Isoenzyme Expression in Bean Seed Germination Treated with Thiamethoxam with and without Drought Stress

The thiamethoxam acts as enhancer, allowing maximizing the expression of seed vigor. Isozymes are products of gene expression, highly influenced by the environment, because genes that control its expression are manifested in certain stages of development and in specific organs and tissues, or even under the action of certain stimulus. This study evaluated the isoenzyme expression in bean seedlings from seeds treated with thiamethoxam. Bean seeds were used, cultivar Pérola and IAPAR Siriri, submitted or not to drought stress. Seeds were treated at doses of thiamethoxam: 0, 1, 2, 3, 4 and 5 mL·kg-1 of seed. Isozymes were extracted from seedlings collected five days after sowing. We evaluated the expression of isoenzymes: Glutamate Oxalacetate transaminase (GOT), acid phosphatase (FAC), peroxidase (PO), Esterase (EST), malate dehydrogenase (MDH) and glutamate dehydrogenase (GTDH). Interpretation of the results was based on visual analysis of electrophoresis gels, considering the presence/absence and the intensity of each electrophoretic band. Bean seeds treated with thiamethoxam with doses up 3 mL·kg-1 of seed are higher germination untreated, even when submitted to drought stress. The expression of isozymes GOT, FAC, PO, EST, MDH and GTDH is not affected significantly in bean seedlings of Pérola and IPR Siriri under different doses of thiamethoxam with and without drought stress.

Sugarcane is used worldwide for sugar, ethanol and energy production. In Brazil, the shift from burned to unburned harvest systems resulted in increases in nitrogen fertilization rates, which can impact root architecture and biomass. The expectation is also an increase in sugarcane biomass. The study hypothesized that high n rates applied to sugarcane fields increases root growth and n stored in roots, promoting higher biomass and n accumulated in shoots. Two experiments were set up in Southeastern Brazil, on a Typic Kandiudox (TK) and Rhodic Eutrudox (RE). Four treatments were studied 1) n application in the plant-cane (0 and 120 kg·ha-1 n) and 2) n application in the ratoon (0 and 150 kg·ha-1 n). The shoot biomass and the root density (by the core method up to 0.6 m) were evaluated over the first ratoon crop cycle, and the n content in those compartments was also examined. There was no carry over effect on n applied at planting in root and shoot biomass in the ratoon crop cycle. At the RE site, the ratoon n fertilization increased root density in the superficial soil layer (0 - 0.2 m) and close to the plants (n addition on root biomass, and biomass and n accumulated in shoot was limited in both sites. Increasing n rates in unburned sugarcane fields do not consistently increases root and shoot biomass under Brazilian field conditions.

S53G SnP in Cathepsin B: Can Be Used as a Molecular Biomarker to Breast Cancer?

Cathepsin B (CTS B) is a proteolytic enzyme that participates in several important biological processes. However, when it is altered can be involved in development of breast cancer, a disease with high incidence and mortality rate among women. Many studies have shown a correlation between high CTS B expression and breast tumor. Furthermore, it has been shown that the SnP rs1803250 (S53G) leads to structural and functional changes in protein that can make it pathogenic. The present study aimed to evaluate a possible association of SnP rs1803250 (S53G) with breast cancer. For this, real-time PCR was performed on a sample collected in the State of Pará, with 127 patients and 122 controls. The SnP frequency in this region was 0.12, according to a research project in progress that aims to identify Amerindians molecular alterations. This indicates that the SnP is found in region with a distribution close to worldwide frequency of 0.09. Our results showed that the SnP was in Hardy-Weinberg Equilibrium in collected sample, but C variant allelic frequency was 0.08 in both patients and control groups, which is extremely similar to global average. Moreover, homozygotes CC was not found in the sample and SnP genotypes frequency in patients and control groups was not significantly different. In addition, statistical analysis showed that the SnP did not have to correlate with tumor subtypes nor with tumor staging. Therefore, according to the analyzed sample, the SnP rs1803250 has no association with breast cancer and it cannot be considered a molecular biomarker for breast cancer.

