Dementia and osteoporosis in a geriatric population: Is there a common link?

AIM To determine the existence of a common pathological link between dementia and osteoporosis through reviewing the current evidence base. METHODS This paper reviews the current literature on osteoporosis and dementia in order to ascertain evidence of a common predisposing aetiology. A literature search of Ovid MEDLINE(1950 to June 2016) was conducted. The keywords "osteoporosis", "osteoporotic fracture", "dementia" and "Alzheimer's disease"(AD) were used to determine the theoretical links with the most significant evidence base behind them. The key links were found to be vitamins D and K, calcium, thyroid disease, statins, alcohol and sex steroids. These subjects were then searched in combination with the previous terms and the resulting papers manually examined. Theoretical, in vitro and in vivo research were all used to inform this review which focuses on the most well developed theoretical common causes for dementia(predominantly Alzheimer's type) and osteoporosis.RESULTS Dementia and osteoporosis are multifaceted disease processes with similar epidemiology and a marked increase in prevalence in elderly populations. The existence of a common link between the two has been suggested despite a lack of clear pathological overlap in our current understanding. Research to date has tended to be fragmented and relatively weak in nature with multiple confounding factors reflecting the difficulties of in vivo experimentation in the population of interest. Despite exploration of various possible mechanisms in search for a link between the two pathologies, this paper found that it is possible that these associations are coincidental due to the nature of the evidence available. One finding in this review is that prior investigation into common aetiologies has found raised amyloid beta peptide levels in osteoporotic bone tissue, with a hypothesis that amyloid beta disorders are systemic disorders resulting in differing tissue manifestations. Colorectal cancer（CRC）, a widespread malignancy, is closely associated with tumor microenvironmental hydrogen peroxide（H2O2） levels. Some clinical trials targeting H2O2for cancer treatment have revealed its paradoxical role as a promoter of cancer progression. Investigating the dynamics of cancer cell H2O2eustress at the single–cell level is crucial. In this study, non–contact hopping probe mode scanning ion conductance microscopy（HPICM） with high-sensitive Pt–functionalized nanoelectrodes was employed to measure dynamic extracellular to intracellular H2O2gradients in individual colorectal cancer Caco–2cells. We explored the relationship between cellular mechanical properties and H2O2gradients.Exposure to 0.1 or 1 mmol/L H2O2eustress increased the extracellular to intracellular H2O2gradient from 0.3 to 1.91 or 3.04, respectively. Notably, cellular f–actin–dependent stiffness increased at 0.1 mmol/L but decreased at 1 mmol/L H2O2eustress. This H2O2–induced stiffness modulated AKT activation positively and glutathione peroxidase 2（GPX2） expression negatively. Our findings unveil the failure of some H2O2-targeted therapies due to their ineffectiveness in generating H2O2, which instead acts eustress to promote cancer cell survival. This research also reveals the complex interplay between physical properties and biochemical signaling in cancer cells' antioxidant defense, illuminating the exploitation of H2O2eustress for survival at the single–cell level. Inhibiting GPX and/or catalase（CAT） enhances the cytotoxic activity of H2O2eustress against CRC cells, which holds significant promise for developing innovative therapies targeting cancer and other H2O2-related inflammatory diseases.

Background and Aims:High-intensity interval training(HIIT)is a therapeutic option for people with nonalcoholic steatohepatitis(NASH).However,the perspectives and experiences of HIIT for people with NASH are unknown,limiting translation of research.We explored the experiences and perspectives of both professionally supervised and self-directed HIIT in people with NASH and evaluated participant-reported knowledge,barriers,and enablers to commencing and sustaining HIIT.Methods:Twelve participants with NASH underwent 12 weeks of supervised HIIT(3 days/week,4×4 minutes at 85–95%maximal heart rate,interspersed with 3 minutes active recovery),followed by 12-weeks of selfdirected(unsupervised)HIIT.One-on-one,semistructured participant interviews were conducted by exercise staff prior to HIIT and following both supervised and self-directed HIIT to explore prior knowledge,barriers,enablers,and outcomes at each stage.Interviews were audio-recorded,transcribed,coded,and thematically analyzed by two independent researchers.Results:four dominant themes were identified:(1)no awareness of/experience with HIIT and ambivalence about exercise capabilities;(2)multiple medical and social barriers to commencing and continuing HIIT;(3)exercise specialist support was a highly valued enabler,and(4)HIIT was enjoyed and provided holistic benefits.Conclusions:People with NASH may lack knowledge of and confidence for HIIT,and experience multiple complex barriers to commencing and continuing HIIT.Exercise specialist support is a key enabler to sustained engagement.These factors need to be addressed in future clinical programs to augment the uptake and long-term sustainability of HIIT by people with NASH so they can experience the range of related benefits.

