Novel multiplex stool-based assay for the detection of early-stage colon cancer in a Chinese population

PIK3CA and TP53 mutations predict overall survival of stage Ⅱ/Ⅲ colorectal cancer patients

AIM To investigate the predictive value of PIK3 CA and TP53 mutation status in colorectal cancer(CRC) patients treated with 5-fluorouracil-based *** In this study, a total of 315 patients with histologically proven CRC were enrolled from Yangpu Hospital affiliated to Shanghai Tongji University between 2007 and 2011. Of these patients, 241 with stage Ⅱ/Ⅲ CRC received 5-fluorouracil-based adjuvant chemotherapy. Formalin-fixed paraffin-embedded lesion samples of the patients with curatively resected CRC were ***-generation sequencing was performed to identify somatic gene mutations. The correlation of PIK3 CA and TP53 mutation status with overall survival(OS) was analyzed using a Cox proportional hazard model and the Kaplan-Meier *** Among the 241 patients with stage Ⅱ/Ⅲ in this cohort, the PIK3 CA and/or TP53 mutation was detected in 177 patients, among which 54 patients had PIK3 CA and TP53 double mutations. The PIK3 CA or TP53 mutation was not significantly correlated with OS in univariate and multivariate analyses. Compared with patients without PIK3 CA and TP53 mutations, those with double PIK3 CA-TP53 mutations showed a significantly worse survival(univariate HR = 2.21; 95%CI: 1.15-4.24; multivariate HR = 2.02; 95%CI: 1.04-3.91). The PIK3 CA mutation located in the kinase domain showed a trend toward a shorter OS compared with wild-type tumors(multivariate HR = 1.56; 95%CI: 1.00-2.44; P = 0.052). The Kaplan-Meier curve showed that patients harboring the PIK3 CA mutation located in the kinase domain had a worse clinical outcome than those with wild-type status(Log-rank P = 0.041)CONCLUSION Double mutation of PIK3 CA and TP53 is correlated with a shorter OS in stage Ⅱ/Ⅲ CRC patients treated with 5-fluorouracil-based therapy.

BACKGROUND Lymphovascular invasion (LVI) is suggested to be an early and important step in tumor progression toward metastasis, but its prognostic value and genetic mechanisms in colorectal cancer (CRC) have not been well investigated. AIM To investigate the prognostic value of LVI in CRC and identify the associated genomic alterations. METHODS We performed a retrospective analysis of 1219 CRC patients and evaluated the prognostic value of LVI for overall survival by the Kaplan-Meier method and multivariate Cox regression analysis. We also performed an array-based comparative genomic hybridization analysis of 47 fresh CRC samples to examine the genomic alterations associated with LVI. A decision tree model was applied to identify special DNA copy number alterations (DCNAs) for differentiating between CRCs with and without LVI. Functional enrichment and protein-protein interaction network analyses were conducted to explore the potential molecular mechanisms of LVI. RESULTS LVI was detected in 150 (12.3%) of 1219 CRCs, and the presence was positively associated with higher histological grade and advanced tumor stage (both P erval: 1.40-2.25, P er 5-year overall survival rate (P ere identified to be significantly related to LVI (P ere located at 22q, 17q, 10q, and 6q. We further constructed a decision tree classifier including seven special DCNAs, which could distinguish CRCs with LVI from those without it at an accuracy of 95.7%. Functional enrichment and proteinprotein interaction network analyses revealed that the genomic alterations related to LVI were correlated with inflammation, epithelial-mesenchymal transition, angiogenesis, and matrix remodeling. Staged dendrite suppression for high safe and stable lithium-sulfur batteries

The unavoidable dendrite growth and shuttle effect have long been stranglehold challenges limiting the safety and practicality of lithium-sulfur ***,we propose a dual-action strategy to address the lithium dendrite issue in stages by constructing a multifunctional surface-negatively-charged nanodiamond layer with high ductility and robust puncture resistance on polypropylene （PP） *** uniformly loaded compact negative layer can not only significantly enhance electron transmission efficiency and promote uniform lithium deposition,but also reduce the formation of dendrite during early deposition *** importantly,under the strong puncture stress encountered during the deterioration of lithium dendrite growth under limiting current,the high ductility and robust puncture resistance（145.88 MPa） of as-obtained nanodiamond layer can effectively prevent short circuits caused by unavoidable lithium *** Li||Li symmetrical cells assembled with nanodiamond layer modified PP demonstrated a stable cycle of over 1000 h at 2 mA cm-2with a polarization voltage of only 29.3 ***,the negative charged layer serves as a physical barrier blocking lithium polysulfide ions,effectively mitigating capacity *** improved cells achieved a capacity decay of only 0.042%per cycle after 700 cycles at 3 C,demonstrating effective suppression of dendrite growth and capacity attenuation,showing promising prospect.

