This study examined if the anti-tumorigenesis effect of Exendin-4 in HT29 and HCT116 colorectal cancer(CrC)involves modulation of SIrT1 and Akt/GSr3K/β-catenin/NF-κB ***29 and HCT116 cells were treated either with increasing levels of Exendin-4(0.0-200μM)or with Exendin-4(at its IC50)in the presence or absence of EX-527(10μM/a selective SIrT1 inhibitor)or Exendin-4(9-39)amide(E(9-39)A)(1μM/an Exendin-4 antagonist).In a dose-dependent manner,Exendin-4 inhibited cell survival,but enhanced levels of lactate dehydrogenase(LDH)and single-stranded DNA(ssDNA)in both HT29 and *** both cell lines and at it has an IC50(45μM for HT29 and 35μM for HCT1165),Exendin-4 also significantly reduced cell survival,migration,and invasion of both cell types,with no effect on the expression GLP-1 receptors(GLPrs)nor of the activity of *** these doses,Exendin-4 also increased the expression of SIrT1 but reduced the acetylation of NF-κB and the expression of Bax and cleaved caspase-3 and in both cell ***,protein levels of p-GS3Kβ(Ser9),total and acetylatedβ-catenin,and Anix2 were significantly decreased,but levels of p-GS3Kβ(Ser9)and p-β-catenin(Ser33/37/Thr41)were significantly increased in both HT29 and HCT116-exendin-4 treated *** the effects exerted by Exendin-4 were completely prevented by Ex527 or E(9-39)*** conclusion,Exendin-4 suppresses the tumorigenesis of HT29 and HCT116 CrC cell activation of GS3Kβ-induced inhibition ofβ-catenin and NF-κβin a SIrT1-dependent mechanism.

Due to the exponential growth of video data,aided by rapid advancements in multimedia *** became difficult for the user to obtain information from a large video *** process of providing an abstract of the entire video that includes the most representative frames is known as static video *** method resulted in rapid exploration,indexing,and retrieval of massive video *** propose a framework for static video summary based on a Binary robust Invariant Scalable Keypoint(BrISK)and bisecting K-means clustering *** current method effectively recognizes relevant frames using BrISK by extracting keypoints and the descriptors from video *** video frames’BrISK features are clustered using a bisecting K-means,and the keyframe is determined by selecting the frame that is most near the cluster *** applying any clustering parameters,the appropriate clusters number is determined using the silhouette *** were carried out on a publicly available open video project(OVP)dataset that contained videos of different *** proposed method’s effectiveness is compared to existing methods using a variety of evaluation metrics,and the proposed method achieves a trade-off between computational cost and quality.

The use of privacy-enhanced facial recognition has increased in response to growing concerns about data securityand privacy in the digital age. This trend is spurred by rising demand for face recognition technology in a varietyof industries, including access control, law enforcement, surveillance, and internet communication. However,the growing usage of face recognition technology has created serious concerns about data monitoring and userprivacy preferences, especially in context-aware systems. In response to these problems, this study provides a novelframework that integrates sophisticated approaches such as Generative Adversarial Networks (GANs), Blockchain,and distributed computing to solve privacy concerns while maintaining exact face recognition. The framework’spainstaking design and execution strive to strike a compromise between precise face recognition and protectingpersonal data integrity in an increasingly interconnected environment. Using cutting-edge tools like Dlib for faceanalysis,ray Cluster for distributed computing, and Blockchain for decentralized identity verification, the proposedsystem provides scalable and secure facial analysis while protecting user privacy. The study’s contributions includethe creation of a sustainable and scalable solution for privacy-aware face recognition, the implementation of flexibleprivacy computing approaches based on Blockchain networks, and the demonstration of higher performanceover previous methods. Specifically, the proposed StyleGAN model has an outstanding accuracy rate of 93.84%while processing high-resolution images from the CelebA-HQ dataset, beating other evaluated models such asProgressive GAN 90.27%, CycleGAN 89.80%, and MGAN 80.80%. With improvements in accuracy, speed, andprivacy protection, the framework has great promise for practical use in a variety of fields that need face recognitiontechnology. recently,nano-systems based on molecular communications via diffusion(MCvD)have been implemented in a variety of nanomedical applications,most notably in targeted drug delivery system(TDDS)***,because the MCvD is unreliable and there exists molecular noise and inter symbol interference(ISI),cooperative nano-relays can acquire the reliability for drug delivery to targeted diseased cells,especially if the separation distance between the nano transmitter and nano receiver is *** this work,we propose an approach for optimizing the performance of the nano system using cooperative molecular communications with a nano relay scheme,while accounting for blood flow effects in terms of drift *** fractions of the molecular drug that should be allocated to the nano transmitter and nano relay positioning are computed using a collaborative optimization problem solved by theModified Central Force Optimization(MCFO)*** the previous work,the probability of bit error is expressed in a closed-form *** is used as an objective function to determine the optimal velocity of the drug molecules and the detection threshold at the nano *** simulation results show that the probability of bit error can be dramatically reduced by optimizing the drift velocity,detection threshold,location of the nano-relay in the proposed nano system,and molecular drug budget.

