Loss of miR-83 extends lifespan and affects target gene expression in an age-dependent manner in Caenorhabditis elegans

MicroRNAs(miRNAs) are short non-coding RNAs that are involved in the post-transcriptional regulation of protein-coding genes. miRNAs modulate lifespan and the aging process in a variety of organisms. In this study, we identified a role of miR-83 in regulating lifespan of Caenorhabditis elegans. mir-83 mutants exhibited extended lifespan, and the overexpression of miR-83 was sufficient to decrease the prolonged lifespan of the mutants. We observed upregulation of the expression levels of a set of miR-83 target genes in young mir-83 mutant adults; while different sets of genes were upregulated in older mir-83 mutant adults. In vivo assays showed that miR-83 regulated expression of target genes including din-1,spp-9 and col-178, and we demonstrated that daf-16 and din-1 were required for the extension of lifespan in the mir-83 mutants. The regulation of din-1 by miR-83 during aging resulted in the differential expression of din-1 targets such as gst-4 and gst-10. In daf-2 mutants, the expression level of miR-83 was significantly reduced compared to wild-type animals. We identified a role for miR-83 in modulating lifespan in C. elegans and provided molecular insights into its functional mechanism.

Increased hazards threatening the United Nations Educational,Scientific and Cultural Organization(UNESCO)-designated sites and endangering cultural heritage and community well-being require attention and ***-sidering the pivotal role of UNESCO sites in conservation and development,this study assessed their levels of disaster *** absence of studies assessing disaster awareness,risk perception,and preparedness among UNESCO site actors,as well as the pivotal place of preparedness within the Disaster Risk Management(DRM)cycle justifies this *** the tenets of the Person-Relative-to-Event framework,we hypothe-sized that a strong positive correlation exists between perceived risks,resources,and disaster *** collect pertinent data,we employed an embedded mixed-method design and conducted an online questionnaire survey yielding 141 responses from 59 *** the results of relevant analyses,wildfires,floods,and droughts are top hazards occurring frequently in UNESCO sites,with significant concerns about pollution and habitat loss during future *** emerged as the most available crucial DRM resource,with higher availability of DRM resources correlating positively and significantly with sites’*** findings con-tribute valuable insights to address missing links for disaster-ready and resilient UNESCO sites,promoting their preservation for future generations.

Bulk geochemistry,Sr,Nd,and O-H isotope systematics are reported for the first time on banded iron formation(BIF)-hosted high-grade iron ore at the northwestern segment of Congo Craton(CC).Located in Mbalam iron ore district,Southern Cameroon,Metzimevin iron ore deposit is a hematite-magnetite BIF system,dominated by SiO_(2)+Fe_(2)O_(3)(97.1 to 99.84 wt%),with low concentrations of clastic elements e.g.,Al_(2)O_(3),TiO_(2),and HFSE,depicting a nearly pure chemical *** REE+Y signature of the iron deposit displays strong positive Eu anomaly,strong negative Ce anomaly,and chondritic to superchondritic Y/Ho ratios,suggestive of formation by mixed seawater-high temperature hydrothermal fluids in oxidising ***^(87)Sr/^(86)Sr ratios of the BIF are higher than the maximum^(87)Sr/^(86)Sr evolution curves for all Archean reservoirs(bulk silicate earth,Archean crust and Archean seawater),indicating involvement of continentally-derived components during BIF formation and ***Ɛ_(Nd)(t)(+2.26 to+3.77)and Nd model age indicate that chemical constituents for the BIF were derived from undifferentiated crustal source,between 3.002 and 2.88 *** variable and diverse O and H isotope data(−1.9‰to 17.3‰and−57‰to 136‰respectively)indicate that the Metzimevin iron ore formed initially from magmatic plumes and later enriched by magmatic-metamorphic-modified meteoric *** balance calculations indicate mineralisation by combined leaching and precipitation,with an average iron enrichment factor of>2.67 and SiO_(2)depletion factor of>*** is associated with an overall volume reduction of 28.27%,reflecting net leaching and volume collapse of the BIF protholith.

Finite domain solution of a KGD hydraulic fracture in the viscosity-dominated regime

This paper describes a numerical algorithm for solving the classic problem of a plane strain(KGD)fracture propagating in an impermeable elastic medium with zero *** method,which takes advantage of the self-similar nature of the solution,combines a domain-based scheme to solve the elasticity equations and a finite volume method to solve the nonlinear lubrication *** work represents a first step towards developing a model able to account for pore pressure diffusion in the medium and corresponding poroelastic effects,noting that these processes are more efficiently solved using a domain-based rather than a boundary integral *** enhance the efficiency and accuracy of the numerical scheme,the far-field crack asymptotics is embedded in the discretized elastic relationship between the fluid pressure and the crack opening,while the coupled fluid-solid tip asymptote is enforced in a weak form when solving the nonlinear lubrication *** proposed technique yields results that closely match the analytical solution,even with a coarse *** approach offers potential for addressing more complex hydraulic fracturing problems in the future.

Assessment of groundwater quantity, quality, and associated health risk of the Tano river basin, Ghana
