AIM To investigate the use of droplet digital polymerase chain reaction(dd PCR) for detecting host m RNA markers in stools as a non-invasive test for colorectal cancer *** dd PCR and quantitative PCR were compared side by side for their performance in the detection of ITGA6 and ITGA6 A transcripts in stool samples obtained from patients with various types of colorectal lesions(advanced adenomas and stage Ⅱ-Ⅳ colorectal cancers) and control(patients displaying no pathological findings) using duplex Taq Man reactions for both methods. ITGA6 and ITGA6 A were chosen for this proof-of-concept study based on their relative medium and low abundance in stool samples, respectively, as established in a previous *** We found that the dd PCR and q PCR methods per-formed equally well in this Taq Man duplex assay for the detection of ITGA6 and ITGA6 A transcripts in stools of patients with colorectal lesions. For ITGA6, receiver operating characteristic(ROC) curve analysis showed comparable areas under the curve of 0.91(P < 0.0001) and 0.89-0.90(P < 0.0001) for the prediction of advanced adenomas and colorectal cancers, respectively. ITGA6 A, which was detected at very low levels in control patients, was found to be significantly elevated(over 40 times) in stage Ⅱ and Ⅲ colorectal cancers(P < 0.0002). Comparison of the two sets of data revealed a strong correlation of the copy numbers obtained by dd PCR and q PCR for both ITGA6 and ITGA6 *** We found that ITGA6 and ITGA6 A detection in stools of patients with colorectal cancers with dd PCR is comparable to that of q PCR using Taq Man assays.

Some physical,frictional and thermal properties of Brachystegia eurycoma seed were studied at different moisture contents in order to explore the possibility of mechanizing the handling and processing *** showed that the seed could be considered an oval disc in *** the moisture range of 2.79%to 27.13%(d.b.),the major,intermediate and minor axial dimensions increased with increase in moisture content from 2.29 mm to 2.45 mm,1.65 mm to 1.91 mm and 0.34 mm to 0.52 mm,*** the above moisture range,one thousand seed weight,particle density,porosity,roundness,sphericity,surface area and angle of repose increased linearly from 0.901 kg to 1.252 kg,2270 kg/m^(3) to 2520 kg/m^(3),11.23%to 15.46%,35%to 47%,67%to 82%,7.67 cm^(2) to 8.48 cm^(2) and 16.8°to 29.2°respectively,while bulk density decreased from 745.4 kg/m^(3) to 613.6 kg/m^(3).Static coefficient of friction on different structural surfaces increased linearly with moisture content and had the highest values on galvanized steel sheet(0.445-0.639),and the lowest values on fiber glass(0.287-0.404).Kinetic coefficient of friction increased linearly with moisture content on different structural surfaces except on galvanized steel sheet where it decreased linearly with increase in moisture content in the above moisture *** had the highest values on hessian bag material(0.266-0.389),and the lowest values on plywood with wood grains perpendicular to the direction of movement(0.204-0.271).Specific heat ranged from 1474 to 5992.7 j/kgK,and increased with increase in moisture content and *** equations were used to express the relationships existing between the seed properties and moisture content.

BACKGROUND Sleep disturbance is a core feature of post-traumatic stress disorder(PTSD).Given the relationship between sleep disturbance and PTSD,there has been a relative paucity of studies examining the potential therapeutic impact of using pharmacotherapy to target sleep disturbance in patients with ***(ESZ)is a non-benzodiazepine y-aminobutyric acid-A receptor agonist indicated for the treatment of sleep and may affect sleep in patients with *** To evaluate the efficacy of ESZ vs placebo(PBO)for patients with PTSD and *** The study was a 12-wk,double blind,randomized controlled trial with 3 mg of ESZ(n=13)or PBO(n=12).RESULTS Patients in both arms experienced significant improvement in PTSD symptoms as assessed by the Clinician-Administered PTSD Scale for DSM-IV(CAPS):ESZ(t11=-3.12,P=0.005)and PBO(t11=-3.5,P=0.002)and by self-report with the Short PTSD Rating Interview(ESZ t11=-3.38,P=0.003 and PBO t11=-4.48,P=0.0005).There were no significant differences between treatments on the CAPS(t22=-0.13,P=0.70)or the Short PTSD Rating Interview(t22=-0.58,P=0.56).Similarly,both treated groups improved on sleep measures as assessed by the Pittsburgh Sleep Quality Index with PTSD Addendum(PSQI)and on total sleep time(TST)and sleep latency assessed by actigraphy with no significant differences between groups(PSQI t22=-0.24,P=0.81;total sleep time t10=0.13,P=0.90 and sleep latency t10=0.68,P=0.50).There was a significant correlation between improvement in sleep and overall improvement in PTSD as measured by change scores on the PSQI and CAPS,r(8)=0.79,P=0.01 for ESZ treated subjects,but not for those treated with PBO r(9)=0.16,P=*** events of ESZ were consistent with the known profile of the medication including dysgeusia(30%,mild),sedation(20%,mild)and headache(20%,moderate to severe).CONCLUSION Results do not support the hypothesis of a specific positive effect of ESZ compared to PBO for measures of PTSD and associated sleep disturban