This study investigates if the anti-tumor effect of Pterostilbene in the SKOV3 ovarian cancer(OC)cell line involves inhibition of cell metabolism and tested in this effect involves modulating AMPK and Akt-induced regulation of ***,SKOV3 cells were cultured in the humidified conditions in DMEM media for 24 h with or without increasing concentration of ***,the cells were incubated with Pterostilbene(IC_(50)=50μM)under similar conditions with or without pre-incubation with Dorsomorphin,an AMPK *** a dose-dependent manner,Pterostilbene inhibited SKOV3 cell survival and increased their lysate levels of lactate dehydrogenase(LDH)and single-stranded DNA(ssDNA).When SKOV3 cells were treated with 50μM Pterostilbene,Pterostilbene significantly suppressed cell migration and invasion,reduced lysate levels of lactic acid and the optical density of Oil red O staining,and increased lysate glucose *** also increased levels of malondialdehyde(MDA),reactive oxygen species(rOS),and induced intrinsic cell apoptosis by upregulating protein levels of Bax and cleaved caspase-3 and reducing protein levels of ***,Pterostilbene reduced mrNA levels of sterol regulatory element-binding protein 1(SrEBP-1),fatty acid synthase(FAS),acetyl CoA carboxylase-1(ACC-1),and AMP-activated protein kinase(AMPK).Furthermore,Pterostilbene increased the protein levels of p-AMPK,p-p53,p-raptor,p-TSC-2,but significantly decreased protein levels of p-Akt,p-TSC-2,p-mTOr,p-S6K1,and *** with Dorsomorphin(CC)abolished all the anti-tumorigenesis effects afforded by Pterostilbene and prevented Pterostilbene-induced phosphorylation of Akt,p53,and *** conclusion,the tumorsuppressive effect of Pterostilbene in SKOV3 cells involves the induction of rOS and inhibition of dysregulation cell metabolism mainly due to AMPK-induced Akt-dependent or independent suppression of mTOr.

Agmatine induces gastric protection against ischemic injury by reducing vascular permeability in rats

Preventive effects of cannabis on neurotoxic and hepatotoxic activities of malathion in rat

Objective: To investigate the effect of Cannabis sativa extract on the development of neuroand hepato-toxicity caused by malathion injection in rats. Methods: The extract of Cannabis sativa was obtained from the plant resin by chloroform treatment. Δ~9-Tetrahydrocannabinol content of the extract(20%) was quantified using gas chromatography–mass spectrometry. The doses of cannabis extract were expressed as Δ~9-tetrahydrocannabinol content of 10 or 20 mg/kg. Malathion(150 mg/kg) was intraperitoneally administered followed after 30 min by the cannabis extract(10 or 20 mg/kg, subcutaneously). rats were euthanized 4 h later. Malondialdehyde(MDA), reduced glutathione(GSH), nitric oxide and paraoxonase-1(PON-1) activity were determined in brain and liver. Brain 5-lipoxygenase and butyrylcholinesterase(BChE) activity were measured as well. Histopathological examination of brain and liver tissue was also performed. results: Compared to controls, malathion resulted in increased oxidative stress in brain and liver. MDA and nitric oxide concentrations were significantly increased(Preased with respect to control levels(Prain 5-lipoxygenase. Brain MDA concentrations were not altered by cannabis treatment. Cannabis at 20 mg/kg, however, caused significant increase in nitric oxide and restored the GSH and PON-1 activity. Brain BChE activity significantly decreased by 26.1%(Pr treatment with 10 mg/kg cannabis. Cannabis showed no effect on brain 5-lipoxygenase. On the other hand, rats treated with cannabis exhibited significantly higher levels of liver MDA, nitric oxide and PON-1 activity compared with the malathion control group. rats treated with only malathion exhibited spongiform changes, neuronal damage in the cerebral cortex and degeneration of some Purkinje cells in the cerebellum.
