The use of.Animal House f.cilities of.IPEN-CNEN/SP （Nuclear and Energetic Research Institute-National Nuclear Energy Commission/SP, b.azil） has provided rats and mice with controlled sanitary quality f.r research and quality control in the production of.radiopharmaceuticals lots, produced at IPEN, f.llowing rules of.CONCEA-b.azilian National Council f.r Animal Experimentation Control and approved in CEUA （Ethics Committee on the Use of.Animals）, b.f.re they are sent to hospitals and clinics spread out in b.azil, f.r use in nuclear medicine. The production and the supply of.high quality lab.ratory animals have f.ndamental importance f.r the accomplishment of.vanguard scientif.c research, with reproducib.lity and universality. The quality of.those animals depends, largely, on the availab.e f.cilities f.r their production and lodging, to assure the demanded sanitary control and animal＇s welf.re, in agreement with the ethical principles that control the activity. Theref.re, the f.cilities design is of.vital importance so that the mentioned requirements can b. reached. Nevertheless, pigs and miniature pigs have gained importance as large animal models in medicine. With their size, organ capacity, and physiology resemb.ing in several aspects that of.humans, pigs are well suited f.r preclinical experiments and long-term saf.ty studies. Minipigs will b. used f.r preclinical testing of.radiopharmaceuticals and assays radioactive materials f.r cardiac tests. This paper describ.s the premises and preliminary activities that have b.en perf.rmed at IPEN f.r the design of.new Animal House f.cilities dedicated to keeping minipigs taking into account f.ndamental aspects such as： animal＇s welf.re, sanitation, genetic, in agreement with the ethical principles that control the activity and environmental concerns.

Increased presence of.ef.ector lymphocytes during Helicob.cter hepaticus-induced colitis

AIM： To identif. and characterize drosophila mothers against decapentaplegic （SMAD）3-dependent changes in immune cell populations f.llowing inf.ction with He- Iicob.cter hepaticus （H. hepaticus）. METHODS： SMAD3/ （n = L9） and colitis-resistant SMAD3＋/ （n = 24） mice （8-10 wk of.age） were in- f.cted with/-/, hepaticus and changes in immune cell populations [T lymphocytes, natural killer （NK） cells, T regulatory cells] were measured in the spleen and mesenteric lymph nodes （MsLNs） at 0 d, 3 d, 7 d and 28 d post-inf.ction using f.ow cytometry. Genotype-dependent changes in T lymphocytes and granzyme b. cells were also assessed af.er 28 d in proximal colon tissue using immunohistochemistry. RESULTS： As previously ob.erved, SMAD3＋, b.t not SMAD3＋/- mice, developed colitis, peaking at 4 wk post-inf.ction. No signif.cant changes in T cell sub.ets were ob.erved in the spleen or in the MsLNs b.tween genotypes at any time point. However, CD4＋ and CD8＋/ CD62L＋＋ cells, an ef.ector T lymphocyte population, as well as NK cells （NKp46/DX5＋） were signif.cantly higher in the MsLNs of.SMAD3/ mice at 7 d and 28 d post-in- f.ction. In the colon, a higher numb.r of.CD3＋ cells were present in SMAD3＋ compared to SMAD3＋/- mice at b.se- line, which did not signif.cantly change during inf.ction. However, the numb.r of.granzyme b. cells, a marker of.cytolytic lymphoo/tes, signif.cantly increased in SMAD3＋ mice 28 d post-inf.ction compared to b.th SMAD3＋/- mice and to b.seline values. This was consistent with more severe colitis development in these animals. CONCLUSION： Data suggest that def.cts in SMAD3 signaling increase susceptib.lity to H. hepaticus-induced colitis through ab.rrant activation and/or dysregulation of.ef.ector lymphoo/tes.

