Non-redundant Contribution of the Plastidial FAD8 ω-3 Desatumse to Glycerolipid Unsatumtion at Different Temperatures in Arabidopsis

Plastidial ω-3 desaturase FAD7 is a major contributor to trienoic fatty acid biosynthesis in the leaves of Arabidopsis plants. However, the precise contribution of the other plastidial ω-3 desaturase, FAD8, is poorly understood. Fatty acid and lipid analysis of several ω-3 desaturase mutants, including two insertion lines of AtFAD7 and AtFAD8, showed that FAD8 partially compensated the disruption of the AtFAD7 gene at 22℃, indicating that FAD8 was active at this growth temperature, contrasting to previous observations that circumscribed the FAD8 activity at low temperatures. Our data revealed that FAD8 had a higher selectivity for 18：2 acyl-lipid substrates and a higher preference for lipids other than galactolipids, particularly phosphatidylglycerol, at any of the temperatures studied. Differences in the mechanism controlling AtFAD7 and AtFAD8 gene expression at different temperatures were also detected. Confocal microscopy and biochemical analysis of FAD8-YFP over-expressing lines confirmed the chloroplast envelope localization of FAD8. Co-localization experiments suggested that FAD8 and FAD7 might be located in close vicinity in the envelope membrane. FAD8-YFP over-expressing lines showed a specific increase in 18：3 fatty acids at 22℃. Together, these results indicate that the function of both plastidial ω-3 desaturases is coordinated in a non-redundant manner.

Syncope is a concerning symptom that affects a large proportion of *** can be related to a heterogeneous group of pathologies ranging from trivial causes to diseases with a high risk of sudden ***,benign causes are the most frequent,and identifying high-risk patients with potentially severe etiologies is crucial to establish an accurate diagnosis,initiate effective therapy,and alter the *** term cardiac syncope refers to those episodes where the cause of the cerebral hypoperfusion is directly related to a cardiac disorder,while arrhythmic syncope is cardiac syncope specifically due to rhythm ***,arrhythmias are the most common cause of cardiac *** bradyarrhythmia and tachyarrhythmia can cause a sudden decrease in cardiac output and produce *** this review,we summarized the main guidelines in the management of patients with syncope of presumed arrhythmic ***,we presented a thorough approach to syncope work-up through different tests depending on the clinical characteristics of the patients,risk stratification,and the management of syncope in different scenarios such as structural heart disease and channelopathies.

S-adenosyl-L-methionine modifies antioxidant-enzymes,glutathione-biosynthesis and methionine adenosyltransferases-1/2 in hepatitis C virus-expressing cells

