Laser capture microdissection and genetic analysis of carbon-labeled Kupffer cells

AIm： To develop a method of labeling and microdissecting mouse Kupffer cells within an extraordinarily short period of time using laser capture microdissection （LCm）. mETHODS： Tissues are complex structures comprised of a heterogeneous population of interconnected cells. LCm offers a method of isolating a single cell type from specific regions of a tissue section. LCm is an essential approach used in conjunction with molecular analysis to study the functional interaction of cells in their native tissue environment. The process of labeling and acquiring cells by LCm prior to mRNA isolation can be elaborate, thereby subjecting the RNA to considerable degradation. Kupffer cell labeling is achieved by injecting India ink intravenously, thus circumventing the need for in vitro staining. The significance of this novel approach was validated using a cholestatic liver injury model. RESULTS： mRNA extracted from the microdissected cell population displayed marked increases in colonystimulating factor-1 receptor and Kupffer cell receptor message expression, which demonstrated Kupffer cell enrichment. Gene expression by Kupffer ceils derived from bile-duct-ligated, versus sham-operated, mice was compared. microarray analysis revealed a significant （2.5-fold, q value 〈 10） change in 493 genes. Based on this fold-change and a standardized Pubmed search, 10 genes were identified that were relevant to the ability of Kupffer cells to suppress liver injury. CONCLUSION; The methodology outlined herein provides an approach to isolating high quality RNA from Kupffer cells, without altering the tissue integrity.

This Opinion piece is offered as a cursory overview of sperm development,function, and transport through the eyes of an equine veterinarian. my professional background is predominantly clinical in nature, but my fascination with sperm function and preservation has led to a fairly sizeable review of the scientific literature over the years in hopes of extracting laboratory findings that have application to my daily activities in the clinical arena. Spermatozoa are quite unique among cellular types with regard to both form and function, and represent the only endogenously derived cell type that exerts its action in a separate being. This paper takes the reader on a voyage with a mammalian spermatozoon, from its formative stages through its transport in the male and female reproductive tracts, and culminating with its interaction with an ovulated oocyte at the time of fertilization. Specific emphasis is placed on equine spermatozoa when notable research findings have been unveiled.

Determinants and rates of land degradation： Application of stationary time-series model to data from a semi-arid environment in Kenya

The causes of land degradation in the African drylands have been shown to vary. Some researchers consider climate to be the major contributor to degradation, with anthropogenic factors playing a minor role. Others reverse the significance of these two factors. A third group attributes land degradation to climate and anthropogenic factors equally. This study was undertaken to establish the factors influencing land degradation in a semi-arid environment in southeastern Kenya and the rate of change in vegetation types for a period of 35 years （1973-2007）. The reduction in grassland cover was used as an indicator of land degradation. Causes of land degradation were determined by a multiple regression analysis. A log-linear regression analysis was used to establish the rate of vegetation change. The multiple and log-linear regression analyses showed：（1） woody vegetation, livestock population and cultivated area to be the main contributors of reduction in grassland cover in the area, and （2） an increase in undesirable woody species, livestock population and cultivated area had a significant （P〈0.05） negative effect on grassland vegetation. Increased human population, low amounts of rainfall and drought showed no significant negative effect on grassland vegetation cover. In conclusion, human and livestock population growth and increased agricultural land have contributed to intensive crop cultivation and overgrazing in the semi-arid lands. This overuse of the semi-arid rangelands has worsened the deterioration of the natural grassland vegetation.

Tumour induced osteomalacia due to a sinonasal hemangiopericytoma:A case report

Tumour induced osteomalacia(TIO) is a rare and often unrecognized cause of hypophosphatemia. We report on a case of TIO due to a hemangiopericytoma originating from the left nasal sinus. The patient was a 55-year-old male with a 3-year history of left hip pain and an undisplaced left hip fracture. Biochemical testing demonstrated low levels of serum phosphate and serum 1,25-dihydroxyvitamin D, and an elevated level of fibroblast growth factor 23. Octreotide scanning demonstrated uptake in the left nasal sinus area and a computed tomography scan revealed a left nasal sinus mass. The patient underwent surgical resection of the mass and histology was consistent with a sinonasal hemangiopericytoma. His serum phosphate levels normalized almost immediately after surgery and he had complete resolution of hip pain. Our case highlights the importance of considering TIO when assessing patients with low serum phosphate.

