Gold nanoshells： Contrast agents for cell imaging by cardiovascular optical coherence tomography

Optical coherence tomography （OCT） has gained considerable attention in interventional cardiovascular medicine and is currently used in clinical settings to assess atherosclerotic lesions and to optimize stent placement. Artery imaging at the cellular level constitutes the first step towards cardiovascular molecular imaging, which represents a major advance in the development of personalized noninvasive therapies. In this work we demonstrate that cardiovascular OCT can be used to detect individual cells suspended in biocompatible fluids. Importantly, the combination of this catheter-based clinical technique with gold nanoshells （GnSs） as intracellular contrast agents led to a substantial enhancement in the backscattered signal produced by individual cells. This cellular contrast enhancement was attributed to the large backscattering cross-section of GnSs at the OCT laser wavelength （1,300 nm）. A simple intensity analysis of OCT cross-sectional images of suspended cells makes it possible to identify the sub-population of living cells that successfully incorporated GnSs. The generalizability of this method was demonstrated using two different cell lines （HeLa and Jurkat cells）. This work provides novel insights into cardiovascular molecular imaging using specifically modified GnSs.

AIM To investigate the main current etiologies of cirrhosis in *** We performed a cross-sectional retrospective multicenter study that included eight hospitals in different areas of Mexico. These hospitals provide health care to people of diverse social classes. The inclusion criteria were a histological, clinical, biochemical, endoscopic, or imaging diagnosis of liver cirrhosis. Data were obtained during a 5-year period(January 2012-December 2017). RESULTS A total of 1210 patients were included. The mean age was 62.5 years(SD = 12.1), and the percentages of men and women were similar(52.0% vs 48.0%). The most frequent causes of liver cirrhosis were hepatitis C virus(HCV)(36.2%), alcoholic liver disease(ALD)(31.2%), and nonalcoholic steatohepatitis(23.2%), and the least frequent were hepatitis B virus(1.1%), autoimmune disorders(7.3%), and other conditions(1.0%).COnCLUSIOn HCV and ALD are the most frequent causes of cirrhosis in Mexico. However, we note that non-alcoholic fatty liver disease(nAFLD) as an etiology of cirrhosis increased by 100% compared with the rate noted previously. We conclude that nAFLD will soon become one of the most frequent etiologies of liver cirrhosis in Mexico.

The American Kennel Club and the Fédération Cynologique Internationale recognize two phenotypic variants of Great Dane breed denominated American and European varieties. Historically, these varieties have been segregated according to morphological characteristics. In an attempt to obtain a better characterization, breeders have been interested in the genetic parameters that could evaluate the within-breed diversity. In this document, we studied the genetic structure of Great Dane breed with 10 STR markers in 88 dogs using capillary electrophoresis. Cluster analysis, population differentiation and phylogenetic analyses revealed that American and European varieties are genetically independent. nevertheless, within the American variety a genetic stratification was found. Additionally, a high misclassification (28%) was detected, which could be due to wrong registration or false paternity. Our results support the importance to deem genetic markers as useful tools in breeding control. Similarly, these studies serve as an accurate reference to establish standards by dog breeding associations and for choosing among dogs for inter-breeding. nevertheless, genetic tools are only a complement of morphological methods, since both are reshuffling the control over the breeding line. notwithstanding, this database provides an overall and scape concerning the impact of genetic diversity within-breed. Unmistakably, more databases are needed to increase the quality of the breeding line as well as the number of STR in order to study, with more detail, the genetic structure in the Great Dane race.

Four new fluorescent sensors(1-4)based on the 4-amino-1,8-naphthalimide fluorophores(naps)have been synthesized based on the classical fluorophore-spacer-receptor *** four compounds all gave rise to emission bands centred at ca.535 nm,which were found to be highly pH dependent,the emission being‘switched on’in acidic media,while being quenched due to PET from the amino moieties to the excited state of the nap at more alkaline *** luminescent pH dependence for these probes was found to be highly dependent on the substitution on the imide site,as well as the polyamine chain attached to the position 4-amino *** the case of sensor 2 the presence of the 4-amino-aniline dominated the pH dependent ***,at higher pH,PET quenching was also found to occur from the polyamine ***,2 is better described as a receptor1-spacer1-fluorophore-spacer2-receptor2 system,where the dominant PET process is due to(normally less favourable)‘directional’PET quenching from the 4-amino-aniline unit to the nap *** trends and pH fluorescence dependences were also seen for 3 and *** compounds were also tested for their imaging potential and toxicity against HeLa cells(using DRAQ5 as nuclear stain which does now show pH dependent changes in acidic and neutral pH)and the results demonstrated that these compounds have reduced cellular viability at moderately high concentrations(with IC50 values between ca.8-30μmol?L^?1),but were found to be suitable for intracellular pH determination at 1μmol?L^?1 concentrations,where no real toxicity was *** allowed us to employ these as lysosomal probes at sub-toxic concentrations,where the nap based emission was found to be pH depended,mirroring that seen in aqueous solution for 1-4,with the main fluorescence changes occurring within acidic to neutral pH.

