid with uncertainty provided a comprehensive overview of clusters of biomedical knowledge and contending topics in cardiovascular *** limitations:The current methods didn’t distinguish the specificity and probabilities of uncertainty cue *** number of sentences surrounding a given triple may also influence the value of *** implications:Our approach identified major uncertain knowledge areas such as diagnostic biomarkers,genetic polymorphism and co-existing risk factors related to cardiovascular diseases in *** areas are suggested to be prioritized;new hypotheses need to be verified,while disputes,conflicts,and contradictions need to be ***/value:We provided a novel approach by combining natural language processing and computational linguistics with informetric methods to extract and measure uncertain knowledge from scientific statements.

The Hippo pathway is a newly identified pathway and evolutionarily conserved from flies to humans mainly regulating cell *** co-activator Yes-associated protein(YAP)functions as a major downstream effector and key node of the Hippo *** of YAP is critical to regulate YAP activity and its corresponding functions.β-adrenergic receptor(β-AR),a typical G protein coupled receptor(GPCR),mediates proliferation in various cell types and regulates multiple physical and pathological ***,the role ofβ-AR in regulating YAP remains ***,we report thatβ-AR can obviously stimulate YAP tyrosine *** mechanism is thatβ-AR stimulation results in tyrosine kinase Src activation and Src phosphorylates YAP tyrosine at *** studies demonstrate that inhibition of Src kinase activity can obviously alleviateβ-AR induced YAP tyrosine phosphorylation and cell *** conclude thatβ-AR can induce YAP tyrosine phosphorylation and also establish the Src/YAP pathway as a critical signaling branch downstream of GPCR.