The Influence of Advanced Paternal Age on Sperm Chromatin Integrity and Early Embryo morphological Development during ICSI

The study determined the impact of advanced male ageing (≥50 years) on sperm chromatin integrity and early embryo morphological development in intra-cytoplasmic sperm injection (ICSI) technique cycles. Six hundred subfertile men were age-grouped;X1 (50 to 59 years), X2 (60 to 69), and X3 (≥70), were compared with 600 fertile males of known fertility (Y, age 25 - 35 years). Oocytes from 254 women, aged ≤ 30 years, were included. Sperm were analyzed using acridine orange fluorescence test (AOT) and categorized: “low”, “inter-mediate” and “high” damage. After ICSI, embryos were evaluated and categorized as “good”, “fair” or “poor” quality. Embryonic morphological development was assessed at three stages: fertilization, early and late paternal effect. The AOT results were: X1: low = 29, intermediate = 53 and high = 118;X2: low = 11, intermediate = 42 and high = 147;X3: low = 8, intermediate = 24 and high = 168;Y: Low = 486, intermediate = 71 and high = 43. The fertilization rate was: X1, 329/350 (93.7%);X2, 298/350 (85.1%);X3, 225/350 (64.1%) and, Y, 350/350 (100%). Associations between increasing age and sperm chromatin damage (χ2 (723.249, 6) p morphologically poor quality embryos over the five days after fertilization. male age ≥ 50 years, is highly associated with abnormal sperm chromatin organization, an inability to adequately fertilize with ICSI methodology, an increase in the number of poor quality embryos and, a corresponding decrease in the number of good quality embryos five days after fertilization.

Assessing Water Availability and Unmet Water Demand Using the WEAP model in the Semi-Arid Bweengwa, Kasaka and magoye Sub-Catchments of Southern Zambia

Located in the semi-arid zone of Zambia, the mutama-Bweengwa, Kasaka and magoye sub-catchments have witnessed a high demand for water due to increase in population and socio-economic activities putting more pressure on water resources. This study assesses the hydrological components and ascertains the available water resources and unmet demand in the sub-catchments using the Water Evaluation And Planning (WEAP) model and hydrometeorological data collected between 1951 and 2018. The model was calibrated and validated on 1971-1981 and 2008-2018 data respectively. The results reveal that the sub-catchments have transitioned from positive to negative water balance with -164.295 mm3/year for mutama-Bweengwa, -19.021 mm3/year for Kasaka and -86.368 mm3/year for magoye. Evaporation was 1815.259 mm3/year for mutama-Bweengwa, 1162.655 mm3/year for Kasaka and 1505.664 mm3/year for magoye. The demand for water has been increasing over time for various purposes such as irrigation, domestic, urban/rural water supply and livestock. The overall water storage in the sub-catchments showed a negative water balance for the year 2018. The observed and simulated peak streamflow were 8.16 m3/s and 7.7 m3/s occurring during the month of January and February respectively. The WEAP model performance achieved R2 of 0.98 during calibration and 0.95 for validation, and an NSE of 0.83 for calibration and 0.85 during validation. The values of objective functions show that the hydrology of the mutama-Bweengwa, Kasaka and magoye sub-catchments as predicted by the WEAP model provides satisfactory confidence for prediction of future streamflow and hence projection based on future scenarios.

Development of Extended-Release Formulation of Ibuprofen Using Blends of Calcium Silicate and Polyvinyl Pyrrolidone as Tablet matrix

A tablet matrix system was developed for ibuprofen and the influence of the polymer blend and concentration on the release rate of the drug was *** containing different concentrations of calcium silicate and PVP(polyvinyl pyrrolidone)were prepared using direct compression and the weight uniformity,crushing strength,friability,drug content uniformity,dissolution profile,and in vitro release kinetics were *** tablets were found to be within the official acceptable limits of physical and chemical parameters except for the thickness test that was below the conformation of extended-release *** crushing strength of the tablets was in the range of 2.5 to 5.6 kg/f,the weight variations of the tablets of all the formulation was less than±5%.The friability of all the formulations was in the range of 0.6%to 1.83%.Tablet thickness and diameter was in range of 3.18 mm to 4.48 mm and 12.53 mm to 12.64 mm *** drug contents of all the formulations were found to be in range of 83.50%to 98%.The release kinetic of F3 containing 20 mg of calcium silicate,40 mg of PVP as matrix formers showed the best linearity(r^2=0.6975)with%drug release of 96 showing that combination of the two polymers(20 mg calcium silicate and 40 mg PVP)for use as a matrix former is best for extended-release formulation of ibuprofen.
