Superior Conjugative Plasmids Delivered by Bacteria to Diverse Fungi

Fungi are nature’s recyclers,allowing for ecological nutrient cycling and,in turn,the continuation of life on *** fungi inhabit the human microbiome where they can provide health benefits,while others are opportunistic pathogens that can cause ***,members of the fungal kingdom,have been domesticated by humans for the production of beer,bread,and,recently,medicine and ***,the great untapped potential exists within the diverse fungal ***,many yeasts are intractable,preventing their use in biotechnology or in the development of novel treatments for pathogenic ***,as a first step for the domestication of new fungi,an efficient DNA delivery method needs to be ***,we report the creation of superior conjugative plasmids and demonstrate their transfer via conjugation from bacteria to 7 diverse yeast species including the emerging pathogen Candida *** create our superior plasmids,derivatives of the 57 kb conjugative plasmid pTA-Mob 2.0 were built using designed gene deletions and insertions,as well as some unintentional ***,a cluster mutation in the promoter of the conjugative gene traj had the most significant effect on improving conjugation to *** addition,we created Golden Gate assembly-compatible plasmid derivatives that allow for the generation of custom plasmids to enable the rapid insertion of designer genetic ***,we demonstrated that designer conjugative plasmids harboring engineered restriction endonucleases can be used as a novel antifungal agent,with important applications for the development of next-generation antifungal therapeutics.

Obesity has been linked to a range of pathologies,including *** contrast,regular physical activity is associated with the prevention or reduced progression of ***,physical activity can improve memory and spatial cognition,reduce age-related cognitive decline,and preserve brain volume,but the mechanisms are not fully ***,we investigated whether any detrimental effects of high-fat diet(HFD)-induced obesity on cognition,motor behavior,adult hippocampal neurogenesis,and brain-derived neurotrophic factor(BDNF)could be mitigated by voluntary exercise training in male C57Bl/6 ***-induced impairment of motor function was not reversed by ***,voluntary wheel running improved long-term memory and increased hippocampal neurogenesis,suggesting that regular physical activity may prevent cognitive decline in obesity.

Methane cracking in molten tin for hydrogen and carbon production—a comparison with homogeneous gas phase process

Methane cracking is considered a bridge technology between gray and green hydrogen production *** this work an experimental study of methane cracking in molten tin is *** tests were conducted in a quartz reactor(i.d.=1.5 cm,l=20 cm)with capillary injection,varying temperature(950–1070℃),inlet methane flow rate(30–60 ml·min^(-1))and tin height(0–20 cm).The influence of the residence time in the tin and in the headspace on methane conversion and on carbon morphology was *** conversions obtained in tin and in the empty reactor were measured and compared with results of detailed kinetic simulations(CRECK).Furthermore,an expression of a global kinetic constant for methane conversion in tin was also *** highest conversion(65%at Q_(0)=30 ml·min^(-1) and t=1070℃)is obtained for homogeneous gas phase reaction due to the long residence time(70 s),the presence of tin leads to a sharp decrease of residence time(1 s),obtaining a conversion of 35%at 1070℃,thus meaning that tin owns a role in the *** characterization(scanning electron microscopy,Raman)reported a change in carbon toward sheet-like structures and an increase of the carbon structural order in the presence of molten tin media.

On a Generalized Centralizer Ring of a Finite Group Which Det ermines the Group

The k-characters are class functions on subsets of G^k called the k-classes of G. For a finite group G, the k-class sums form a basis for a subring of CG^k, which we call the k-S-ring of G、and we think of these rings as generalized centralizer rings. We show that for a finite group G the 3-S-ring determines G. More specifically, the group characters and set products of certain 3-classes of G, which we call “uniform 3-classes", determine G.