Dear Editor,Triple-negative breast cancer(TnBC)accounts for 20%of all breast carcinomas and lacks a validated targeted therapy;thus,currently,cytotoxic chemotherapy is the treatment of choice[1].Compared with other breast cancer types,patientswith TnBCare younger,have larger tumors,a higher risk of metastasis,and a higher rate of recurrence[2].Among all the subtypes described by Lehmann et al.[3],mesenchymal-like and mesenchymal stem-like(MSL)subtypes had the lowest 5-year distant metastasis-free survival rates.

Microbiological Quality of Asadero Cheese Manufactured with a Plant Based Coagulant from <i>Solanum elaeagnifolium</i>

In Chihuahua, north Mexico, the berries of Solanum elaeagnifolium (trompillo or silverleaf nightshade) have been used in the manufacture of artisanal filata-type asadero cheese. Solanum elaeagnifolium is a wild plant that possesses proteases in its fruit;those enzymes exhibit general proteolytic activities, which are useful in traditional asadero cheesemaking as a rennet substitute. These type of cheese is softer than those made with rennin or chymosin due to their water content and proteolysis. Thus, the aim of this work was to explore microbiological susceptibility of asadero cheese made with an extract from ripe berries and compared with those elaborated with commercial rennet. Crude extract of such fruit was obtained by salting out with ammonium sulfate (40%). Cheeses were obtained by a standardized process only changing the enzyme, packaged in plastic bag or vacuum bag and storage at 4℃ -?6℃. The study included the analysis of spoilage bacteria (total aerobic count, yeasts and molds), indicators of hygiene (total coliforms and E. coli) and fecal contamination (fecal coliforms) after 1, 7, 14, 21 and 28 days of production. Although cheese made with S. elaeagnifolium had lower microbial growth, the aerobic count and yeasts and molds count of all cheese samples showing a 28 days shelf life. Total coliforms, in limits beyond the established ones by the Mexican legislation for human consumption with no effect of the enzyme type used for production, as well as for fecal coliforms. Differences were detected just for packaged method, suggesting that good manufacturing practices take place not the plant coagulant.

Effectiveness of an intervention for reducing social stigma towards mental illness in adolescents

AIM: To evaluate the effectiveness of an intervention for reducing social stigma towards mental illness in adolescents. The effect of gender and knowledge of someone with mental illness was measured. METHODS: Two hundred and eighty secondary school students were evaluated using the Community Attitudes towards Mental Illness(CAMI) questionnaire. The schools were randomized and some received the intervention and others acted as the control group. The programme consisted of providing information via a documentary film and of contact with healthcare staff in order to reduce the social stigma within the school environment. RESULTS: The intervention was effective in reducing the CAMI authoritarianism and social restrictiveness subscales. The intervention showed significant changes in girls in terms of authoritarianism and social restrictiveness, while boys only showed significant changes in authoritarianism. Following the intervention, a significant reduction was found in authoritarianism and social restrictiveness in those who knew someone with mental illness, and only in authoritarianism in those who did not know anyone with mental illness. COnCLUSIOn: The intervention was effective to reduce social stigma towards people with mental illness, especially in the area of authoritarianism. Some differences were found depending on gender and whether or not the subjects knew someone with mental illness.

在西班牙进行环境污染物对猛禽种群压力的最早研究可追溯到20世纪80年代,通过多种角度使用多种前哨猛禽物种。然而,由于没有国家的监测计划,所以研究无论在时间或是空间上都是零星的。对金属的暴露并没有随着时间的推移变化,但存于卵和组织中的铅浓度有所减少。一般来说,在西班牙猛禽样本中检测到的金属浓度通常很低,不足以产生毒副作用。除DDT和DDE,在过去的20年中西班牙猛禽类中大部分以有机氯为基础的杀虫剂已经减少。在几个不同物种的卵中所发现的DDE浓度可以部分地解释在西班牙猛禽能够成功繁殖的原因。
