Edaravone protects Pc12 cells from ischemic-like injury via attenuating the damage to mitochondria

Background： Edaravone had been validated to effectively protect against ischemic injuries. In this study, we investigated the protective effect of edaravone by observing the effects on anti-apoptosis, regulation of Bcl-2/Bax protein expression and recovering from damage to mitochondria after OGD （oxygen-glucose deprivation）-reperfusion. Methods： Viability of Pc 12 cells which were injured at different time of OGD injury, was quantified by measuring MTT （2-（4,5-dimethylthia-zol-2-yl）-2,5-diphenyltetrazolium bromide） staining. In addition, Pc 12 cells＇ viability was also quantified after their preincubation in different concentration of edaravone for 30 min followed by （OGD）. Furthermore, apoptotic population of Pc 12 cells that reinsulted from OGD-reperfusion with or without preincubation with edaravone was determined by flow cytometer analysis, electron microscope and Hoechst/Pl staining. Finally, change of Bcl-2/Bax protein expression was detected by Western blot. Results：（1） The viability of Pc12 cells decreased with time （1-12 h） after OGD. We regarded the model of OGD 2 h, then replacing DMEM （Dulbecco＇s Modified Eagle＇s Medium） for another 24 h as an OGD-reperfusion in this research. Furthermore, most Pc 12 cells were in the state of apoptosis after OGD-reperfusion. （2） The viability of Pc 12 cells preincubated with edaravone at high concentrations （1, 0.1, 0.01 μmol/L） increased significantly with edaravone protecting Pc 12 cells from apoptosis after OGD-reperfusion injury. （3） Furthermore, edaravone attenuates the damage of OGD-reperfusion on mitochondria and regulated Bcl-2/Bax protein imbalance expression after OGD-reperfusion. conclusion： Neuroprotective effects of edaravone on ischemic or other brain injuries may be partly mediated through inhibition of Bcl-2/Bax apoptotic pathways by recovering from the damage of mitochondria.

A highly efficient coupling of glycosyl stannanes and sulfonium salts enabled by synergistic Pd/cu catalysis is disclosed, facilitating the construction of c-aryl/alkenyl glycals under mild conditions in high *** protocol tolerates a wide scope of functional groups including ketone, cyano, ester, amide, nitro,halide. The one-pot formal c–H glycosylation starting from arene is demonstrated with a reaction sequence of dibenzothiophenylation/Stille coupling. Besides, a gram-scale reaction is performed successfully,showing the high applicability of this protocol.

Molecular recognition of aromatic hydrocarbons by four endo-functionalized molecular tubes has been studied by 1H NMR spectroscopy, computational methods, and single crystal X-ray crystallography. The binding selectivity is rationalized by invoking shape complementarity and dipole alignment. The non- covalent inter.ctions are proved to predominantly be c/N-H...π inter.ctions.

BAcKGROUND The main pathophysiological basis of coronavirus disease 2019(cOVID-19)causing respiratory failure is a cytokine storm and interleukin-6(IL-6)is an important component of the cOVID-19 cytokine *** a specific antagonist of IL-6,tocilizumab may block the cytokine storm of *** Diagnosis and Treatment Guidelines of New coronavirus Pneumonia(7 th Edition)includes tocilizumab as a recommended drug for immunotherapy in severe and critical cOVID-19 ***,the specific clinical efficacy of tocilizumab in the treatment of cOVID-19 patients is worth *** To determine the clinical efficacy of tocilizumab in inhibiting the cytokine storm in *** In total,19 severe and critical cOVID-19 patients were enrolled in this study,and were treated with tocilizumab in Optical Valley campus of Hubei Maternal and child Health care Hospital from February 20 to March 31,*** imaging manifestations and clinical data before and after treatment were analyzed retrospectively,including routine peripheral venous blood tests,routine blood biochemical tests,coagulation test,c-reactive protein(cRP),IL-6,and arterial blood gas *** Of the 19 patients in this group,13(68.4%)had significantly improved symptoms of cOVID-19(5 patients were discharged directly and 8 patients were transferred after improvement)following *** case was invalid,1 case was exacerbated,and 4 deaths(21.1%)were observed(all critical cases).The lymphocyte count,cRP,lactic acid,oxygenation index,fibrinogen(FIB)and IL-6 levels were significantly different in the improved *** Tocilizumab treatment is effective against IL-6 in cOVID-19 patients,but it does not completely inhibit the inflammation and cytokine storm in all patients with *** the clinical treatment of cOVID-19 patients,attention should be paid to the timing of drug administration and other adjuvant treatments.

The saturated solubility of carbon and nitrogen in Fe-c-N and Fe-c-B-N melts was measured experimentally at 1 485℃ to obtain the activity inter.ction coefficients between components in these melts. A new method was used to treat experimental results. Using thermodynamic derivation and calculation, some important inter.c- tion coefficients between components in these melts were obtained.

Analysis of χ Phase Precipitation in Aged 2205 Duplex Stainless Steel

Analysis ofZ phase precipitation in 2205 duplex stainless steel aged at 700 and 750 ~c has been investigated systematically. The experimental results showed that X phase forms prior to the precipitation of δ phase and disappears once tr phase starts to precipitate. This phenomenon indicates that a phase nucleated and consumed the Z phase. The δ phase nucleated mairdy at ferrite/austenite interface and grew inwards into the ferrite phase. The morphology of a phase reveals a coral-like structure at the temperature of 700 ℃ for 120 min followed by quenching in water. The decomposition of ferrite occurs via the following eutectoid reaction： F---＊o＇＋y2. The selected area diffraction pattern of zone axes is[3 1 3-][3 1 3], indicating a characteristic orientation relationship between X phase and δ-ferrite.

Using the data obtained from the LGT soil profile, this article attempts to illustrate the process of modem soil formation in the Guanzhong areas and its micromorphological features. The micromorphology is observed under a petrographic microscope, and its image is quantitatively measured by LEIcAL Qwin 2.6 software. Micromorphological observations of the thin sections show that the assemblage of minerals in different horizons is very similar, which is mainly composed of Q and P1. However, there are obvious differences in c/F15μm ratio, mineral content, and coarse features. The pedofeatures is mainly composed of clay, calcite, and amorphous Fe. Ap horizon is characterized by abundant needleshaped secondary calcite, secondary clay, and earthworm fecal pellet. Bc horizon is characterized by a large quantity of secondary calcite with various shapes. Bt1and Bt2 horizons are characterized by abundant clay hypocoatings and a small quantity of secondary calcite. All the results of this research suggest that Earth-cumulic Orthic Anthrosols consist of both the upper Ap horizon, which cause loessal dung and eolian dust deposition, and cultivation occurs simultaneously during the process of Ap horizon-formation, and the lower Bc horizon, which is aeolian sedimentary at the time of relative aridity during late Holocene.

The endoplasmic reticulum(er),which is composed of a continuous network of tubules and sheets,forms the most widely distributed membrane system in eukaryotic *** a result,it engages a variety of organelles by establishing membrane contact sites(McSs).These contacts regulate organelle positioning and remodeling,including fusion and fission,facilitate precise lipid exchange,and couple vital signaling ***,we systematically review recent advances and converging themes on er-involved organellar *** molecular basis,cellular influence,and potential physiological functions for er/nuclear envelope contacts with mitochondria,Golgi,endosomes,lysosomes,lipid droplets,autophagosomes,and plasma membrane are summarized.