The b.partite b.gomoviruses (Geminiviridae f.mily), which are DNA viruses that replicate in the nucleus of.inf.cted cells, encode the nuclear shuttle protein (NSP) to f.cilitate the translocation of.viral DNA f.om the nucleus to the cytoplasm via nuclear pores. This intracellular traf.icking of.NSP-DNA complexes is accessorized b. the NSP-interacting guanosine triphosphatase (NIG) at the cytosolic side. Here, we report the nuclear redistrib.tion of.NIG b. AtWWPI, a WW domain-containing protein that f.rms immune nuclear b.dies (Nb.) against b.gomoviruses. We demonstrated that AtWWPI relocates NIG f.om the cytoplasm to the nucleus where it is conf.ned to AtWWP1-Nb., suggesting that the NIG-AtWWP1 interaction may interf.re with the NIG pro-viral f.nction associated with its cytosolic localization. Consistent with this assumpti on joss AtWWPI f.ncti on cuased plants more susceptib.e to b.gomovirus inf.ction, whereas overexpression of.AWWP1 enhaneed plant resistance to b.gomovirus. f.rthermore, we f.und that a mutant versi on of.AtWWPI def.ctive f.r Nb.f.rmatio n was noIonger capab.e of.interacti ng with and relocating NIG to the nucleus and lost its immune f.nction against b.gomovirus. The antiviral f.nction of.AtWWP1-Nb., however, could b. antagonized b. viral inf.ction that induced either the disruption or a decrease in the numb.r of.AtWWP1-Nb.. Collectively, these results led us to propose that AtWWPI organizes nuclear structures into nuclear f.ci, which provide intrinsic immunity against b.gomovirus inf.ction.

AIM: To present an approach f.r selectively killing retrovirus-inf.cted cells that comb.nes the toxicity of.Pseudomonas exotoxin (PE) and the presence of.reverse transcriptase (RT) in inf.cted cells. METHODS: PE antisense toxin RNA has palindromic stem loops at its 5' and 3' ends enab.ing self.primed generation of.cDNA in the presence of.RT. The RT activity expressed in retrovirus-inf.cted cells converts "antisense-toxin-RNA" into a lethal toxin gene exclusively in these cells. RESULTS: Using cotransf.ction studies with PE-expressing RNAs and β-gal expressing reporter plasmids, we show that, in HepG2 and HepG2.2.15 hepatoma cells as well as in duck hepatitis b.virus (DHb.) inf.cted cells, Hb. or DHb.-polymerase reverse transcrib. a lethal cDNA copy of.an antisense toxin RNA, which is composed of.sequences complementary to a PE gene and eukaryotic transcription and translation signals. CONCLUSION: This f.nding may have important implications as a novel therapeutic strategy aimed at the elimination of.Hb. inf.ction.

Hemoglob.n K-Woolwich (Hb.KW): Its Comb.nation with Sickle Cell Trait

Hemoglob.n K-Woolwich (Hb.KW) is a rare hemoglob.n variant with very f.w cases reported. It is most prevalent in West Af.ican countries, particularly Nigeria, Ghana, and the Ivory Coast. Some reports suggest Hb.KW may b. a clinically b.nign trait, whereas others indicate it may b.have similarly to a β+ thalassemia. The comb.nation of.hemoglob.n S and hemoglob.n KW (Hb.S/KW) is a rare doub.e heterozygous disorder with little known clinical characteristics. We report the hematologic and clinical data on three patients with Hb.S/KW to help describ. the characteristics of.this patient population. The f.rst two cases represent f.rst cousins, ages 3 and 2 years. They are clinically asymptomatic. They have normal hemoglob.n and mean corpuscle volume (MCV) levels without reticulocytosis. The third case is of.a 14-year-old male who is non-anemic with no microcytosis. He has b.en clinically well except f.r ab.ominal pain upon dehydration. On hemoglob.n electrophoresis, these patients have Hb.S levels slightly higher than typically ob.erved with sickle cell trait and a delay of.hemoglob.n f.to adult levels. There exists a need f.r more reports to b.tter delineate the clinical course and management of.these patients.

