e.R is a plasma-me.brane.re.ide.t re.e.tor re.ponsible.for re.ognition of microbial e.ongation factortu （e.-tu） and thus trigge.ing plant innate.immunity to fe.d off phytopathoge.s. Functional e.R must be.subje.t to the.e.doplasmic re.iculum quality control （e.QC） machine.y for the.corre.t folding and prope. asse.bly in orde. to re.ch its final de.tination. Ge.e.ic studie. have.de.onstrate. that erd2b, a counte.part of the.ye.st or mammalian HDe. re.e.tor erd2 for re.aining prote.ns in the.e.doplasmic re.iculum （e.） lume., is re.uire. for e.R function in plants （Li e. al., 2009）. In this study, we.characte.ize. the.Arabidopsis glucosidase.Ⅱ β--subunit via the.H De. motif against the.non-re.undant prote.n database. Data mining also re.e.le. that the.glucosidase.Ⅱ β--subunit ge.e.has a highly similar e.pre.sion patte.n to erd2b and the.othe. known e.QC compone.ts involve. in e.R bioge.e.is. Importantly, the.t-DNA inse.tion line. of the.glucosidase.Ⅱ β-subunit ge.e.showe. that e.R-controlle. re.ponse. we.e.substantially re.uce. or comple.e.y blocke. in the.e.mutants. the.re.ponse. include.se.dling growth inhibition, induction of marke. ge.e., MAP kinase.activation, and callose.de.osition, trigge. by pe.tide.e.f18, a full mimic of e.F-tu. take. toge.he., ourdata indicate.a re.uire.e.t of the.glucosidase.Ⅱ β-subunitfor e.R function.

AIM： to study the.e.pre.sion of e.dothe.ial and inducible.nitric oxide.synthase. （e.OS and iNOS） and the.r role.in inflammatory bowe. dise.se.（IBD）. Me.HODS： We.e.amine. the.e.fe.t of se.a obtaine. from patie.ts with active.Crohn＇s dise.se.（CD） and ulce.ative.colitis （UC） on the.function and viability of human umbilical ve.n e.dothe.ial ce.ls （HUVe.）. HUVe.s we.e.culture. for 0-48 h in the.pre.e.ce.of a me.ium containing poole. se.um of he.lthy controls, or se.um from patie.ts with active.CD or UC. e.pre.sion of e.OS and iNOS was visualize. by immunofluore.ce.ce. and quantifie. by the.de.sitome.ry of We.te.n blots. Prolife.ation activity was asse.se. by compute.ize. image.analyse. of Ki-67 immunore.ctive.ce.ls, and also te.te. in the.pre.e.ce.of the.NOS inhibitor, 10^-4 mol/L L-NAMe. Apoptosis and ne.rosis was e.amine. by the.anne.in-V-biotin me.hod and by propidium iodide.staining, re.pe.tive.y. Re.ULtS： In HUVe. imme.iate.y afte. e.posure.to UC, se.um e.OS was marke.ly induce., re.ching a pe.k at 12 h. In contrast, a de.re.se.in e.OS was obse.ve. afte. incubation with CD se.a and the.e.OS le.e. was minimal at 20 h compare. to control （18%±16% vs 23%± 15% P〈0.01）. UC or CD se.um cause. a significant incre.se.in iNOS compare. to control （UC： 300%±21%; CD： 275% ± 27% vs 108% ± 14%, P〈0.01）. Apoptosis/ne.rosis characte.istics did not diffe. significantly in e.the. e.pe.ime.t. Incre.se. prolife.ation activity was de.e.te. in the.pre.e.ce.of CD se.um or afte. tre.tme.t with L-NAMe. Culture. showe. tube.like.formations afte. 24 h tre.tme.t with CD se.um. CONCLUSION： IBD se.a e.oke. change. in the.ratio of e.OS/iNOS, whe.e.s did not influe.ce.the.viability of HUVe.. the.e.involve. down-re.ulation of e.OS and up-re.ulation of iNOS simultane.usly, le.ding to incre.se. prolife.ation activity and possibly a re.uce. antiinflammatory prote.tion of e.dothe.ial ce.ls.