Pattern of Development in Foals from Four Different Breeds between Birth and Weaning

Several studies have shown a major development in height and cannon bone circumference in foals from birth to weaning and from different breeds. However, in Brazil, there is little information about foal development. For this reason, this study has used data on foals from seven different farms in Pernambuco, northeast Brazil, where horses have a major economic importance. The aim of this study was to compare foal’s development from four breeds and to develop a regression formula in order to estimate such development. It was used data from 210 suckling foals (Arabians n = 18, Campolina n = 86, Mangalarga Marchador n = 42 and Quarter Horses n = 64), raised in similar managem.nt practices. Their height at withers, heart girth, and cannon bone girth were measured monthly from birth to weaning. data we reanalyzed by AnOVA and Tukey Test, with P established in 5%. The highest mean of height at withers was observed in the Arabian foals, and it was similar to Campolina and Mangalarga Marchador foals. The last breed presented a higher percentage of gain in height (~36%) and higher heart girth. On the other hand, Campolina foals had the highest mean of cannon bone girth. There was a significant interaction between breed and development phase in heart and cannon bone girths. All regression for mulas obtained from this data was very significant, and it was showed that they can predict height at withers in the four breeds studied. Heart and cannon bone girths showed medium value of R2 and large significance. It is important to highlight that fetal programing produces significant impact on foal development, even after weaning. So, this study would have observed some more changes if its scope had been extended beyond this phase.

AIM To investigate the association between 16 insertiondeletions(InDEL) polymorphisms, colorectal cancer(CRC) risk and clinical features in an admixed *** O n e h u n d re d a n d fo r ty p a t i e n t s w i t h C R C a n d 140 cancer-free subjects were examined. Genomic DnA was extracted from peripheral blood samples. Polymorphisms and genomic ancestry distribution were assayed by Multiplex-PCR reaction, separated by capillary electrophoresis on the ABI 3130 Genetic Analyzer instrument and analyzed on Gene Mapper ID v3.2. Clinicopathological data were obtained by consulting the patients' clinical charts, intra-operative documentation, and pathology *** Logistic regression analysis showed that polymorphism variations in IL4 gene was associated with increased CRC risk, while TYMS and UCP2 genes were associated with decreased risk. Reference to anatomical localization of tumor Del allele of nFKB1 and CASP8 were associated with more colon related incidents than rectosigmoid. In relation to the InDEL association with tumor node metastasis(TnM) stage risk, the Ins alleles of ACE, HLAG and TP53(6 bp InDEL) were associated with higher TnM stage. Furthermore, regarding InDEL association with relapse risk, the Ins alleles of ACE, HLAG, and UGT1A1 were associated with early relapse risk, as well as the Del allele of TYMS. Regarding InDEL association with death risk before 10 years, the Ins allele of SGSM3 and UGT1A1 were associated with death *** The InDEL variations in ACE, UCP2, TYMS, IL4, nFKB1, CASP8, TP53, HLAG, UGT1A1, and SGSM3 were associated with CRC risk and clinical features in an admixed population. These data suggest that this cancer panel might be useful as a complem.ntary tool for better clinical managem.nt, and more studies need to be conducted to confirm these findings.

In order to analyze crime hot spots, we use Kernel estimation. The choice of Kernel function and Band-width is critical in kernel density estimation, which decides the accuracy of the estimation. We choose Gauss kernel and further obtain the optimal Band-width in the sense of square error MISE. Using Kernel estimation, not only can we calculate the density of crime in the region, but also accurately show the areas with the relative high-crime density and get the maximum point according to the information about the previous criminal spots. Last we use Kernel estimation to predict Peter Sutcliffe "the Yorkshire Ripper" 11th criminal location based on the previous criminal locations in the Serial murders. Finally we can get the range of the criminal hot zone: Longitude: 53.6875n - 53.8125n;Altitude: 1.775W - 1.815 W. In fact, the coordinate of Peter’s 11th criminal loca tion is （53.817n, 1.784W）.From this, it can be seen that our estimation is relatively accurate.
