Pregnancy and fetal outcomes of chronic hepatitis C mothers with viremia in China

BACKGROUND Data that assess maternal and infant outcomes in hepatitis C virus(HCV)-infected mothers are limited.AIM To investigate the frequency of complications and the associated risk factors.METHODS We performed a cohort study to compare pregnancy and fetal outcomes of HCVviremic mothers with those of healthy mothers.Risk factors were analyzed with logistic regression.RESULTS Among 112 consecutive HCV antibody-positive mothers screened,we enrolled 79 viremic mothers.We randomly selected 115 healthy mothers from the birth registry as the control.Compared to healthy mothers,HCV mothers had a significantly higher frequency of anemia[2.6%(3/115)vs 19.0%(15/79),Per(3278.3±462.0 vs 3105.1±459.4 gms;P=0.006),and the mean head circumference was smaller(33.3±0.6 vs 33.1±0.7 cm;P=0.03).In a multivariate model,HCV-infected mothers were more likely to suffer anemia[adjusted odds ratio(OR):18.1,95%confidence interval(CI):4.3-76.6],require caesarian sections(adjusted OR:2.6,95%CI:1.4-4.9),and have nuchal cords(adjusted OR:5.6,95%CI:2.4-13.0).Their neonates were also more likely to have smaller head circumferences(adjusted OR:2.1,95%CI:1.1-4.3)and lower birth weights than the average(≤3250 gms)with an adjusted OR of 2.2(95%CI:1.2-4.0).The vertical transmission rate was 1%in HCV-infected mothers.CONCLUSION Maternal HCV infections may associate with pregnancy and obstetric complications.We demonstrated a previously unreported association between maternal HCV viremia and a smaller neonatal head circumference,suggesting fetal growth restriction.

Liquid biopsy for non-invasive assessment of liver injury in hepatitis B patients

BACKGROUND Hepatitis B is a major public health problem in China. Accurate liver injury assessment is essential for clinical evidence-based treatment. Liver biopsy is considered the gold standard method to stage liver disease, but it is not widely used in resource-limited settings. Therefore, non-invasive liquid biopsy tests are needed. AIM To assess liver injury in hepatitis B patients using quantified cell free DNA combined with other serum biomarker as a liquid biopsy-based method. METHODS A cohort of 663 subjects including 313 hepatitis B patients and 350 healthy controls were enrolled. Ultrasound-guided liver biopsies followed by histopathological assessments were performed for the 263 chronic hepatitis B patients to determine the degree of liver injury. Cell-free DNA was quantified using a novel duplex real-time polymerase chain reaction assay. RESULTS Compared with healthy controls, patients with hepatitis B virus (HBV) infection had significantly higher plasma DNA, serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), bilirubin, and HBV DNA levels (Perum ALT, AST, bilirubin, and plasma DNA levels of patients with markedsevere inflammation were significantly higher than those with mild-moderate inflammation (Pere was a statistically significant correlation between hepatocyte inflammation severity and serum bilirubin (R^2=0.673, Per the curves of serum ALT, bilirubin, plasma DNA, and their combination to distinguish between patients with mild–moderate and marked-severe inflammation were 0.8059, 0.7910, 0.7921, and 0.9564, respectively. CONCLUSION The combination of plasma DNA, serum ALT, and bilirubin could be a candidate liquid biopsy for non-invasive assessment of liver injury in hepatitis B patients.

To the Editor:The epidemic caused by 2019 novel coronavirus(2019-nCoV)has drawn public attention.[1]Huge infected population and enormous economic loss make it the urgent public health event to deal with.Confirmatory test targeting virus RNA was established at the early stage of outbreak and then used for 2019-nCoV infection diagnosis.[2]However,high risk of laboratory infection,high-qualified personnel,and strict operation condition hampered its application into primary hospitals and community clinics.[3]In this study,the serum immunoglobulin(Ig)M and IgG antibodies were detected in 2019-nCoV confirmed cases of different stages.Furthermore,three different immunological assays,chemiluminescent immunoassay(CLIA),gold immunochromatographic assay(GICA),and enzyme-linked immunosorbent assay(ELISA)were used for IgM and IgG detection.

While some individuals infected by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)present mild-to-severe disease,many SARS-CoV-2-infected individuals are asymptomatic.We sought to identify the distinction of immune response between asymptomatic and moderate patients.We performed single-cell transcriptome and T-cell/B-cell receptor(TCR/BCR)sequencing in 37 longitudinal collected peripheral blood mononuclear cell samples from asymptomatic,moderate,and severe patients with healthy controls.Asymptomatic patients displayed increased CD56^(bri)CD16^(-) natural killer(NK)cells and upregulation of interferon-gamma in effector CD4^(+) and CD8^(+) T cells and NK cells.They showed more robust TCR clonal expansion,especially in effector CD4^(+) T cells,but lack strong BCR clonal expansion compared to moderate patients.Moreover,asymptomatic patients have lower interferon-stimulated genes(ISGs)expression in general but large interpatient variability,whereas moderate patients showed various magnitude and temporal dynamics of the ISGs expression across multiple cell populations but lower than a patient with severe disease.Our data provide evidenee of different immune signatures to SARS-CoV-2 in asymptomatic infections.

