Prompt and Easy Activation by Specific Thioredoxins of Calvin Cycle Enzymes of Arabidopsis thaliana Associated in the GAPDH/CP12/PRK Supramolecular Complex

The Calvin cycle enzymes glyceraldehyde-3-phosphate dehydrogenase （GAPDH） and phosphoribulokinase （PRK） can form under oxidizing conditions a supramolecular complex with the regulatory protein CP12. Both GAPDH and PRK activities are inhibited within the complex, but they can be fully restored by reduced thioredoxins （TRXs）. We have investigated the interactions of eight different chloroplast thioredoxin isoforms （TRX f1, m1, m2, m3, m4, y1, y2, x） with GAPDH （A4, B4, and B8 isoforms）, PRK and CP12 （isoform 2）, all from Arabidopsis thaliana. In the complex, both A4-GAPDH and PRK were promptly activated by TRX f1, or more slowly by TRXs ml and m2, but all other TRXs were ineffective. Free PRK was regulated by TRX f1, m1, or m2, while B4- and Bs-GAPDH were absolutely specific for TRX fl. Interestingly, reductive activation of PRK caged in the complex was much faster than reductive activation of free oxidized PRK, and activation of A4-GAPDH in the complex was much faster （and less demanding in terms of reducing potential） than activation of free oxidized B4- or BB-GAPDH. It is proposed that CP12-assembled supramolecular complex may represent a reservoir of inhibited enzymes ready to be released in fully active conformation following reduction and dissociation of the complex by TRXs upon the shift from dark to low light. On the contrary, autonomous redox-modulation of GAPDH （B- containing isoforms） would be more suited to conditions of very active photosynthesis.

Sunlight represents the energy source for photosynthesis and plant growth. When growing in the field, plant photosynthesis has to manage strong fluctuations in light intensities. Regulation based on the thio- redoxin （Trx） system is believed to ensure light-responsive control of photosynthetic reactions in the chlo- roplast. However, direct evidence for a role of this system in regulating dynamic acclimation of photosyn- thesis in fluctuating conditions is largely lacking. In this report we show that the ferredoxin-dependent Trxs ml and m2 as well as the NADPH-dependent NTRC are both indispensable for photosynthetic acclimation in fluctuating light intensities. Arabidopsis mutants with combined deficiency in Trxs ml and m2 show wild- type growth and photosynthesis under constant light condition, while photosynthetic parameters are strongly modified in rapidly alternating high and low light. Two independent trxmlm2 mutants show lower photosynthetic efficiency in high light, but surprisingly significantly higher photosynthetic efficiency in low light. Our data suggest that a main target of Trx ml and m2 is the NADP-malate dehydrogenase involved in export of excess reductive power from the chloroplast. The decreased photosynthetic efficiency in the high-light peaks may thus be explained by a reduced capacity of the trxm lm2 mutants in the rapid light acti-vation of this enzyme. In the ntrc mutant, dynamic responses of non-photochemical quenching of excita- tion energy and plastoquinone reduction state both were strongly attenuated in fluctuating light intensities, leading to a massive decrease in PSII quantum efficiency and a specific decrease in plant growth under these conditions. This is likely due to the decreased ability of the ntrc mutant to control the stromal NADP（H） redox poise. Characterisation of the granular dynamics at the interface between a pipe and a granular flow in a rotating drum

A new mobile bed heat exchanger is presented in this work which is composed of a flowing granular material in a rotating drum and a cylindrical pipe with potential interest in different energy applications as cooling,heating or heat recovery *** optimal design of the device requires a characterisation of the phenomena involved at the interface between the granular flow and the *** process is modelled by the discrete element method and a global classification of the flow patterns around the pipe is presented with respect to the three main control parameters of the problem:the Froude number,the diameter ratio and the relative filling height of the *** second part is devoted to the characterisation of the structure of the flow at the interface(velocity field,density field)in particular in a so-called Biflow regime where granular motion occurs above as well as below the pipe which is favourable to transfer by convection.A typical behavior at the interface with the pipe consists of a zone I with high velocities of particles at the top of the pipe,a second zone with quasistatic particles or low velocity particles at the front and at the bottom of the pipe and a last zone ll of depletion of particles at the back of the *** Froude number has a limited effect on the features of this structure on the first layer in the range of Froude numbers considered whereas the relative height is a more determinant parameter to control the relative magnitude of velocities in zone I and zone Il as well as the extent of the depletion *** first hydrodynamical characterisation can shed light on the dynamical regimes with improved transfer between the particles and the pipe boundary.

Extensive Bi-Atrial Remodeling on Hypertensive Patient with Permanent Atrial Fibrillation Delayed Diagnosis of Fatal Cardiac Lambda Amyloidosis

Background: Amyloidosis is a disease characterized by the deposition of fibrillar proteins in tissues. The nature of the protein defines the type of amyloidosis. Cardiac involvement is most often secondary to deposits of transthyretin and immunoglobulin light chains. Treatment depends on the type of amyloidosis. Cardiac light chain amyloidosis is a medical emergency. Aim: To highlight the importance of an early diagnosis of cardiac light chain amyloidosis. Case Presentation: We report the case of an 88-year-old hypertensive female patient with sustained atrial fibrillation and recurrent heart failure, in whom echocardiography showed concentric left ventricle hypertrophy with mildly reduced left ventricle ejection fraction (LVEF) to 45%. Bone scintigraphy was normal. Serum analysis showed increased lambda free light chains. Accessory salivary gland biopsy revealed weak Kappa light chain staining and clear overexpression of lambda light chain deposits. The diagnosis of stage 3B cardiac amyloidosis secondary to lambda light chain myeloma was made. After a multidisciplinary meeting, it was decided to start treatment with DARATUMUMAB + LENALIDOMIDE. Patient’s general condition deteriorated with the occurrence of febrile pancytopenia. Chemotherapy was stopped and management was limited to comfort care until the patient’s death. Conclusion: Cardiac light-chain amyloidosis must be diagnosed early as it can be rapidly fatal.