Disturbance cue communication is shaped by emitter diet and receiver background risk in Trinidadian guppies

In animal communication systems,individuals that detect a cue(i.e.,“receivers”)are often influenced by characteristics of the cue emitter.for instance,in many species,receivers avoid chemical cues that are released by emitters experiencing disturbance.These chemical“disturbance cues”appear to benefit receivers by warning them about nearby danger,such as a predator’s approach.While the active ingredients in disturbance cues have been largely unexplored,by-products of metabolized protein are thought to play a role for some species.If so,the content(quality)and volume(quantity)of the emitter’s diet should affect their disturbance cues,thus altering how receivers perceive the cues and respond.Guppies Poecilia reticulata are a species known to discriminate among disturbance cues from different types of donors,but dietary variation has yet to be explored.In this study,we found evidence that diet quality and quantity can affect disturbance cues released by guppy emitters(i.e.,experimental“donors”).Receivers discriminated between donor cue treatments,responding more strongly to cues from donors fed a protein-rich bloodworm diet(Experiment 1),as well as an overall larger diet(Experiment 2).We also found that receivers exposed to higher background risk were more sensitive to disturbance cue variation,with the strongest avoidance responses displayed by high-risk receivers toward disturbance cues from donors fed the high-quality diet.Therefore,diet,and perhaps protein specifically,affects either the concentration or composition of disturbance cues released by guppies.Such variation may be important in information signaling in social species like the guppy.

Effects of Acute Exercise on Verbal, Mathematical, and Spatial Insight Creativity

This experiment was designed to accommodate the diversified verbal,mathematical,and spatial skills of young adults,which have not yet to be collectively evaluated in research projects focused on acute exercise and creativity among college students.While emerging research suggests that acute moderate-intensity exercise may influence human creativity,creativity during and after exercise has not been experimentally investigated.Such differences are plausible,as previous work demonstrates that memory can be differentially influenced based on whether the memory task occurs during or after exercise.Therefore,the purpose of this study was to examine the effects of acute moderate‐intensity treadmill walking,for 15 min,on verbal,mathematical,and spatial insight creativity performance while considering the timing of the exercise and creativity tasks.Among a sample of 20 undergraduate students,all completed three randomized laboratory visits in this within-subjects design:control condition,insight problem-solving following exercise,and insight problem-solving during exercise.They also completed six insight creativity tasks(two verbal,two mathematical,and two spatial tasks)per visit,with the order of task-presentation randomized and counterbalanced across the three visits.Average insight creativity scores were similar across the three exercise manipulations:That is,verbal insight[f(2,18)=0.689,P=0.51],mathematical insight[f(2,18)=0.033,P=0.97],and spatial insight[f(2,18)=1.0,P=0.38]performance were not statistically significant across the three visits.Thus,moderate-intensity acute exercise may not appreciably influence verbal,mathematical or spatial insight creativity.

Effect of feeding broilers diets differing in susceptible phytate content

Measurements of total phytate phosphorus content of diets may be deceptive as they do not indicate substrate availability for phytase; it may be that measurements of phytate susceptible to phytase effects are a more accurate measure of phosphorus(P) availability to the bird. To verify this hypothesis, an experiment was conducted to compare diets formulated to contain either high or low susceptible phytate, supplemented with either 0 or 500 fTU/kg phytase. Susceptible phytate was determined by exposing the feed samples to conditions that mimicked the average pH of the proximal gastrointestinal tract(pH 4.5) and the optimum temperature for phytase activity(37℃) and then measuring phytate dissolved. Ross 308 birds(n = 240) were fed one of 4 dietary treatments in a 2 x 2 factorial design; 2 diets with high(8.54 g/kg, 57.90% of total phytate) or low(5.77 g/kg, 46.33% of total phytate) susceptible phytate, containing 0 or 500 fTU/kg phytase. Diets were fed to broilers(12 replicate pens of 5 birds per pen) from d 0 to 28 post hatch. Birds fed diets high in susceptible phytate had greater phytate hydrolysis in the gizzard(P fCR(P = 0.003) than birds fed the low susceptible phytate diets,irrespective of phytase presence. Birds fed the high susceptible diets also had greater P solubility in the gizzard and Ca and P solubility in the jejunum and ileum(P femur Ca and P(P fed the low susceptible diets. All the susceptible phytate was fully degraded in the tract in the absence of added phytase, suggesting the assay used in this study was able to successfully estimate the amount of total dietary phytate that was susceptible to the effects of phytase when used at standard levels. The adaptability of the central nervous system has been revealed in several model systems.Of particular interest to central nervous system-injured individuals is the ability for neural components to be modified for regain of function.In both types of neurotrauma,traumatic brain injury and spinal cord injury,the primary parasympathetic control to the gastrointestinal tract,the vagus nerve,remains anatomically intact.However,individuals with traumatic brain injury or spinal cord injury are highly susceptible to gastrointestinal dysfunctions.Such gastrointestinal dysfunctions attribute to higher morbidity and mortality following traumatic brain injury and spinal cord injury.While the vagal efferent output remains capable of eliciting motor responses following injury,evidence suggests impairment of the vagal afferents.Since sensory input drives motor output,this review will discuss the normal and altered anatomy and physiology of the gastrointestinal vagal afferents to better understand the contributions of vagal afferent plasticity following neurotrauma.