Genotyping of strains of Yersinia pestis from Marmota baibacina-Spermophilus undulates Plague Focus of Tianshan Mountains in China

The airn of this study is to carry out genotyping of 61 Y.pestis strains isolated from Marmota baibacina-Spermophilus undulates Plague Focus of Tianshan Mountains in China. Primer pairs targeting the 22 DFRs were designed for detecting the genotypes of 61 strains. As a result, 61 strains of Y.pestis were divided into four genotypes 1, 2, 3, and 4. Genotypes 1, 2 and 3 were found from west part in Northern Tianshan Mountains Plague Focus, but type 1 was only from Nileke county. The type of strains from Aheqi was different from those of Atushi counties in Southern Tianshan Mountains and similar to strains in Northern Tianshan Mountains Plague Focus. The type 4 distributed over Atushi county, which was identical with that of strains from Marmota caudae Plague Focus of the Pamiri Plateau. It is concluded that geonotyping is identical with ecotyping made by Shuli Ji. Tianshan Mountains Plague Focus and Marmota caudae Plague Focus of the Pamiri Plateau have a cross spreading profile.

Quantitative characterizations of anisotropic dynamic properties in organic-rich shale with different kerogen content

Understanding the quantitative responses of anisotropic dynamic properties in organic-rich shale with different kerogen content(KC)is of great significance in hydrocarbon exploration and development.Conducting controlled experiments with a single variable is challenging for natural shales due to their high variations in components,diagenesis conditions,or pore fluid.We employed the hot-pressing technique to construct 11 well-controlled artificial shale with varying KC.These artificial shale samples were successive machined into prismatic shape for ultrasonic measurements along different directions.Observations revealed bedding perpendicular P-wave velocities are more sensitive to the increasing KC than bedding paralleling velocities due to the preferential alignments of kerogen.All elastic stiffnesses except C_(13)are generally decreasing with the increasing KC,the variation of C_(1) and C_(33)on kerogen content are more sensitive than those of C_(44)and C_(66).Apparent dynamic mechanical parameters(v and E)were found to have linear correlation with the true ones from complete anisotropic equations independent of KC,which hold value towards the interpretation of well logs consistently across formations,Anisotropic mechanical parameters(ΔE and brittlenessΔB)tend to decrease with the reducing KC,withΔB showing great sensitivity to KC variations.In the range of low KC(erve as a significant characterization for highly organic-rich(>10%)shale,compilation of data from natural organic rich-shales globally verified the similar systematic relationships that can be empirically used to predict the fraction of KC in shales.

Molybdenum and cadmium co-induce necroptosis through Th1/Th2 imbalance-mediated endoplasmic reticulum stress in duck ovaries

Cadmium(Cd)and excess molybdenum(Mo)pose serious threats to animal health.Our previous study has determined that Cd and/or Mo exposure can cause ovarian damage of ducks,while the specific mechanism is still obscure.To further investigate the toxic mechanism of Cd and Mo co-exposure in the ovary,forty 8-day-old female ducks were randomly allocated into four groups for 16 weeks,and the doses of Cd and Mo in basic diet per kg were as follows:control group,Mo group(100 mg Mo),Cd group(4 mg Cd),and Mo+Cd group(100 mg Mo+4 mg Cd).Cadmium sulfate 8/3-hydrate(CdSO_(4)·8/3H_(2)O)and hexaammonium molybdate((NH_(4))_(6)Mo_(7)O_(24)·4H_(2)O)were the origins of Cd and Mo,respectively.At the 16th week of the experiment,all ovary tissues were collected for the detection of related indexes.The data indicated that Mo and/or Cd induced trace element disorders and Th1/Th2 balance to divert toward Th1 in the ovary,which activated endoplasmic reticulum(er)stress and then provoked necroptosis through triggering RIPK1/RIPK3/MLKL signaling pathway,and eventually caused ovarian pathological injuries and necroptosis characteristics.The alterations of above indicators were most apparent in the joint group.Above all,this research illustrates that Mo and/or Cd exposure can initiate necroptosis through Th1/Th2 imbalance-modulated er stress in duck ovaries,and Mo and Cd combined exposure aggravates ovarian injuries.This research explores the molecular mechanism of necroptosis caused by Mo and/or Cd,which reveals that er stress attenuation may be a therapeutic target to alleviate necroptosis.

Effect of er_(30)Cu_(70) on microstructure and properties of sintered Nd-Fe-B by grain boundary reconstruction

Usually it is generally believed that the er element forms the er_(2)Fe_(14)B phase,which will seriously deteriorate the magnetic properties.Distinctly,here we report the balance of corrosion resistance and coercivity in Nd-Fe-B sintered magnets through using simple er_(30)Cu_(70) additive whose price is much lower than Dy and Tb.By reasonably controlling er_(30)Cu_(70) addition,the corrosion resistance is improved at the minimum limit of reducing the magnetic properties.Through studying the influence mechanism of er element,it is found that the main effect of er elements is to replace the Nd elements at the edge of the main phase grains to form a(er,Nd)_(2)Fe_(14)B shell with low H_(A),resulting in the reduction of magnetic properties.The improvement of corrosion resistance mainly comes from the more stable Cu element introduced at the grain boundary.At the same time,the target magnets also show different advantages under different heat treatment methods.Above findings may spur progress towards developing the lowcost permanent magnets that rival the commercial Nd-Fe-B counterpart.