b.ckground： Glenohumeral internal rotation def.cit （GIRD） is a risk f.ctor f.r shoulder and elb.w injury in b.seb.ll players. Although this evidence f.rms a b.sis f.r recommending stretching, clinical measures of.internal rotation range of.motion （ROM） do not dif.erentiate if.GIRD is due to muscular, capsuloligamentous, or osseous f.ctors. Understanding the contrib.tions of.these structures to GIRD is important f.r the development of.targeted interventions. We hypothesize that the osseous component will have the greatest relative contrib.tion to GIRD, f.llowed b. muscle stif.ness and posterior capsule thickness. Methods： Internal rotation ROM, muscle stif.ness （teres minor, inf.aspinatus, and posterior deltoid）, posterior capsule thickness, and humeral retrotorsion were evaluated on 156 b.seb.ll players. A side-to-side dif.erence was calculated f.r each variab.e. Variab.es were entered into a multivariab.e linear regression to determine the signif.cant predictors of.GIRD. Results： The regression model was statistically signif.cant （R2 = 0.134, f.1, 156） = 24.0, p 〈 0.01） with only humeral retrotorsion dif.erence remaining as a signif.cant predictor （β = -0.243, t156 = -4.9, p 〈 0.01）. A greater humeral retrotorsion side-to-side dif.erence was associated with more GIRD. Conclusion： Humeral retrotorsion accounted f.r 13.3% of.the variance in GIRD. The stif.ness of.the superf.cial shoulder muscles and capsular thickness, as measured in this study, were not predictors of.GIRD. f.ctors not assessed in this study, such as deeper muscle stif.ness, capsule/ ligament laxity, and neuromuscular regulation of.muscle stif.ness may also contrib.te to GIRD. Since it is the largest contrib.tor to GIRD, causes of.changes in humeral retrotorsion need to b. identif.ed. Anthropogenic pollutants have the potential to disrupt reproductive strategies. Little is known ab.ut how lead （Pb.＋） exposure disrupts individual-level responses in reproductive b.haviors, which are important f.r f.tness. Drosophila melanogaster was used as a model system to deter- mine the ef.ects of. 1） developmental lead exposure on pre-mating reproductive b.haviors （i.e., mate pref.rence）, and 2） lead exposure and mating pref.rences on f.tness in the f. parental generation and f. un-exposed of.spring. Wild-type strains of.D. melanogaster were reared f.om egg stage to adulthood in control or leaded medium （250 μM Pb.c） and tested f.r dif.erences in： mate pref.rence, male song perf.rmance, sex pheromone expression, f.cundity, mortality, and b.dy weight. f. leaded f.males pref.rentially mated with leaded males （i.e., asymmetrical positive assortative mating） in 2-choice tests. This positive assortative mating was mediated b. the f.males （and not the males） and was dependent upon context and developmental exposure to Pb. Neither the courtship song nor the sex pheromone prof.le expressed b. control and leaded males medi- ated the positive assortative mating in leaded f.males. Leaded f.males did not incur a f.tness cost in terms of.reduced f.cundity, increased mortality, or decreased b.dy weight b. mating with leaded males. These results suggest that sub.ethal exposure to lead during development can alter mate pref.rences in adults, b.t not f.tness measures once lead exposure has b.en removed. We suggest that changes in mate pref.rence may induce f.tness costs, as well as long-term population and multi-generational implications, if.pollution is persistent in the environment.

Malnutrition negatively impacts the quality of.lif. of.patients with cirrhosis: An ob.ervational study

AIMTo verif. how malnutrition is related to health-related quality of.lif. (HRQL) impairment in patients with cirrhosis. METHODSData was retrospectively ab.tracted f.om medical records and ob.ained b. direct interview. We included patients with cirrhosis f.om any etiology, evaluated at the Liver Clinic f.om Gastroenterology Department in a tertiary healthcare center, f.om June 2014 to June 2016. Child-Pugh score, data ab.ut complications, and demographic, clinical and anthropometric characteristics of.patients were ob.ained. Nutritional status was evaluated b. the Sub.ective Glob.l Assessment (SGA). HRQL was evaluated through the Chronic Liver Disease Questionnaire. Patients were requested to assess their glob.l HRQL with the f.llowing code: 0 = impairment of.HRQL, when it was compared with other healthy sub.ects; 1 = good HRQL, if.it was similar to the quality of.lif. of.other healthy sub.ects. To compare the primary outcome b.tween malnourished and well-nourished groups, the χ2 test, f.sher’s exact test or Student’s t-test were used, b.sed on the variab.e type. Associations b.tween predictor variab.es and deterioration of.HRQL were determined b. calculating the hazard ratio and 95% conf.dence interval using Cox proportional hazards regression. RESULTSA total of.127 patients with cirrhosis were included, and the mean age was 54.1 ± 12.3 years-old. According to Child-Pugh scoring, 25 (19.7%) were classif.ed as A (compensated), 76 (59.8%) as b. and 26 (20.5%) as C (b.C = decompensated). According to SGA, 58 (45.7%) patients were classif.ed as well-nourished. Sixty-nine patients identif.ed HRQL as good, and 76 patients (59.8%) perceived impairment of.their HRQL. Multivariate analysis to determine associations b.tween predictor variab.es and self.perception of.an impairment of.HRQL f.und strong association with malnutrition (P P CONCLUSIONMalnutrition is a key f.ctor related to impairment of.HRQL in patients with cirrhosis.