Landslide Hazard and Community-based Risk Reduction Effort in Karanganyar and the Surrounding Area,Central Java,Indonesia

Karanganyar and the surrounding area are situated in a dynamic volcanic arc region, where landslide frequently occurs during the rainy season. The rain-induced landslide disasters have been resulting in 65 fatalities and a substantial socioeconomical loss in last December 2007. Again, in early february 2009, 6 more people died, hundreds of people temporary evacuated and tens of houses damaged due to the rain-induced landslide. Accordingly, inter-disciplinary approach for geological, geotechnical and social investigations were undertaken with the goal for improving community resilience in the landslide vulnerable villages. Landslide hazard mapping and communitybased landslide mitigation were conducted to reduce the risk of landslides. The hazard mapping was carried out based on the susceptibility assessment with respect to the conditions of slope inclination, types and engineering properties of lithology/soil as well as the types of landuse. All of those parameters were analyzed by applying weighing and scoring system which were calculated by semi qualitative approach (Analytical Hierarchical Process). It was found that the weathered andesitic-steep slope (steeper than 30o) was identified as the highest susceptible slope for rapid landslide, whilst the gentle colluvial slope with inter-stratification of tuffaceous clay-silt was found to be the susceptible slope for creeping. finally, a programme for landslide risk reduction and control were developed with special emphasize on community-based landslide mitigation and early warning system. It should be highlighted that the social approach needs to be properly addressed in order to guarantee the effectiveness of landslide risk reduction.

High-throughput quantification of microbial birth and death dynamics using fluorescence microscopy

Background: Microbes live in dynamic environments where nutrient concentrations fluctuate. Quantifying fitness in terms of birth rate and death rate in a wide range of environments is critical for understanding microbial evolution and ecology. Methods^ Here, using high-throughput time-lapse microscopy, we have quantified how Saccharomyces cerevisiae mutants incapable of synthesizing an essential metabolite (auxotrophs) grow or die in various concentrations of the required metabolite. We establish that cells normally expressing fluorescent proteins lose fluorescence upon death and that the total fluorescence in an imaging frame is proportional to the number of live cells even when cells form multiple layers. We validate our microscopy approach of measuring birth and death rates using flow cytometry, cell counting, and chemostat culturing. Results: for lysine-requiring cells, very low concentrations of lysine are not detectably consumed and do not support cell birth, but delay the onset of death phase and reduce the death rate compared to no lysine. In contrast, in low hypoxanthine, hypoxanthine-requiring cells can produce new cells, yet also die faster than in the absence of hypoxanthine. for both strains, birth rates under various metabolite concentrations are better described by the sigmoidal-shaped Moser model than the well-known Monod model, while death rates can vary with metabolite concentration and time. Conclusions: Our work reveals how time-lapse microscopy can be used to discover non-intuitive microbial birth and death dynamics and to quantify growth rates in many environments.

Inhibition of Wnt signaling by frizzled7 antibody-coated nanoshells sensitizes triple-negative breast cancer cells to the autophagy regulator chloroquine

Nano Research volume 13,pages1693–1703(2020)Cite this article 137 Accesses 6 Altmetric Metrics details Abstract Despite improvements in our understanding of the biology behind triple-negative breast cancer(TNBC),it remains a devastating disease due to lack of an effective targeted therapy.Inhibiting Wnt signaling is a promising strategy to combat TNBC because Wnt signaling drives TNBC progression,chemoresistance,and stemness.However,Wnt inhibition can lead to upregulation of autophagy,which confers therapeutic resistance.This provides an opportunity for combination therapy,as autophagy inhibitors applied concurrently with Wnt inhibitors could increase treatment efficacy.Here,we applied the autophagy inhibitor chloroquine(CQ)to TNBC cells in combination with frizzled7 antibody-coated nanoshells(fZD7-NS)that suppress Wnt signaling by blocking Wnt ligand/fZD7 receptor interactions,and evaluated this dual treatment in vitro.We found that fZD7-NS can inhibit Axin2 and CyclinD1,two targets of canonical Wnt signaling,and increase the expression of LC3,an autophagy marker.When fZD7-NS and CQ are applied together,they reduce the expression of several stemness genes in TNBC cells,leading to inhibition of TNBC cell migration and self-renewal.Notably,co-delivery of fZD7-NS and CQ is more effective than either therapy alone or the combination of CQ with free fZD7 antibodies.This demonstrates that the nanocarrier design is important to its therapeutic utility.Overall,these findings indicate that combined regulation of Wnt signaling and autophagy by fZD7-NS and CQ is a promising strategy to combat TNBC.