AIM: To elucidate the mechanism(s) by which S-adenosyl-L-methionine(SAM) decreases hepatitis C virus(HCV) ***: We examined the effects of SAM on viral expression using an HCV subgenomic replicon cell culture system. Huh7 HCV-replicon cells were treated with 1 mmol/L SAM for different times(24-72 h), then total RNA and proteins were isolated. c DNA was synthesized and real time-PCR was achieved to quantify HCV-RNA, superoxide dismutase 1 and 2(SOD-1, SOD-2) catalase, thioredoxin 1, methionine adenosyltransferase 1A and 2A(MAT1A, MAT2A) expression, and GAPDH and RPS18 as endogenous genes. Expression of cellular and viral protein was evaluated by western-blot analysis using antibodies vs HCV-NS5 A, SOD-1, SOD-2, catalase, thioredoxin-1, MAT1 A, MAT2 A, GAPDH and actin. Total glutathione levels were measured at different times by Ellman's recycling method(0-24 h). Reactive oxidative species(ROS) levels were quantified by the dichlorofluorescein assay(0-48 h); Pyrrolidin dithiocarbamate(PDTC) was tested as an antioxidant control and H2O2 as a positive oxidant ***: SAM exposition decreased HCV-RNA levels 50%-70% compared to non-treated controls(24-72 h). SAM induced a synergic antiviral effect with standard IFN treatment but it was independent of IFN signaling. In addition, 1 mmol/L SAM exposition did not modify viral RNA stability, but it needs cellular translation machinery in order to decrease HCV expression. Total glutathione levels increased upon SAM treatment in HCV-replicon cells. Transcriptional antioxidant enzyme expression(SOD-1, SOD-2 and thioredoxin-1) was increased at different times but interestingly, there was no significant change in ROS levels upon SAM treatment, contrary to what was detected with PDTC treatment, where an average 40% reduction was observed in exposed cells. Along 24 years, our team in the University of Los Andes, Mérida-Venezuela, has developed several softwares intended to have educational applications in medicine. All of them were just prototypes and were never completed. We have always thought that information systems have a very important place in education and that in the future they would have a leading role. We have been thinking that the future paradigm of education is based on informatics, communications and their applications. Nevertheless, our insight and autocriticism has open new windows, points of view and approaches of this "new paradigm" and some questions have invaded our minds. Metaphorically speaking, are information systems applications a teacher, a model, a way, a mean, a goal? Or;might it be just a sort of a tool, comparable with a book;or even more dispensable ones like a pencil, a piece of paper, an eraser or a pencil-sharpener? In this paper we will briefly present some of our products(information systems), their features, applications, rationale, some difficulties that we have encountered while introducing them. We will describe the evolution of the new ideas and developments that we incorporated and finally we will discuss some of the thoughts that have emerged. Answers to these questions might be given by the readers.

Morphometric Comparison of White Tambaqui （Piaractus brachypomus） in Lentic and Lotic Environments

The purpose of this paper is looking for associations between environmental factors and morphological parameters in tambaqui （Piaractus brachypomus） individuals to differentiate this species in lentic environments （lake） and lotic （river）. In this, regard studied 30 specimens, 15 from lentic environment （lake） and 15 from lotic （river）. Also, on right profile of these 25 morphometric variables were measured. On data matrix a PCA （principal components analysis） based on morphometric correlations matrix, which was defined in the new morphologic space of these specimens （3 principal components） which explain 73.23% of variability. Fish projection in first two principal components showed a morphological differentiation between two environments （lentic and lotic）, with variables as, horizontal eye diameter, length maxilla, suggesting greater response of these fish in lotic environment by their adaptation to light conditions, predators threat and food distribution. Finally, in lotic environment these fish have thinner caudal peduncle, indicating greater plasticity, namely stylized fish.

Natural regeneration dynamics of three dry deciduous forest species in Chacocente Wildlife Reserve, Nicaragua

A study was conducted to examine the natural regeneration of three dry forest species, Lysiloma divaricatum （Jacq.）, Tabebuia ochracea （Cham.） and Lonchocarpus minimiflorus （Donn. Sin.） over a three-year period and to analyze heterogeneity of regeneration in relationship to topographic slope and incidence of light in Chacocente Wildlife Reserve in Nicaragua. Permanent sample plots were established in 2001, and all individuals with height as low as 10 cm and diameter as large as 10 cm were recorded for three consecutive years from 2001 to 2003. The results show that the density of naturally regenerated individuals varied significantly among species, as well as over time. L. minimiflorus and T. ochracea had higher densities compared with L. divaricatum, and the net change in population density was slightly positive for T. ochracea only. Regeneration was more abundant in the gentle and steep slopes under partial exposure of the crown to sunlight, thus resulting in aggregated pattern of distribution, especially for L. minimiflorus and T. ochracea. We concluded that natural regeneration alone is not sufficient to maintain the desired number of stocks of these species, and an immediate restoration measure should be taken to assist the natural regeneration process.