Background:Insomnia is a prevalent clinical condition and Shangxia Liangji formula(SXLJF)is a well-established method of ***,the specific mechanism of action of SXLJF remains ***:The mouse model of insomnia was established by intraperitoneal injection of ***-two mice were randomly divided into a negative control group,model group,SXLJF group(18.72 g/kg/day),and positive control group(diazepam,2 mg/kg)and treated with the corresponding drugs for 7 consecutive *** open field test and pentobarbital-induced sleeping test were ***-MS-based untargeted metabolomics and network pharmacology were applied to explore the potential targets of SXLJF for treating ***,key targets were validated using ***:Behavioral tests demonstrated that SXLJF reduced the total distance,average velocity,central distance,and sleep latency,and prolonged sleep *** and network pharmacology revealed potential targets,signaling pathways,metabolic pathways,and metabolites associated with the anti-insomnia effects of ***,tyrosine hydroxylase(TH)and tyrosine metabolism emerged as crucial metabolic pathways and targets,***-qPCR results supported the role of TH in the mechanism of SXLJF in treating ***:In conclusion,TH and tyrosine metabolism may represent significant targets and pathways for SXLJF in treating insomnia.

高静压联合天然抑制剂处理对浑浊苹果汁多酚氧化酶活性及相关品质的影响

探究高静压(high hydrostatic pressure,HHP)联合阿魏酸(ferulic acid,FA)、表没食子儿茶素没食子酸酯(epigallocatechin gallate,EGCG)处理对浑浊苹果汁中多酚氧化酶(polyphenol oxidase,PPO)活性及相关品质的影响。结果表明,添加0.1 g/100 mL的FA或者0.2 g/100 mL的EGCG均能显著降低样品中PPO活性,并显著提高其亮度(P<0.05)。单独HHP处理(550 MPa、5 min)样品的PPO相对残余活性高达82.70%,而经过HHP+FA或HHP+EGCG处理后,样品的PPO相对残余活性分别降至66.35%和41.50%。在冷藏2周期间,样品的PPO活性持续下降,而颜色参数保持稳定。贮藏第14天,HHP+FA组的褐变指数最小,仅有4.37,而HHP+EGCG组的PPO相对残余活性最低(24.71%)。此外,两组样品都保留了80%以上的总酚和90%以上的抗氧化能力。综上,HHP与FA/EGCG的联合处理可以有效抑制浑浊苹果汁的酶促褐变,具有进一步推广应用的潜力。

Sintering behavior and properties of SiC/Si_3N_4 composite

SiC/Si_3N_4 composite was pressureless sintered using self-propagating high-temperature combustion synthesis a-Si_3N_4 powder and appropriate amount of b-SiC powder. Both of the sintering additive systems were used,which were YAN（Y_2O_3–Al_2O_3–AlN） and YN（Y_2O_3–AlN）.The influences of b-SiC content on sintering behavior of SiC/Si_3N_4 composite were investigated. The results show that the density, shrinkage, bending strength, hardness, and fracture toughness of the samples with two sintering additive systems increase first with the increase of the contents of b-SiC and then decrease even when b-SiC contents continually increase. The tendency of weight loss of the samples is opposite. The b-SiC content of the samples with the best mechanical properties is different in two systems. For YAN system, the best mechanical properties of the samples are gained when b-SiC content reaches 10 %, while as for YN system it is 5 %. The properties of YN samples are superior to YAN samples. J phase（2Y_2O_3 ·Si_2N_2O） forms in YN system easily. A small amount of M phase（Y_2O_3·Si_3N_4） is observed in YAN system.

Investigation on removing recalcitrant toxic organic polluters in coking wastewater by forward osmosis

Investigation was made on the efficiency of two commercial membranes in removing via forward osmosis(FO)the low molecular weight organic compounds typical of coking wastewater. The membranes were supplied by Poten and HTI companies. The organics in the simulated coking water were indole and pyrridine. Under FO mode, the rejection to the organics by Poten membrane was around 50%, whereas that for HTI membrane was obviously higher, ranging from 65% to 74%. The response of the two membranes in terms of Water flux and reverse salt flux(RSF) towards changing feed/draw solution(DS) flow rates in FO mode showed similar tendency,but different degree. Generally, the flux in FO using HTI membranes was lower. For HTI membrane, FO operated with pressure retarded osmosis(PRO) mode was also performed and the overall rejection of the organics was slightly lower than that in FO mode. In the long term FO test within 15 days, both Poten and HTI membranes displayed flux reduction and rejection enhancement. But the variation with Poten membrane was much more obvious. Discussion was carried out about the reasons and the mechanisms behind the FO performance difference between two membranes and the variation in flux and rejection with operation conditions. Characterizations by SEM, FTIR, AFM, XRD and XPS were tried to support the proposed explanations.