Wild tigers are in a critical state with an estimated population decline of more than 95%over the past *** the capacity and effectiveness of law enforcement in reducing poaching of tigers is an immediate priority to secure remaining wild populations in source *** 2008-2010,standardized patrol-based law enforcement monitoring(lEM)was established under the Tigers Forever Program across 8 key tiger sites in order to improve and evaluate law enforcement ***-based monitoring has the distinct advantage of providing regular and rapid information on illegal activities and ranger performance,although,until recently,it has received relatively little scrutiny from the conservation *** present paper outlines a framework for implementation of lEM in tiger source sites using MIST,a computerized management information system for ranger-based data *** framework addresses many of the technical,practical and institutional challenges involved in the design,implementation,sustainability and evaluation of *** of such a framework for lEM is a cost-effective strategy to improve the efficiency of law enforcement efforts,to increase the motivation of enforcement staff and to promote the accountability of law enforcement agencies in addressing threats to *** combined with independent,systematic and science-based monitoring of tigers and their prey,lEM has great potential for evaluating the effectiveness of protection-based conservation investments.

Amyotrophic lateral sclerosis(AlS)and frontotemporal dementia(FTD)are both devastating neurodegenerative *** affecting different regions of the nervous system(FTD affecting primarily the frontal and temporal lobes,whilst AlS presents with motor neuron loss),there is significant overlap between these conditions in terms of genetics,pathology,and disease mechanisms,and they are therefore often grouped as a spectrum of symptoms under the heading FTD/AlS(Abramzon et al.,2020).Significantly,there is currently no cure for AlS or ***,recent mechanistic insight points to a novel pathway to target for potential therapeutic intervention.

Does the scent of a potential mate prevent the resorption of oocytes by apoptosis in Nauphoeta cinerea？

We expect organisms to have evolved mechanisms to gather and use the information available within their environments, to steer resource investment decisions toward the traits that will give the greatest fitness returns. Pheromones are social signals, a common purpose of which is to act as indicators of mate presence. Consequently, pheromones have the potential to act as signals to increase or maintain reproductive investment over that of competing life-history traits. In the cockroach, Nauphoeta cinerea （Oliver）, females pay costs of maintaining investment in reproduction when there are no males, and males produce pheromones that are known to effect female reproductive outcomes. Whether the pheromones have an influence on resource physiology is unknown. We subjected newly eclosed females to either a synthetic blend of male pheromones or control. We dissected females at 7, 12, 17 or 22 days. We measured apoptosis levels and size of all oocytes within the vitellerium, and measured dry fat body mass. Synthetic male pheromone blend did not have an effect on any measure of female reproductive or somatic resource physiology. Although negative results are always problematic, the success of the synthetic pheromone method in the past suggests that females may be insensitive to male pheromones in the pre-oviposition period, and may rely on mating stimulus rather than pheromone as the cue for oocyte maintenance and growth. Previous studies reporting effects of male pheromones on female reproduction suggest that the period of pheromone sensitivity may be between mating and birth.

Background Pediatric post coronavirus disease 2019(COVID-19)condition(PPCC)is a heterogeneous syndrome,which can significantly affect the daily lives of *** study aimed to identify clinically meaningful phenotypes in children with PPCC,to better characterize and treat this *** Participants were children with physician-diagnosed PPCC,referred to the academic hospital Amsterdam UMC in the Netherlands between November 2021 and March *** factors and information on post-COVID symptoms,comorbidities,and impact on daily life were *** clusters were identified using an unsupervised and unbiased approach for mixed data *** Analysis of 111 patients(aged 3–18 years)revealed three distinct clusters within *** 1(n=62,median age=15 years)predominantly consisted of girls(74.2%).These patients suffered relatively more from exercise intolerance,dyspnea,and smell *** 2(n=33,median age=13 years)contained patients with an even gender distribution(51.5%girls).They suffered from relatively more sleep problems,memory loss,gastrointestinal symptoms,and *** 3(n=16,median age=11 years)had a higher proportion of boys(75.0%),suffered relatively more from fever,had significantly fewer symptoms(median of 5 symptoms compared to 8 and 10 for clusters 1 and 2 respectively),and experienced a lower impact on daily *** This study identified three distinct clinical PPCC phenotypes,with variations in sex,age,symptom patterns,and impact on daily *** findings highlight the need for further research to understand the potentially diverse underlying mechanisms contributing to post-COVID symptoms in children.