Comparative study of the efficacy and safety of bromfenac, nepafenac and diclofenac sodium for the prevention of cystoid macular edema after phacoemulsification

AIM： To compare the efficacy, tolerability and safety of bromfenac 0.09%, nepafenac 0.1% or diclofenac 0.1% for the prophylaxis of the cystoid macular edema（CME） after phacoemulsification. METHODS： Group sequential observational comparative study. After phacoemulsification, patients received two months for topical treatment of either diclofenac sodium, bromfenac or nepafenac. All patients received concomitant topical tobramycin 0.3% and topical prednisolone 1%. We measured CME using optical coherence tomography（OCT） central foveal thickness, macular thickness and total macular volume. RESULTS： We enrolled 243 patients from January to June 2015, and 35% received diclofenac, 32.9% bromfenac and 32.1% nepafenac. When we compared pre-operative to three weeks to two months, bromfenac was more effective in reducing foveal volume（21.3 and 35.4 mm3, respectively）, compared with the diclofenac（1.3 and 11.5 mm3, respectively）, and the nepafenac group, became more edematous 6.4 and 5.3, respectively. Totally 133 patients completed the post-surgical satisfaction questionnaire. Patients complained of eye stickiness in 13.8% whom we gave nepafenac, versus 10.3% whom we gave diclofenac sodium, and in 0 whom we gave bromfenac. CONCLUSION： Bromfenac is the best tolerated and is more effective than diclofenac and nepafenac in reducing CME after phacoemulsification.

Human CD4 ＋ effector T lymphocytes generated upon TCR engagement with self-peptides respond defectively to IL-7 in their transition to memory cells

The peripheral repertoire of CD4＋ T lymphocytes contains autoreactive cells that remain tolerant through several mechanisms. However, nonspecific CD4＋ T cells can be activated in physiological conditions as in the course of an ongoing immune response, and their outcome is not yet fully understood. Here, we investigate the fate of human naive CD4＋ lymphocytes activated by dendritic cells （DCs） presenting endogenous self-peptides in comparison with lymphocytes involved in alloresponses. We generated memory cells （Tmem） from primary effectors activated with mature autologous DCs plus interleukin （IL）-2 （Tmauto）, simulating the circumstances of an active immune response, or allogeneic DCs （Tmallo）. Tmem were generated from effector cells that were rested in the absence of antigenic stimuli, with or without IL-7. Tmem were less activated than effectors （demonstrated by CD25 downregulation） particularly with IL-7, suggesting that this cytokine may favour the transition to quiescence. Tmauto and TmaHo showed an effector memory phenotype, and responded similarly to polyclonal and antigen-specific stimuli. Biochemically, IL-7-treated Tma^o were closely related to conventional memory lymphocytes based on Erk-l/2 activation, whereas Tmauto were more similar to effectors. Autologous effectors exhibited lower responses to IL-7 than allogeneic cells, which were reflected in their reduced proliferation and higher cell death. This was not related to IL-7 receptor expression but rather to signalling deficiencies, according to STAT5 activation These results suggest that ineffective responses to IL-7 could impair the transition to memory cells of naive CD4＋ T lymphocytes recognizing self-peptides in the setting of strong costimulation.

本论文是以翻译报告做为主要任务,更具体地说,是著名作家兼翻译家傅雷在其著作《傅雷家书》中使用简单过去时（pretérito perfecto simple）以及未完成过去时（pretérito imperfecto del indicativo）的用法。进行中译西翻译的过程中最大困难之一就是准确使用西班牙文的简单过去时以及未完成过去时,这是因为汉语表达这两种时态的方式跟西班牙文很不一样,因此经常会引起疑问或歧义。本研究参考了先前研究之后决定将康苏尔洛·马科斯·马丁内（Consuelo Marcos Martínez）博士和兰教授以及其他研究员的结论作为该论文的理论框架。在分析译文的过程中,发现若翻译员以该论文所提及的理论框架来进行翻译任务,他将能做出一个较准确的选择,但也同时发现在很多情况下这两种时态是可互换的。因此,奥尔特加教授与卡斯塔涅达教授（Ortega y Casta（?）eda）的结论能为两者之间的细微差别提供理论和见解。