The G-quadruplex （G4） elements comprise a class of nucleic acid structures formed by stacking of guanine base quartets in a quadruple helix. This （34 DNA can form within or across single-stranded DNA molecules and is mutually exclusive with duplex B-form DNA. The reversibility and structural diversity of G4s make them highly versatile genetic structures, as demonstrated by their roles in various functions including telomere metabolism, genome maintenance, immunoglobulin gene diversification, transcription, and translation. Sequence motifs capable of forming G4 DNA are typically located in telomere repeat DNA and other non-telomeric genomic loci. To investigate their potential roles in a large-genome model plant species, we computationaily identified 149,988 non-telomeric G4 motifs in maize （Zea mays L., B73 AGPv2）, 29% of which were in non-repetitive genomic regions. G4 motif hotspots exhibited non-random enrichment in genes at two locations on the antisense strand, one in the 5~ UTR and the other at the 5~ end of the first intron. Several genic G4 motifs were shown to adopt sequence-specific and potassium-dependent G4 DNA structures in vitro. The G4 motifs were prevalent in key regulatory genes associated with hypoxia （group VII ERFs）, oxidative stress （D j-1/GATasel）, and energy status （AMPK/ SnRK） pathways. They also showed statistical enrichment for genes in metabolic pathways that function in glycolysis, sugar degradation, inositol metabolism, and base excision repair. Collectively, the maize G4 motifs may represent conditional regulatory elements that can aid in energy status gene responses. Such a network of elements could provide a mechanistic basis for linking energy status signals to gene regulation in maize, a model genetic system and major world crop species for feed, food, and fuel.

Background: There is growing evidence to support the beneficial effects of supplementing direct-fed microbials(DFM) on performance, health status, and immune responses of weaned pigs. Therefore, the objective of this study was to investigate dietary supplementation of Bacillus subtilis(DSM 25841) on growth performance, diarrhea, gut permeability and immunity of weaned pigs experimentally infected with a pathogenic F-18 Escherichia coli(E. coli).Results: The F18 E. coli infection reduced(P jejunum compared with non-challenged control. Supplementation of Bacillus subtilis linearly enhanced average daily gain of E. coli infected pigs from d 0 to 5 post-inoculation(PI)(P jejunal mucosa when pigs were fed with the control diet. Supplementation of Bacillus subtilis linearly up-regulated(P jejunal mucosa of E. coli infected pigs. In addition, E. coli infection increased(P < 0.05) the m RNA expression of several immune genes(IL1 A, IL1 B, and IL7 on d 5 PI, and IL1 B, IL6, IL7, and TNF on d 11 PI) in the ileal mucosa of weaned pigs. Cholangiopathies are a group of diseases primarily or secondarily affecting bile duct cells, and result in cholangiocyte proliferation, regression, and/or transformation. Their etiopathogenesis may be associated with a broad variety of causes of different nature, which includes genetic, neoplastic, immune-associated, infectious, vascular, and drug-induced alterations, or being idiopathic. miRNAs, small non-coding endogenous RNAs that post-transcriptionally regulate gene expres sion, have been associated with pathophysiological processes in different organs and cell types, and are postulated as potential targets for diagnosis and therapy. In the current manuscript, knowledge regarding the role of miRNAs in the development and/or progression of cholangiopathies has been reviewed and the most relevant findings in this promising field of hepatology have been highlighted.

Dendrosenecio brassiciformis(菊科、巨大千里光属)为直立、一次结实植物;局限性分布于肯尼亚阿伯德尔山脉：常与Lobelia telekii和Lobelia aberdarica构成混生居群。该物种具有许多特殊的表型：茎干中心储水、巨大的莲座状叶片、叶片昼夜节律运动、干枯的叶片堆砌在茎干周围——这些是其应对极端环境的适应性进化。然而,有关***的遗传信息却很少,这使得开展该物种系统学鉴定与进化方面的研究比较困难。在本工作开展前,千里光族(Senecioneae)仅有一个物种(jacobaea vulgaris)有叶绿体基因组全长的报道。得益于第二代测序技术的发展,尤其是测序价格和准确度方面的进步,我们对D. brassiciformis进行了高通量测序,并获得了其叶绿体基因组全长序列。利用该物种和另外9个菊科物种、1个茄科物种的叶绿体基因组开展了系统发育分析：分别利用反向重复序列(inverted repeat)、小单拷贝序列(small single copy)、大单拷贝序列(large single copy regions)、叶绿体全长(whole chloroplast genome)构建了系统进化树。结果表明D. brassiciformis的叶绿体基因组为一环状,全长为150,555bp,并包含4个主要区域：1个小单拷贝序列区域、1个大单拷贝序列区域,它们被2个反向重复序列区域间隔开来;平均GC含量为37.5%;和其它菊科植物的叶绿体基因组类似,D. brassiciformis的叶绿体基因组包含有121个单拷贝基因,有19个基因在反向重复区域有多个拷贝。系统进化分析表明,在10个内类群中,D. brassiciformis与j. vulgaris的进化关系最近。本论文有助于对D. brassiciformis的理解,以及开展Dendrosenecio和其它高等植物的叶绿体基因组研究。