Geochemical Analysis of Albian-Maastrichtian Formations in the Offshore Basin of the Abidjan Margin: Rock-Eval Pyrolysis Study

The Albian-Maastrichtian interval of the Ivorian sedimentary basin has been the subject of numerous sedimentological, biostratigraphic, and geophysical studies. However, its geochemical characteristics remain relatively unexplored. This study aims to determine the oil potential and the nature of the organic matter it contains. It focuses on the geochemical analysis (physicochemical method) of two oil wells located in the offshore sedimentary basin of Côte d’Ivoire, specifically in the Abidjan margin. A total of 154 cuttings samples from wells TMH-1X and TMH-2X were analyzed to determine their oil potential and the nature of the organic matter (OM) they contain. The analyses were performed using Rock-Eval pyrolysis, a method that characterizes the amount of hydrocarbons generated by the organic matter present in the rocks. The key parameters measured include Total Organic Carbon (TOC), Hydrogen Index (HI), oil potential (S2), and maximum pyrolysis temperature (Tmax). These parameters are used to assess the amount of organic matter, its thermal maturity, and its potential to generate hydrocarbons in the studied wells. The results show significant variations between different stratigraphic levels. In well TMH-1X, the Cenomanian and Campanian intervals stand out with very good quantities of organic matter (OM) with good oil potential, although often immature. In contrast, other stages such as the Albian and Turonian contain organic matter in moderate to low quantities, often immature and of continental type, which limits their capacity to generate hydrocarbons. In well TMH-2X, a similar trend is observed. Despite an abundance of organic matter, the oil potential remains low in most of the studied stages. The organic matter is primarily of type III (continental origin) and thermally immature, indicating a low potential for hydrocarbon generation. The synthesis of new 4-imino-4H-chromeno[2,3-d]pyrimidin-3(5H)-amine in four steps including one step under microwave dielectric heating is reported. The structural identity of the synthesized compounds was established according to their spectroscopic analysis, such as FT-IR, NMR and mass spectroscopy. These new compounds were tested for their antiproliferative activities on seven representative human tumoral cell lines (Huh7 D12, Caco2, MDA-MB231, MDA-MB468, HCT116, PC3 and MCF7) and also on fibroblasts. Among them, only the compounds 6c showed micromolar cytotoxic activity on tumor cell lines (1.8 50 50 > 25 μM). Finally, in silico ADMET studies ware performed to investigate the possibility of using of the identified compound 6c as potential anti-tumor compound.

Invasive rats strengthen predation pressure on bird eggs in a South Pacific island rainforest

Invasive rats （Rattus spp.） are known to have pervasive impacts on island birds, particularly on their nesting success. To conserve or restore bird populations, numerous invasive rat control or eradication projects are undertaken on islands worldwide. However, such projects represent a huge investment and the decision-making process requires proper assessment of rat impacts. Here, we assessed the influence of two sympatric invasive rats （Rattus rattus and R. exulans） on native bird eggs in a New Caledonian rainforest, using artificial bird-nest monitoring. A total of 178 artificial nests containing two eggs of three different sizes were placed either on the ground or 1.5 m high and monitored at the start of the birds＇ breeding season. Overall, 12.4% of the nests were depredated during the first 7 days. At site 1, where nests were monitored during 16 days, 41.8% of the nests were depredated. The main predator was the native crow Corvus moneduloides, responsible for 62.9% of the overall predation events. Rats were responsible for only 22.9% of the events, and ate only small and medium eggs at both heights. Our experiment suggests that in New Caledonia, predation pressure by rats strengthens overall bird-nest predation, adding to that by native predators, Experimental rat control operations may allow reduced predation pressure on nests as well as the recording of biodiversity responses after rat population reduction.

Wolbachia Infection in Populations of Ostrinia furnacalis: Diversity, Prevalence, Phylogeny and Evidence for Horizontal Transmission

Wolbachia are maternally inherited endosymbiotic bacteria infecting a wide range of arthropods and filarial nematodes. They can induce various reproduction alterations in their hosts, including thelytokous parthenogenesis, cytoplasmic incompatibility （CI）, feminization of genetic males and male killing. Here we investigated diversity and prevalence patterns of Wolbachia infection in 43 geographical populations of the Asian corn borer, Ostriniafurnacalis, in China and one population in North Korea. Based on Wolbachia surface protein gene （wsp） sequences, nine strains of Wolbachia （wFurl-wFur9）, belonging to supergroups A and B, were identified in populations of O. furnacalis with an average infection rate of 10.5%. Superinfection commonly appeared in individuals of O. furnacalis and coinfection patterns were very complex. There was no specific pattern for the prevalence and distribution of the nine Wolbachia strains suggesting an intricate evolutionary history of Wolbachia infection in this species. The genetic similarity of the wFurl-wFur9 strains with those detected in two parasitoids of O. furnacalis, Macrocentrus cingulum and Lydella grisescens, strongly suggests host-parasitoid horizontal transmission.
